Most biofilms in their natural environments are likely to consist of consortia of species that influence each other in synergistic and antagonistic manners. However, few reports specifically address interactions within multispecies biofilms. In this study, 17 epiphytic bacterial strains, isolated from the surface of the marine alga Ulva australis, were screened for synergistic interactions within biofilms when present together in different combinations. Four isolates, Microbacterium phyllosphaerae, Shewanella japonica, Dokdonia donghaensis, and Acinetobacter lwoffii, were found to interact synergistically in biofilms formed in 96-well microtiter plates: biofilm biomass was observed to increase by >167% in biofilms formed by the four strains compared to biofilms composed of single strains. When exposed to the antibacterial agent hydrogen peroxide or tetracycline, the relative activity (exposed versus nonexposed biofilms) of the four-species biofilm was markedly higher than that in any of the single-species biofilms. Moreover, in biofilms established on glass surfaces in flow cells and subjected to invasion by the antibacterial protein-producing Pseudoalteromonas tunicata, the four-species biofilms resisted invasion to a greater extent than did the biofilms formed by the single species. Replacement of each strain by its cell-free culture supernatant suggested that synergy was dependent both on species-specific physical interactions between cells and on extracellular secreted factors or less specific interactions. In summary, our data strongly indicate that synergistic effects promote biofilm biomass and resistance of the biofilm to antimicrobial agents and bacterial invasion in multispecies biofilms.

The role of bacterial surface components in combination with bacterial functional signals in the process of biofilm formation has been increasingly studied in recent years. Plants support a diverse array of bacteria on or in their roots, transport vessels, stems, and leaves. These plant-associated bacteria have important effects on plant health and productivity. Biofilm formation on plants is associated with symbiotic and pathogenic responses, but how plants regulate such associations is unclear. Certain bacteria in biofilm matrices have been found to induce plant growth and to protect plants from phytopathogens (a process termed biocontrol), whereas others are involved in pathogenesis. In this review, we systematically describe the various components and mechanisms involved in bacterial biofilm formation and attachment to plant surfaces and the relationships of these mechanisms to bacterial activity and survival.

The Role of Bacterial Biofilms and Surface Components in Plant-Bacterial Associations

In this study, aerobic granules to treat wastewater containing p-nitrophenol (PNP) were successfully developed in a sequencing batch reactor (SBR) using activated sludge as inoculum. A key step was the conditioning of the activated sludge seed to enrich for biomass with improved settleability and higher PNP degradation activity by implementing progressive decreases in settling time and stepwise increases in PNP concentration. The aerobic granules were cultivated at a PNP loading rate of 0.6 kg/ m3 x day, with glucose to boost the growth of PNP-degrading biomass. The granules had a clearly defined shape and appearance, settled significantly faster than activated sludge, and were capable of nearly complete PNP removal. The granules had specific PNP degradation rates that increased with PNP concentration from 0 to 40.1 mg of PNP/L, peaked at 19.3 mg of PNP/(g of VSS) x h (VSS = volatile suspended solids), and declined with further increases in PNP concentration as substrate inhibition effects became significant. Batch incubation experiments show that the PNP-degrading granules could also degrade other phenolic compounds, such as hydroquinone, p-nitrocatechol, phenol, 2,4-dichlorophenol, and 2,6-dichlorophenol. The PNP-degrading granules contained diverse microbial morphotypes, and PNP-degrading bacteria accounted for 49% of the total culturable heterotrophic bacteria. Denaturing gradient gel electrophoresis analysis of 16S rRNA gene fragments showed a gradual temporal shift in microbial community succession as the granules developed from the activated sludge seed. Specific oxygen utilization rates at 100 mg/L PNP were found to increase with the evolution of smaller granules to large granules, suggesting that the granulation process can enhance metabolic efficiency toward biodegradation of PNP. The results in this study demonstrate that it is possible to use aerobic granules for PNP biodegradation and broadens the benefits of using the SBR to target treatment of toxic and recalcitrant organic compounds.

Characterisation of heterotrophic nitrifying and aerobic denitrifying Klebsiella pneumoniae CF-S9 strain for bioremediation of wastewater

Thirty-two strains of nonflocculating bacteria isolated from sewage-activated sludge were tested by a spectrophotometric assay for their ability to coaggregate with one other in two-membered systems. Among these strains, eight showed significant (74 to 99%) coaggregation with Acinetobacter johnsonii S35 while only four strains coaggregated, to a lesser extent (43 to 65%), with Acinetobacter junii S33. The extent and pattern of coaggregation as well as the aggregate size showed good correlation with cellular characteristics of the coaggregating partners. These strains were identified by sequencing of full-length 16S rRNA genes. A. johnsonii S35 could coaggregate with strains of several genera, such as Oligotropha carboxidovorans, Microbacterium esteraromaticum, and Xanthomonas spp. The role of Acinetobacter isolates as bridging organisms in multigeneric coaggregates is indicated. This investigation revealed the role of much-neglected nonflocculating bacteria in floc formation in activated sludge.

Coaggregation among Nonflocculating Bacteria Isolated from Activated Sludge

The effects of different cultivation periods and growth media on the cell surface hydrophobicity and coaggregation ability of Acinetobacter johnsonii S35 with Oligotropha carboxidovorans S23 (both sewage sludge isolates) were examined. It was observed that when cultivated in polypeptone medium, A. johnsonii S35 shows high hydrophobicity and strong coaggregation (98% aggregation index) with O. carboxidovorans S23, which remain unaltered after a 1-day, 3-day or 7-day cultivation period. When A. johnsonii S35 was cultivated with different carbon substrates, large coaggregates and a high aggregation index were observed using acetate-grown cells (96%), followed by ethanol-(83%) and vegetable oil-grown cells (72%). Coaggregates were small and narrow in ethanol-grown cells and still smaller in case of the vegetable oil-grown cells. Although no direct correlation could be observed, more hydrophobic cells showed a higher aggregation index. None of the culture supernatants of A. johnsonii S35 showed emulsification of hexadecane or a significant ability to aggregate O. carboxidovorans S23 cells, indicating that the coaggregation factor of A. johnsonii S35 is cell-bound. These results demonstrate that the coaggregation ability of A. johnsonii S35 is constitutive and is not lost under diverse cultivation conditions.
