While many experiment with drugs, relatively few individuals develop a true addiction. We hypothesized that, in rats, such individual differences in the actions of addictive drugs might be determined by postnatal rearing conditions. To test this idea, we investigated whether stimulant- and stress-induced activation of nucleus accumbens dopamine transmission and dopamine-dependent behaviours might differ among adults rats that had been either repeatedly subjected to prolonged maternal separation or a brief handling procedure or left undisturbed (non-handled) during the first 14 days of life. We found that, in comparison with their handled counterparts, maternally separated and non-handled animals are hyperactive when placed in a novel setting, display a dose-dependent higher sensitivity to cocaine-induced locomotor activity and respond to a mild stressor (tail-pinch) with significantly greater increases in nucleus accumbens dopamine levels. In addition, maternally separated animals were found to sensitize to the locomotor stimulant action of amphetamine when repeatedly stressed under conditions that failed to sensitize handled and non-handled animals. Finally, quantitative receptor autoradiography revealed a lower density of nucleus accumbens-core and striatal dopamine transporter sites in maternally separated animals. Interestingly, we also found greatly reduced D(3) dopamine receptor binding and mRNA levels in the nucleus accumbens-shell of handled animals. Together, these findings provide compelling evidence that disruptions in early postnatal rearing conditions can lead to profound and lasting changes in the responsiveness of mesocorticolimbic dopamine neurons to stress and psychostimulants, and suggest a neurobiological basis for individual differences in vulnerability to compulsive drug taking.

Influence of early postnatal rearing conditions on mesocorticolimbic dopamine and behavioural responses to psychostimulants and stressors in adult rats.

Dodecaborate anions of the type B12X122− and B12X11Y2− (X=H, Cl, Br, I and Y=OH, SH, NH3+, NR3+) form strong (Ka up to 106 L mol−1, for B12Br122−) inclusion complexes with γ-cyclodextrin (γ-CD). The micromolar affinities reached are the highest known for this native CD. The complexation exhibits highly negative enthalpies (up to −25 kcal mol−1) and entropies (TΔS up to −18.4 kcal mol−1, both for B12I122−), which position these guests at the bottom end of the well-known enthalpy-entropy correlation for CDs. The high driving force can be traced back to a chaotropic effect, according to which chaotropic anions have an intrinsic affinity to hydrophobic cavities in aqueous solution. In line with this argument, salting-in effects revealed dodecaborates as superchaotropic dianions.

Water Structure Recovery in Chaotropic Anion Recognition: High-Affinity Binding of Dodecaborate Clusters to γ-Cyclodextrin.

Following up on scattered reports on interactions of conventional chaotropic ions (for example, I- , SCN- , ClO4 - ) with macrocyclic host molecules, biomolecules, and hydrophobic neutral surfaces in aqueous solution, the chaotropic effect has recently emerged as a generic driving force for supramolecular assembly, orthogonal to the hydrophobic effect. The chaotropic effect becomes most effective for very large ions that extend beyond the classical Hofmeister scale and that can be referred to as superchaotropic ions (for example, borate clusters and polyoxometalates). In this Minireview, we present a continuous scale of water-solute interactions that includes the solvation of kosmotropic, chaotropic, and hydrophobic solutes, as well as the creation of void space (cavitation). Recent examples for the association of chaotropic anions to hydrophobic synthetic and biological binding sites, lipid bilayers, and surfaces are discussed.

The Chaotropic Effect as an Assembly Motif in Chemistry.

Thermo-electrochemical cells have potential to generate thermoelectric voltage 1 order higher than that given by semiconductor materials. To overcome the current issues in thermoelectric energy conversion, it is of paramount importance to grow and fulfill the full potential of thermo-electrochemical cells. Here we report a rational supramolecular methodology that yielded the highest Seebeck coefficient of ca. 2.0 mV K–1 around ambient temperatures. This is based on the encapsulation of triiodide ions in α-cyclodextrin, whose equilibrium is shifted to the complexation at lower temperatures, whereas it is inverted at elevated temperatures. This temperature-dependent host–guest interaction provides a concentration gradient of redox ion pairs between two electrodes, leading to the eminent performance of the thermo-electrochemical cells. Supramolecular Thermo-Electrochemical Cells: Enhanced Thermoelectric Performance by Host-Guest Complexation and Salt-Induced Crystallization

The synthetic macrocycle cucurbit[6]uril forms host–guest inclusion complexes with molecular dibromine and diiodine. As evidenced by their crystal structures, the encapsulated dihalogens adapt a tilted axial geometry and are held in place by two different types of halogen-bonding interactions, one with a water molecule (bond distances 2.83 A for O···Br and 3.10 A for O···I) and the other one with the ureido carbonyl groups of the molecular container itself (bond distances 3.33 A for O···Br and 3.49 A for O···I). While the former is of the conventional type, involving the lone electron pair of an oxygen donor, the latter is perpendicular, involving the π-system of the carbonyl oxygen (N–C═O···X dihedrals ca. 90°). Such perpendicular interactions resemble those observed in protein complexes of halogenated ligands.
