This paper describes fabrication techniques for the fabrication of glassy carbon microstructures. Molding of a resin of poly furfuryl . alcohol using elastomeric molds yields polymeric microstructures, which are converted to free-standing glassy carbon microstructures by . 2 . heating T ; 500-11008C under argon. This approach allows the preparation of macroscopic structures several mm with microscopic . features ; 2 mm . Deformation of the molds during molding of the resin allows preparation of curved microstructures. Fabrication of glassy carbon microstructures by soft lithography

Sensing and quantification of DNA and proteins are becoming increasingly important in biochemistry, medicine and biotechnology. Traditional analytical techniques are lagging behind the demand for more information in less time, at a lower cost. An important step forward in this pursuit is to identify an analyte using its electrochemical behavior and to convert its presence and concentration into perceivable and distinct electrical signals. This review covers the strategies for electrochemical sensing of biomolecules, mainly, DNA and proteins by label-based and label-free approaches using disposable electrochemical printed chips and carbon nanotube based field effect transistors. Issues, such as ease of preparation, robustness, sensitivity, and realization of mass production of the detection strategies are also considered. A good coverage of the published literature, mostly, a wide treatment of original research articles reporting novel principles has been made. Finally, this review may help the researchers in developing an understanding of miniaturized electrochemical biosensors and their possible applications in medical and food science, with directions for future research.

Electrochemical Biosensors for Medical and Food Applications

The newly developed DNA-biosensor is a very promising tool for the investigation and study of the action of drugs specifically designed to interact with DNA. In this work the electrochemical reduction of nitroimidazole drugs was studied in the presence of DNA immobilized onto the surface of a glassy carbon electrode. This enabled preconcentration of the drug onto the electrode surface, which was then electrochemically reduced to the corresponding hydroxylamine which followed by reoxidation give the nitroso compound and subsequently an azoxycompound. Moreover, as the target of the nitroimidazole action was the DNA, the damage caused to DNA on the electrode surface by a reduction product of this drug could be detected in situ.

Voltammetric Behavior of Nitroimidazoles at a DNA-Biosensor

Publisher Summary The electrochemical reduction of nitroimidazoles follows a complex mechanism and in theory the nitro group is able to receive up to six electrons to form the corresponding amine. Under anaerobic and low oxygen pressure conditions metronidazole follows a reduction mechanism similar to that of nitrobenzene. Electrochemical reduction of nitroimidazole derivatives shows two reduction waves in aqueous acid media, the first involving four electrons and corresponding to the reduction of the nitro group to form the intermediate hydroxylamine (-NHOH) and the second involving two electrons and corresponding to the reduction of the hydroxylamine to amine (-NH2). Using three different electrode materials (bare glassy carbon electrode, mercury thin film electrode, and DNA biosensor), it has been verified that hydroxylamine formation involves four electrons and is pH dependent. This chapter shows that the DNA biosensor developed provides a new perspective to the research and study of the mechanism of action of nitroimidazoles with DNA.
