Fine magnetic particles containing useful proteins bound thereto, which comprise fine magnetic particles formed in the cells of a magnetic bacterium and a hybrid protein bound to the organic membranes covering the particles, the hybrid protein being one composed of a membrane protein inherently bound to the organic membranes and one or more useful proteins bound thereto biologically by, for example, fusion. The biologically immobilized proteins do not undergo activity reduction. Enzymes, antibodies and other useful proteins can be obtained in a state immobilized onto fine magnetic particles merely by culturing a transformed magnetic bacterium and separating the fine magnetic particles formed in the bacterial cells. When a functional protein is thus immobilized, the function of such a protein can be exhibited efficiently at a desired location because the protein can be controlled magnetically. As it is possible to genetically control the site of protein expression, it becomes possible to conduct a highly sensitive detection when a binding protein such as protein A and a marker protein such as a photoprotein are expressed in proximity to each other on the magnetic particles.

PURPOSE:To enable accurate immunological measurement of hemoglobin in excrement, by making a glycosidase type bacteriolysis enzyme act upon a sample. CONSTITUTION:An excrement sampling spoon is coated with a buffer containing an antihuman hemoglobin antibody by being immersed thereinto for 24hr. The spoon sticks into excrements to sample a fixed amount thereof. On the other hand, NaN3 and a bacteriolysis enzyme such as lysozyme are added to an agar solution to gelatinize. The excrement sampling spoon having excrements sampled is inserted into the agar gel and allowed to stand for a fixed time. Then the spoon is taken out and washed to be immersed into an alkali phosphodase labelled antibody solution. After washed, the spoon is immersed into a phenyl phosphate substrate liquid to incubate and after the addition of a coloring liquid, reaction is stopped to measure the degree of coloration.

Detection of hemoglobin in excrement

PROBLEM TO BE SOLVED: To provide a method for specifically detecting patients affected by pneumonia/bronchitis by Streptococcus pneumoniae by rapidly and simply detecting without invasive pain the antigen protein derived from Streptococcus pneumoniae in urine or serum specimens from patients infected with Streptococcus pneumoniae. SOLUTION: The method for testing the disease caused by Streptococcus pneumoniae of the patient includes detecting ribosomal protein L7/L12 of Streptococcus pneumoniae in the urine or serum of the patient using the antibodies against ribosomal protein L7/L12 of Streptococcus pneumoniae. Detection method of streptococcus pneumoniae

PURPOSE:Immobilized magnetic fine particles of a physiologically active substance, obtained by separating from magnetotactic bacteria, having a large surface area due to the small particle size and very large amount of the supported physiologically active substance. CONSTITUTION:The above-mentioned magnetic fine particles are separated from magnetotactic bacteria and can be used even in a state of either being covered with a coating film consisting mainly of a protein or free from the coating film. In general, the magnetic fine particles having the coating film has an advantage in that the harmony with the living body is good. In order to collect the above-mentioned magnetic fine particles or separate the particles from magnetotactic bacteria gathered by culture fluid, the concentration by a micro-centrifuge is carried out and the bacterial walls are dissolved with a lysozyme solution. The centrifugation may then be repeated. Examples of the kind of the physiologically active substance to be immobilized by the separated magnetic fine particles include enzymes, e.g. hydrolase or oxidoreductase, immunity-related substances, growth factors, nucleic acid-related substances, physiologically active substances derived from plants and other physiologically active substances.
