Journal ArticleDOI

Analysis of the reactions used for the preparation of drug candidate molecules.

TL;DR: The purpose of this perspective is to indicate the range of chemistries used in the manufacture of drug candidate molecules and to highlight certain gaps in current technologies.
Journal ArticleDOI

Lipase catalysed resolution of the Lotrafiban intermediate 2,3,4,5-tetrahydro-4-methyl-3-oxo-1H-1,4-benzodiazepine-2-acetic acid methyl ester in ionic liquids: comparison to the industrial t-butanol process

TL;DR: In this article, the results suggest that ionic liquids can be very favorable reaction media for industrial bioconversion processes, which also overcome many of the safety and environmental concerns of conventional organic solvents.
Journal ArticleDOI

The Development of a Manufacturing Route for the GPIIb/IIIa Receptor Antagonist SB-214857-A. Part 2: Conversion of the Key Intermediate SB-235349 to SB-214857-A

TL;DR: In this article, the process development to the manufacturing route to (2S)-7-([4,4‘-bipiperidin]-1-ylcarbonyl)-2,3,4,5-tetrahydro-4-methyl-3-oxo-1H-1,4-benzodiazepine-2-acetic acid hydrochloride (SB-214857-A, lotrafiban) is described.
Journal ArticleDOI

A new synthesis of the GPIIb/IIIa receptor antagonist SB-214857-A

TL;DR: A new synthesis of lotrafiban SB-214857-A was reported in this paper, where the key steps were the preparation of racemic (4methyl-3-oxo,2,3,4,5-tetrahydro-1H-benzo[e][1,4]diazepin-2-yl)-acetic acid methyl ester from 2-nitrobenzyl alcohol, using an immobilised lipase enzyme and a palladium-catalysed aminocarbonylation reaction.
