Electrografting refers to the electrochemical reaction that permits organic layers to be attached to solid conducting substrates. This definition can be extended to reactions involving an electron transfer between the substrate to be modified and the reagent, but also to examples where a reducing or oxidizing reagent is added to produce the reactive species. These methods are interesting as they provide a real bond between the surface and the organic layer. Electrografting applies to a variety of substrates including carbon, metals and their oxides, but also dielectrics such as polymers. Since the 1980s several methods have been developed, either by reduction or oxidation, and some of them have reached an industrial stage. This critical review describes the methods that are used for electrografting, their mechanism, the formation and growth of the layers as well as their applications (742 references).

Electrografting: a powerful method for surface modification

Rapid progress in identifying biomarkers that are hallmarks of disease has increased demand for high-performance detection technologies. Implementation of electrochemical methods in clinical analysis may provide an effective answer to the growing need for rapid, specific, inexpensive, and fully automated means of biomarker analysis. This Review summarizes advances from the past 5 years in the development of electrochemical sensors for clinically relevant biomolecules, including small molecules, nucleic acids, and proteins. Various sensing strategies are assessed according to their potential for reaching relevant limits of sensitivity, specificity, and degrees of multiplexing. Furthermore, we address the remaining challenges and opportunities to integrate electrochemical sensing platforms into point-of-care solutions.

Electrochemical Methods for the Analysis of Clinically Relevant Biomolecules

A comprehensive review on the development and state of the art of colorimetric and fluorometric sensor arrays is presented Chemical sensing aims to detect subtle changes in the chemical environment by transforming relevant chemical or physical properties of molecular or ionic species (ie, analytes) into an analytically useful output Optical arrays based on chemoresponsive colorants (dyes and nanoporous pigments) probe the chemical reactivity of analytes, rather than their physical properties (eg, mass) The chemical specificity of the olfactory system does not come from specific receptors for specific analytes (eg, the traditional lock-and-key model of substrate-enzyme interactions), but rather olfaction makes use of pattern recognition of the combined response of several hundred olfactory receptors In a similar fashion, arrays of chemoresponsive colorants provide high-dimensional data from the color or fluorescence changes of the dyes in these arrays as they are exposed to analytes This provides chemical sensing with high sensitivity (often down to parts per billion levels), impressive discrimination among very similar analytes, and exquisite fingerprinting of extremely similar mixtures over a wide range of analyte types, in both the gas and liquid phases Design of both sensor arrays and instrumentation for their analysis are discussed In addition, the various chemometric and statistical analyses of high-dimensional data (including hierarchical cluster analysis (HCA), principal component analysis (PCA), linear discriminant analysis (LDA), support vector machines (SVMs), and artificial neural networks (ANNs)) are presented and critiqued in reference to their use in chemical sensing A variety of applications are also discussed, including personal dosimetry of toxic industrial chemical, detection of explosives or accelerants, quality control of foods and beverages, biosensing intracellularly, identification of bacteria and fungi, and detection of cancer and disease biomarkers

Publisher Summary The electroactivity of nucleic acids was discovered several years ago. It was shown that at mercury electrodes, adenine and cytosine were reduced in ssDNA while guanine produced an anodic signal due to the oxidation of guanine reduction product. The DNA signals at mercury electrodes are highly sensitive to changes in DNA structure due to DNA denaturation and renaturation, as well as to minor structural changes resulting from DNA premelting and DNA damage. The changes in DNA structure are reflected not only by the DNA faradaic responses but also by non-faradaic signals due to adsorption/desorption of DNA. Several interesting principles have been used in the development of the DNA sensors—such as amplified electrochemical analysis, investigation of charge transport, and tracing of changes in conformation of DNA. The s ensors for DNA hybridization and DNA damage lead the field of the electrochemistry of nucleic acids. The chapter briefly summarizes some properties of the elimination voltammetry with linear scan (EVLS) method. Oscillographic polarography, at controlled alternating current, was used in the electrochemical analysis of nucleic acids.

