The Genetics of Autism Spectrum Disorders

TL;DR: Three approaches to identifying genetic factors that contribute to the pathogenesis of ASDs are reviewed: common variants and genome-wide association studies (GWAS); 2) rare variants and copy number variation (CNV) studies, and 3) familial forms of autism and the role of next-generation sequencing (NGS) methods.
Journal ArticleDOI

The Philadelphia Neurodevelopmental Cohort: A publicly available resource for the study of normal and abnormal brain development in youth.

TL;DR: The Philadelphia Neurodevelopmental Cohort is a large-scale study of child development that combines neuroimaging, diverse clinical and cognitive phenotypes, and genomics and is evolving to be a significant resource for the broader neuroscience community for studies of normal and abnormal neurodevelopment.
Journal ArticleDOI

The Philadelphia Neurodevelopmental Cohort: constructing a deep phenotyping collaborative

TL;DR: The PNC assessment mechanism yielded psychopathology data with strong factorial validity in a large diverse community cohort of genotyped youths and can advance understanding of complex inter-relationships among genes, cognition, brain, and behavior involved in neurodevelopment of common mental disorders.
Journal ArticleDOI

The psychosis spectrum in a young U.S. community sample: findings from the Philadelphia Neurodevelopmental Cohort.

TL;DR: Youths with spectrum symptoms had reduced accuracy across neurocognitive domains, reduced global functioning, and increased odds of depression, anxiety, behavioral disorders, substance use and suicidal ideation, which have public health relevance for prevention and early intervention.
