Journal ArticleDOI
A novel concept of reversing neuromuscular block: chemical encapsulation of rocuronium bromide by a cyclodextrin-based synthetic host.
Anton Bom,Mark Bradley,Kenneth S. Cameron,John K. Clark,Jan van Egmond,Helen Feilden,Elizabeth J. MacLean,Alan W. Muir,Ronald Palin,David C. Rees,Zhang Mingqiang +10 more
Journal ArticleDOI
Cyclodextrin-derived host molecules as reversal agents for the neuromuscular blocker rocuronium bromide: synthesis and structure-activity relationships.
Julia Adam,D. Jonathan Bennett,Anton Bom,John K. Clark,Helen Feilden,Edward J Hutchinson,Ronald Palin,Alan Prosser,David C. Rees,Georgina M. Rosair,Donald Stevenson,Gary J Tarver,Zhang Mingqiang +12 more
TL;DR: A series of mono- and per-6-substituted cyclodextrin derivatives were synthesized as synthetic receptors (or host molecules) of rocuronium bromide and can be used as reversal agents of the neuromuscular blocker to assist rapid recovery of patients after surgery.
Journal ArticleDOI
Discovery and SAR of org 24598-a selective glycine uptake inhibitor.
Angus R. Brown,Ian Craig Carlyle,John K. Clark,William Hamilton,Samuel George Gibson,George McGarry,Sandra McEachen,Rae Duncan Robertson,Simon Nicholas Thorn,Glenn Walker +9 more
TL;DR: The discovery of Org 24598, one of the first potent and selective inhibitors of the glycine transporter is discussed and in vitro structure-activity relationships (SARs) data for interaction of a ligand with this system is discussed.
Journal ArticleDOI
Conformationally Constrained Anesthetic Steroids That Modulate GABAA Receptors
Alison Anderson,Andrew C. Boyd,John K. Clark,Lee Fielding,David K. Gemmell,Niall M. Hamilton,Maurice S. Maidment,Valerie May,Ross McGuire,Petula McPhail,Francis H. Sansbury,Hardy Sundaram,Robert Taylor +12 more
TL;DR: For example, the authors showed that for optimal GABA(A) receptor modulation, the hydrogen bond-accepting substituent should be near perpendicular to the plane of the D-ring on the beta-face of the alpha-hydroxypregnan-20-one.
Journal ArticleDOI
Quaternary Salts of E2020 Analogues as Acetylcholinesterase Inhibitors for the Reversal of Neuromuscular Block
John K. Clark,Phill Cowley,Alan W. Muir,Ronald Palin,Eleanor Pow,Alan B. Prosser,Robert Taylor,Zhang Mingqiang +7 more
TL;DR: A series of benzylpiperidinium and Benzylpyrid inium quaternary salts have been synthesised and tested for inhibition of acetylcholinesterase and reversal of neuromuscular block induced by vecuronium.
