Research on chitin as a marine resource is pointing to novel applications for this cellulose-like biopolymer. Discovery of nondegrading solvent systems has permitted the spinning of filaments, for example, for use as surgical sutures. New methods for preparing the bioactive alkyl glycoside of N-acetyl-D-glucosamine (the monomer unit of chitin) and a microcrystalline chitin has encouraged their use as promoters for growth of bifidobacteria and as an aid in digestion of high-lactose cheese whey by domestic animals. Chitin-protein complexes of several crustacean species show great variability in ratios of chitin to covalently bound protein and in residual protein in the "purified" chitins.

Publisher Summary This chapter discusses the cell wall synthesis in apical hyphal growth. It appears that the hyphal apex is best viewed as a highly polarized system of exocytosis. Wall materials, extracellular enzymes, and probably other substances are excreted at the growing end of a tubular cell. The most obvious cellular features that accompany this polarized system are (1) the unidirectional flow of vesicles in the cytoplasm fusing with the plasma membrane at the apex, (2) the gradients in wall synthesis at the apex, and (3) the cytoplasmic gradients in ion distribution that are maintained at the apex. New microscopic techniques reveal a cytoskeletal organization of the cytoplasm at the apex, which may be crucial to its polarized activity. Growth of the wall at the hyphal apex requires that the wall in this region has plastic properties, which contrast with the requirement of rigidity elsewhere in the hypha. A widely held view involves the participation of wall-lytic enzymes in plasticizing the wall at the apex and in allowing new wall material to be inserted. A critical evaluation of the evidence presented to support this view makes this hypothesis less attractive. As an alternative a steady-state model is discussed based on recent observations in the author's laboratory. This model holds that the assemblage of polymers synthesized at the apex is inherently plastic. However, this assemblage develops rigidity by interactions, in the wall, between and among the various individual polymers present while the wall segment moves in subapical directions during elongation. This model seems to fit many of the original observations made on living hyphae.

Publisher Summary This chapter highlights some of the recent and important findings obtained from studies conducted on the synthesis, structure, physical state, modification, organization, and degradation of chitin in insect tissues, as well as the interplay of chitin with chitin-binding proteins, the regulation of genes responsible for chitin metabolism, and, finally, the targeting of chitin metabolism for insect-control purposes. Chitin is the major polysaccharide present in insects and many other invertebrates as well as in several microbes, including fungi. It serves as the skeletal polysaccharide of several animal phyla, such as the Arthropoda, Annelida, Molluska, and Coelenterata. In several groups of fungi, chitin replaces cellulose as the structural polysaccharide. In insects, it is found in the body wall or cuticle, gut lining or peritrophic matrix (PM), salivary gland, trachea, eggshells, and muscle attachment points. In the course of evolution, insects have made excellent use of the rigidity and chemical stability of the polymeric chitin to assemble both hard and soft extracellular structures such as the cuticle (exoskeleton) and PM respectively, both of which enable insects to be protected from the environment while allowing for growth, mobility, respiration, and communication. All of these structures are primarily composites of chitin fibers and proteins with varying degrees of hydration and trace materials distributed along the structures.

Chitin Metabolism in Insects

A "soluble" form of chitin synthetase was separated from a membrane-rich fraction by exposure to the enzyme substrate (uridine diphosphate N-acetyl-D-glucosamine) and activator (N-acetyl- D-glucosamine). The solubilized enzyme catalyzed the synthesis of chitin microfibrils similar, if not identical, to those formed in vivo by the fungus. Cell wall microfibrils were thus abundantly formed in the absence of a living cell or its membranes.
