Determination of carboplatin in plasma and tumor by high-performance liquid chromatography-mass spectrometry

Firstly, metformin and repaglinide were degraded under high temperature/humidity, UV/VIS light, in different pH and oxidative conditions. Secondly, a new validated LC-UV method was examined, as to whether it validly determined these drugs in the presence of their degradation products and whether it is suitable for estimating degradation kinetics. Finally, the respective LC-MS method was used to identify the degradation products. In addition, using FT-IR method, the stability of metformin and repaglinide was scrutinized in the presence of polyvinylpyrrolidone (PVP), mannitol, magnesium stearate, and lactose. Significant degradation of metformin, following the first order kinetics, was observed in alkaline medium. In the case of repaglinide, the most significant and quickest degradation, following the first order kinetics, was observed in acidic and oxidative media (0.1 M HCl and 3% H2O2). Two new degradation products of metformin and nine new degradation products of repaglinide were detected and identified when the stressed samples were examined by our LC-MS method. What is more, the presence of PVP, mannitol, and magnesium stearate proved to affect the stability of metformin, while repaglinide stability was affected in the presence of PVP and magnesium stearate.

Determination of Chemical Stability of Two Oral Antidiabetics, Metformin and Repaglinide in the Solid State and Solutions Using LC-UV, LC-MS, and FT-IR Methods.

Hypertension is a condition in which blood pressure is elevated to an extent where benefit is obtained from blood pressure lowering. The risk of complications is proportional to the level that blood pressure raises. Calcium channel blockers are a class of compounds used in the treatment of hypertension. The dihydropyridine (DHP) group, a subclass of the calcium channel blocker works almost exclusively on L-type calcium channels in the peripheral arterioles and reduce blood pressure by reducing total peripheral resistant. Long acting DHP is preferred because they are more convenient for patients and avoid the large fluctuations in plasma drug concentration which are associated with side effects. Amlodipine is the most distinct DHP and the most popular. The drug was patented in the year 1986 and its commercial sale began by 1990. The current article provides a state of art about the analytical and bioanalytical techniques available for the quantification of drug as a single entity and in combined pharmaceutical formulations between 1989 and 2019.

Analytical and Bioanalytical Techniques for the Quantification of the Calcium Channel Blocker - Amlodipine: A Critical Review.

In this present study, we investigated the utilization of whey powder as an alternate carbon source and statistical optimization of medium components on lovastatin production by Aspergillus terreus MTCC 1281. Central composite design (CCD) of response surface methodology (RSM) was successfully employed to optimize the four medium variables whey powder, yeast extract, sodium chloride, and copper sulphate at five levels. Regression analysis explained that the linear and square effects of the variables were found significant with R2 value of 0.937. Maximum lovastatin production of 358.2 mg/L was obtained with the predicted optimal concentration of whey powder 1.5%, yeast extract 0.75%, sodium chloride 0.04%, and copper sulphate 0.019%.The results of this study indicated that statistical optimization approach leads to high lovastatin production by using whey powder as a carbon source.

Utilization of whey powder as an alternate carbon source for production of hypocholesterolemic drug by Aspergillus terreus MTCC 1281

A simple, specific, accurate and precise HPTLC method has been developed for the determination of Artesunate and Mefloquine Hydrochloride in bulk dosage form by using HPTLC plates (Merck) precoated with silica gel 60F254 on aluminum sheets, with a band width of 5 mm using Camag 20 µl syringe and Camag – Linomat IV Sample Applicator. The sample was analysed using Methanol: Chloroform in the ratio of 4 : 1 v/v and detected by absorbance mode at 230nm. The assay was performed and validated extensively for specificity, linearity, accuracy, precision, recovery, limit of quantization and detection .The proposed method was precise, rapid, accurate, specific and reproducible.

Method development and validation of artesunate and mefloquine hydrochloride in bulk and dosage form by hptlc

The present work deals with method development and analytical validation of a novel, precise and accurate HPTLC methods for the simultaneous estimation of Amlodipine besylate and Olmesartan medoxomil. Literature review has shown that High performance liquid chromatography and UV-Visible Spectroscopy methods have been reported for the estimation of these drugs, but no HPTLC method has been done, thus this study had to be done. A analytical method development for the simultaneous estimation of Amlodipine besylate and Olmesartan medoxomil was developed. For simultaneous HPTLC method, the analytical separation was achieved on aluminium plates pre-coated with Silica Gel 60 F254 Acetonitrile: Water: Toluene (6:3:1) v/v/v used as the mobile phase with densitometric scanning at 270 nm. Good and acceptable linearity was obtained for the drug in the range of 5 - 15 μg/mL with r2 > 0.999. All the precision measurements made were well within the acceptable limits. The percentage recovery was found to be 99.59 & 99.61 % HPTLC method for Amlodipine besylate and Olmesartan medoxomil respectively. The parameters of validation were in accordance with the outlined ICH guidelines for method development in the estimation of drugs. Hence in conclusion the method can be applied day today lab practice for the determination of Amlodipine besylate and Olmesartan medoxomil simultaneously using HPTLC instrument.

Method Development and Validation of simultaneous estimation for Amlodipine besylate and Olmesartan medoxomil by HPTLC method

High incidences and cancer cases in the world for the past decade have resulted in the need to conduct extensive research on the cancer medications. Carboplatin is a platinum compound used mainly in the treatment of lung, head and neck, as well as many other cancers. No method has been reportedly done and developed for the routine analysis of carboplatin using UV-Vis. Many have reported HPLC and Mass spectroscopy. A method was developed and validated for the analysis of carboplatin in the standard and injection form. 10mg of the carboplatin standard was transferred into a 10ml volumetric flask, and diluted up to the mark using 0.1M NaOH. The solution was further diluted to a 1μg/ml solution and scanned using UV-Vis. The ʎ-max was found to be 295nm. A calibration curve was plotted and at 2-10μg/ml Beer’s law was obeyed. The Linear equation was y=0.0564x+0.0057. The r value was 0.9996, which was within acceptable limits. Precision using inter-day and intra-day precision were done yielding %RSD values of 0.734 and 0.504 respectively, which were acceptable according to ICH guidelines (≤2). A method was therefore successfully developed and validated for routine use in the analysis of Carboplatin in the standard and parenteral dosage form.

Method development and validation for the estimation of carboplatin in pharmaceutical pure and dosage form using UV-Vis method

A new simple, precise, sensitive, economical RP-HPLC method was developed for the estimation of Metformin hydrochloride and Repaglinide in bulk and tablet dosage form. The ?max for Metformin hydrochloride and Repaglinide was found to be nm and nm respectively. The linearity in the concentration range of 25?g/ml to 150 ?g/ml (r2=0.999) and 0.1 to 0.6 ?g/ml (r2=0.999) for Metformin hydrochloride and Repaglinide respectively. The chromatographic values for Metformin hydrochloride and Repaglinide were found to be satisfactory on BDS Hypersil 18, 250 x 4.6mm, 5?m, column using mobile phase of ACN: Buffer in the ratio of 58: 42 v/v with the flow rate of 1.0ml/min. The developed method was validated according to ICH guidelines and found to be accurate, precise, rugged and robust and found to be in good accordance with the prescribed values. The newly developed method can be used for routine analysis estimation of Metformin hydrochloride and Repaglinide in bulk and tablet dosage form in pharmaceutical industry.