Electrochemistry of nucleic acids.

There remains a great challenge in the sensitive detection of microRNA because of the short length and low abundance of microRNAs in cells. Here, we have demonstrated an ultrasensitive detection platform for microRNA by combining the tetrahedral DNA nanostructure probes and hybridization chain reaction (HCR) amplification. The detection limits for DNA and microRNA are 100 aM and 10 aM (corresponding to 600 microRNAs in a 100 μL sample), respectively. Compared to the widely used supersandwich amplification, the detection limits are improved by 3 orders of magnitude. The uncontrolled surface immobilization and consumption of target molecules that limit the amplification efficiency of supersandwich are eliminated in our platform. Hybridization chain reaction amplification of microRNA detection with a tetrahedral DNA nanostructure-based electrochemical biosensor.

A simple and robust “off−on” signaling genosensor platform with improved selectivity for single-nucleotide polymorphism (SNP) detection based on the electronic DNA hairpin molecular beacons has been developed. The DNA beacons were immobilized onto gold electrodes in their folded states through the alkanethiol linker at the 3′-end, while the 5′-end was labeled with a methylene blue (MB) redox probe. A typical “on−off” change of the electrochemical signal was observed upon hybridization of the 27−33 nucleotide (nt) long hairpin DNA to the target DNA, in agreement with all the hitherto published data. Truncation of the DNA hairpin beacons down to 20 nts provided improved genosensor selectivity for SNP and allowed switching of the electrochemical genosensor response from the on−off to the off−on mode. “Off−On” Electrochemical Hairpin-DNA-Based Genosensor for Cancer Diagnostics

A DNA molecular beacon approach was used for the analysis of interactions between DNA and Methylene Blue (MB) as a redox indicator of a hybridization event. DNA hairpin structures of different length and guanine (G) content were immobilized onto gold electrodes in their folded states through the alkanethiol linker at the 5'-end. Binding of MB to the folded hairpin DNA was electrochemically studied and compared with binding to the duplex structure formed by hybridization of the hairpin DNA to a complementary DNA strand. Variation of the electrochemical signal from the DNA-MB complex was shown to depend primarily on the DNA length and sequence used: the G-C base pairs were the preferential sites of MB binding in the duplex. For short 20 nts long DNA sequences, the increased electrochemical response from MB bound to the duplex structure was consistent with the increased amount of bound and electrochemically readable MB molecules (i.e. MB molecules that are available for the electron transfer (ET) reaction with the electrode). With longer DNA sequences, the balance between the amounts of the electrochemically readable MB molecules bound to the hairpin DNA and to the hybrid was opposite: a part of the MB molecules bound to the long-sequence DNA duplex seem to be electrochemically mute due to long ET distance. The increasing electrochemical response from MB bound to the short-length DNA hybrid contrasts with the decreasing signal from MB bound to the long-length DNA hybrid and allows an "off"-"on" genosensor development.

DNA interactions with a Methylene Blue redox indicator depend on the DNA length and are sequence specific

A new DNA modifier containing triazene, ferrocene, and activated ester functionalities was synthesized and applied for electrochemical grafting and characterization of DNA at glassy carbon (GC) and gold electrodes. The modifier was synthesized from ferrocenecarboxylic acid by attaching a phenyltriazene derivative to one of the ferrocene Cp rings, while the other Cp ring containing the carboxylic acid was converted to an activated ester. The modifier was conjugated to an amine-modified DNA sequence. For immobilization of the conjugate at Au or GC electrodes, the triazene was activated by dimethyl sulfate for release of the diazonium salt. The salt was reductively converted to the aryl radical which was readily immobilized at the surface. DNA grafted onto electrodes exhibited remarkable hybridization properties, as detected through a reversible shift in the redox potential of the Fc redox label upon repeated hybridization/denaturation procedures with a complementary target DNA sequence.
