AAV2 Gene Therapy Readministration in Three Adults with Congenital Blindness
Jean Bennett,Jean Bennett,Manzar Ashtari,Jennifer Wellman,Kathleen A. Marshall,Laura Cyckowski,Daniel C. Chung,Daniel C. Chung,Sarah McCague,Eric A. Pierce,Eric A. Pierce,Yifeng Chen,Jeannette L. Bennicelli,Xiaosong Zhu,Gui-Shuang Ying,Junwei Sun,J. Fraser Wright,Alberto Auricchio,Francesca Simonelli,Kenneth S. Shindler,Federico Mingozzi,Katherine A. High,Katherine A. High,Albert M. Maguire,Albert M. Maguire +24 more
TL;DR: The researchers report that the lack of immune response and the robust safety profile in this readministration gene therapy study may be due in part to the immune-privileged nature of the eye, and the low dose and very pure preparation of the AAV vector.
Journal ArticleDOI
Safety and durability of effect of contralateral-eye administration of AAV2 gene therapy in patients with childhood-onset blindness caused by RPE65 mutations: a follow-on phase 1 trial
Jean Bennett,Jean Bennett,Jennifer Wellman,Kathleen A. Marshall,Sarah McCague,Manzar Ashtari,Manzar Ashtari,Julie DiStefano-Pappas,Okan U. Elci,Daniel C. Chung,Junwei Sun,Junwei Sun,J. Fraser Wright,Dominique Cross,Puya Aravand,Laura Cyckowski,Jeannette L. Bennicelli,Federico Mingozzi,Alberto Auricchio,Eric A. Pierce,Jason Ruggiero,Bart P. Leroy,Bart P. Leroy,Francesca Simonelli,Katherine A. High,Katherine A. High,Albert M. Maguire +26 more
TL;DR: To the knowledge, AAV2-hRPE65v2 is the first successful gene therapy administered to the contralateral eye and the results highlight the use of several outcome measures and help to delineate the variables that contribute to maximal benefit from gene augmentation therapy in this disease.
Journal ArticleDOI
Medial temporal structures and memory functions in adolescents with heavy cannabis use
Manzar Ashtari,Brian B. Avants,Laura Cyckowski,Kelly L. Cervellione,David Roofeh,Philip A. Cook,James C. Gee,Serge Sevy,Sanjiv Kumra +8 more
TL;DR: Compared hippocampal morphology in 14 "treatment-seeking" adolescents with a history of prior heavy cannabis use with an average of 6.7 month of supervised abstinence lends support to a theory that cannabis use may impart long-term structural and functional damage.
Journal ArticleDOI
The human visual cortex responds to gene therapy-mediated recovery of retinal function.
Manzar Ashtari,Laura Cyckowski,Justin F. Monroe,Kathleen A. Marshall,Daniel C. Chung,Alberto Auricchio,Francesca Simonelli,Bart P. Leroy,Albert M. Maguire,Kenneth S. Shindler,Jean Bennett +10 more
TL;DR: Functional MRI data suggest that despite severe and long-term visual impairment, treated LCA2 patients have intact and responsive visual pathways, and suggest that gene therapy resulted in not only sustained and improved visual ability, but also enhanced contrast sensitivity.
Journal ArticleDOI
Plasticity of the human visual system after retinal gene therapy in patients with Leber’s congenital amaurosis
Manzar Ashtari,Hui Zhang,Philip A. Cook,Laura Cyckowski,Kenneth S. Shindler,Kathleen A. Marshall,Puya Aravand,Arastoo Vossough,James C. Gee,Albert M. Maguire,Chris I. Baker,Jean Bennett,Jean Bennett +12 more
TL;DR: It is demonstrated that reversing blindness with gene therapy promoted long-term structural plasticity in the visual pathways emanating from the treated retina of LCA patients, and improvements and normalization along the visual fibers corresponding to the site of retinal injection of the gene therapy vector carrying the therapeutic gene in the treated eye compared to the visual pathway for the untreated eye of L CA patients.
