Effect of a Proposed Trastuzumab Biosimilar Compared With Trastuzumab on Overall Response Rate in Patients With ERBB2 (HER2)-Positive Metastatic Breast Cancer: A Randomized Clinical Trial.
Hope S. Rugo,Abhijit Barve,Cornelius F. Waller,Miguel Hernandez-Bronchud,Jay Herson,Jinyu Yuan,Rajiv Sharma,Mark Baczkowski,Mudgal Kothekar,Subramanian Loganathan,Alexey Manikhas,Igor Bondarenko,Guzel Mukhametshina,Gia Nemsadze,Joseph D. Parra,Maria Luisa T. Abesamis-Tiambeng,Kakhaber Baramidze,Charuwan Akewanlop,Ihor Vynnychenko,Virote Sriuranpong,Gopichand Mamillapalli,Sirshendu Ray,Eduardo Yanez Ruiz,Eduardo J. Pennella +23 more
TL;DR: Among women with ERBB2-positive metastatic breast cancer receiving taxanes, the use of a proposed biosimilar compared with trastuzumab resulted in an equivalent overall response rate at 24 weeks, suggesting a clinically effective biosimilar may help increase access to this therapy.
Journal ArticleDOI
The Use of Interim Analysis for Sample Size Adjustment
Jay Herson,Janet Turk Wittes +1 more
TL;DR: The sample size adjustment process is found to have negligible effect on significance level and to deliver reasonable power, and through proper use of a data monitoring board the potential for unblinding is negligible.

Data and Safety Monitoring Committees in Clinical Trials
TL;DR: Issues Due to Maturing of DMC Processes and Evolution of the Pharmaceutical Industry Resignation from a DMC are issues due to maturing of the DMC processes and evolution of the pharmaceutical industry.
Journal ArticleDOI
Guidelines for data and safety monitoring for clinical trials not requiring traditional data monitoring committees
Dennis O. Dixon,Ralph S. Freedman,Jay Herson,Michael Hughes,KyungMann Kim,Michael H. Silverman,Catherine M. Tangen +6 more
TL;DR: Recommendations are given for appropriate elements of a data monitoring plan, decision criteria for institution of aData Monitoring Committee (DMC), and critical elements for a DMC to consider in exploratory trials.
