Metabolome analysis by capillary electrophoresis-mass spectrometry.

An overview of the use of CE-MS in the field of metabolomics is provided. Metabolomics is concerned with the comprehensive analysis of endogenous low-molecular-weight compounds in biological samples. CE-MS has demonstrated to be a powerful technique for the profiling of polar metabolites in biological samples. This review covers the use of various CE separation modes, capillary coatings, MS analyzers, sample preparation techniques, and data analysis methods used in CE-MS for metabolomics. The applicability of CE-MS in metabolomics research is illustrated by giving examples of the analysis of bacterial extracts, plant extracts, urine, plasma, and cerebrospinal fluid samples. The relevant CE-MS metabolomics studies published between 2000 and 2008 are presented in tabular form, including information on sample type and pretreatment and MS detection mode. Future developments with regard to the use of alternative ionization techniques, the use of coupled separation systems and the potential of microchip CE systems for metabolomics are discussed.

CE-MS in metabolomics.

This review gives an overview of applications of CE coupled to MS detection published in the literature of the last three years. The works discussed in this paper comprise a wide range of different fields of application. These include important sections such as the analysis of biomolecules, the analysis of pharmaceuticals and their metabolites in different matrices, environmental analysis, and also investigations on the composition of technical products.

Biomarkers are biomolecules that serve as indicators of biological and pathological processes, or physiological and pharmacological responses to a drug treatment. Because of the high abundance of albumin and heterogeneity of plasma lipoproteins and glycoproteins, biomarkers are difficult to identify in human serum. Due to the clinical significance the identification of disease biomarkers in serum holds great promise for personalized medicine, especially for disease diagnosis and prognosis. This review summarizes some common and emerging proteomics techniques utilized in the separation of serum samples and identification of disease signatures. The practical application of each protein separation or identification technique is analyzed using specific examples. Biomarkers of cancers of prostate, breast, ovary, and lung in human serum have been reviewed, as well as those of heart disease, arthritis, asthma, and cystic fibrosis. Despite the advancement of technology few biomarkers have been approved by the Food and Drug Administration for disease diagnosis and prognosis due to the complexity of structure and function of protein biomarkers and lack of high sensitivity, specificity, and reproducibility for those putative biomarkers. The combination of different types of technologies and statistical analysis may provide more effective methods to identify and validate new disease biomarkers in blood.

Methodology and Applications of Disease Biomarker Identification in Human Serum

We report the use of capillary electrophoresis-electrospray ionization-mass spectrometry (CE-ESI-MS) for the determination of antiretroviral dideoxynucleosides (ddNs), their nucleotides, and a set of ribonucleosides and ribonucleotides. A CE system for separation of most commonly used antiretroviral ddNs has been developed based on a basic buffer with a volatile electrolyte suitable for ESI-MS detection in an untreated capillary column. Positive and negative ionization modes are investigated and compared for sensitive and stable electrospray performance. A 14-compound mixture of nucleosides and nucleotides is profiled in a single capillary zone electrophoresis separation with a distinct elution order: electroosmotic flow, ddNs, mononucleotides, dinucleotides, and trinucleotides in less than 18 min. The fragmentation pathways of the nucleosides and nucleotides in ESI-MS have been interpreted. Concentration limits of detection are 100 to 200 nM with an injection volume of approximately 10 nL. This technique has been used to detect naturally occurring nucleotides and to study the metabolism of lamivudine (3TC) in the human hepatoma cell line Hep G2. 3TC and its metabolites 3TC-monophosphate, 3TC-diphosphate, and 3TC-triphosphate were detected after 10 h of incubation of 3TC with the cells.
