Abundance and diversity of microfungi in leaf litter of a lowland rain forest in Costa Rica

Root, arbuscular-mycorrhizal (AM), soil faunal (protozoa and microarthropods), and microbial responses to field exposure to CO2 for six growing seasons were measured in spring 1997 in two adjacent grassland communities. The grasslands showed contrasting root responses to CO2 enrichment: whereas root length was not affected in the sandstone grassland, it was greater in the serpentine grassland, as was specific root length. AM fungal hyphal lengths were greater in the sandstone, but were unaffected in the serpentine community. This lent support to the hypothesis that there may be a tradeoff in resource allocation to more fine roots or greater mycorrhizal extraradical hyphal length. AM root infection was greater in both communities at elevated CO2, as was the proportion of roots containing arbuscules. Our data on total hyphal lengths, culturable and active fungi, bacteria, and protozoa supported the hypothesis that the fungal food chain was more strongly stimulated than the bacterial chain. This study is one of the first to test these hypotheses in natural multi-species communities in the field.

A method with the combined advantages of soil particle washing, selective inhibitors, and an indicator substrate was developed to isolate saprophytic basidiomycetes from soil. Organic particles were washed from soil and plated on a medium containing lignin, guaiacol, and benomyl, which reduced mold growth and allowed detection of basidiomycetes producing laccase or peroxidase. The 64 soil samples yielded 67 basidiomycete isolates, representing 51 groups on the basis of morphology and physiology. This method should facilitate investigations into the biodiversity of soil basidiomycetes and yield organisms that are useful in bioremediation of soils contaminated with pesticides or other recalcitrant aromatic compounds.

Isolation of saprophytic basidiomycetes from soil.

An automatic soil-washing apparatus is described which enabled the washing of fungus spores out of large numbers of soil samples simultaneously. Tests of the apparatus indicated that superficial spores were removed from most of the soil particles. This allowed organisms growing in soil as vegetative mycelium to be isolated more readily without the serious competition commonly encountered from organisms sporulating heavily in the soil. Virtually no cross-contamination of soil samples occurred in the multichambered apparatus.
