Genetic diversity is an area of concern for sustaining crop yield. Information on genetic relatedness/diversity among Gossypium arboreum L. cultivars/genotypes is scanty. We have used random amplified polymorphic DNA (RAPD) analysis to assess the genetic divergence/relationship among 30 genotypes/cultivars of G. arboreum. Of 45 primers surveyed, 63% were polymorphic. Out of the total number of loci amplified, 36% were polymorphic. The calculated genetic similarity between the cultivars/genotypes was in the range of 47.05–98.73%. Two genotypes, HK-244 and Entry-17, were the most distantly related. The average genetic relatedness among all the genotypes was 80.46%. However, most of the cultivated varieties showed a close genetic relationship, indicating a narrow genetic base in comparison to the non-cultivated germplasm. The calculated coefficients were used to construct a dendrogram using the unweighted pair group of arithmetic means (UPGMA) algorithm, which grouped the genotypes/cultivars into two major and three smaller clusters. The study is the first comprehensive analysis of the genetic diversity of G. arboreum germplasm and identifies cultivars that will be useful in extending the genetic diversity of cultivated varieties and future genome mapping projects.

In order to evaluate the genetic diversity of sea- island cotton (Gossypium barbadense), 237 commonly mapped SSR markers covering the cotton genome were used to genotype 56 sea- island cotton accessions. A total of 218 polymorphic primer pairs (91.98%) amplified 361 loci, with a mean of 1.66 loci. Polymorphism information content values of the SSR primers ranged from 0.035 to 0.862, with a mean of 0.320. The highest mean polymorphism information content value for the SSR motifs was from a compound motif (0.402), and for the chromosomes it was Chr10 (0.589); the highest ratio of polymorphic primers in Xinjiang accessions was from Chr21 (83.33%). Genetic diversity was high in Xinjiang accessions. AMOVA showed that variation was 8 and 92% among populations and within populations, respectively. The 56 sea-island accessions were divided into three groups in the UPGMA dendrogram: Xinhai5 was in the first group; accessions from Xinjiang, except the five main ones, were in the second group, and the other 34 accessions

Genetic diversity and identification of some Turkish cotton genotypes (Gossypium hirsutum L.) by RAPD-PCR analysis

Random Amplified Polymorphic DNA (RAPD) analysis was used to evaluate genetic diversity among commercial Indian cotton varieties. Fifteen varieties belonging to Gossypium hirsutum L and seven to G. arboreum L were analyzed with 50 random decamer primers using the polymerase chain reaction (PCR). Twenty six of the primers detected polymorphism in all 22 cotton varieties. A total of 371 bands were amplified, 87% of which were polymorphic. Cluster analysis by the unweighted pair group method of arithmetic means (UPGMA) showed that diploids and tetraploids can be divided in two groups at a similarity of 30%. Diploid variety C402W showed the least similarity to all the others in the group. Among tetraploids, closely related varieties Pusa 8-6, 4515 and RS 875 were distinctly different from the rest. The analysis revealed that the intervarietal genetic relationships of several varieties is related to their pedigree. The results also revealed that tetraploids show a much narrower genetic base (similarity range of 65–95%) than the diploids (similarity range 54–88%). The results obtained can be used for the selection of parents to generate a mapping population and begin a breeding programme.
