The quest for alternative drugs to the well-known cisplatin and its derivatives, which are still used in more than 50% of the treatment regimes for patients suffering from cancer, is highly needed.1,2 Despite their tremendous success, these platinum compounds suffer from two main disadvantages: they are inefficient against platinum-resistant tumors, and they have severe side effects such as nephrotoxicity. The latter drawback is the consequence of the fact that the ultimate target of these drugs is ubiquitous: It is generally accepted that Pt anticancer drugs target DNA, which is present in all cells.3,4 Furthermore, as a consequence of its particular chemical structure, cisplatin in particular offers little possibility for rational improvements to increase its tumor specificity and thereby reduce undesired side effects.

In this context, organometallic compounds, which are defined as metal complexes containing at least one direct, covalent metal−carbon bond, have recently been found to be promising anticancer drug candidates. Organometallics have a great structural variety (ranging from linear to octahedral and even beyond), have far more diverse stereochemistry than organic compounds (for an octahedral complex with six different ligands, 30 stereoisomers exist!), and by rational ligand design, provide control over key kinetic properties (such as hydrolysis rate of ligands). Furthermore, they are kinetically stable, usually uncharged, and relatively lipophilic and their metal atom is in a low oxidation state. Because of these fundamental differences compared to “classical coordination metal complexes”, organometallics offer ample opportunities in the design of novel classes of medicinal compounds, potentially with new metal-specific modes of action. Interestingly, all the typical classes of organometallics such as metallocenes, half-sandwich, carbene-, CO-, or π-ligands, which have been widely used for catalysis or biosensing purposes, have now also found application in medicinal chemistry (see Figure ​Figure11 for an overview of these typical classes of organometallics).

In this Perspective, we report on the recent advances in the discovery of organometallics with proven antiproliferative activity. We are emphasizing those compounds where efforts have been made to identify their molecular target and mode of action by biochemical or cell biology studies. This Perspective covers more classes of compounds and in more detail than a recent tutorial review by Hartinger and Dyson.(5) Furthermore, whereas recent reviews and book contributions attest to the rapid development of bioorganometallic chemistry in general,6,7 this Perspective focuses on their potential application as anticancer chemotherapeutics. Another very recent review article categorizes inorganic anticancer drug candidates by their modes of action.(8) It should be mentioned that a full description of all currently investigated types of compounds is hardly possible anymore in a concise review. For example, a particularly promising class of organometallic anticancer compounds, namely, radiolabeled organometallics, has been omitted for space limitations. Recent developments of such compounds have been reviewed in detail by Alberto.(9)

Organometallic Anticancer Compounds

In undergraduate level organometallic chemistry courses students are usually taught that organometallic compounds are toxic and unstable in air and water. While this is true of many complexes, some are also non-toxic and stable in air and water. Indeed, bioorganometallic chemistry, the study of biomolecules or biologically active molecules that contain at least one carbon directly bound to a metal, is a thriving subject, and air and water stability is a general pre-requisite. This interdisciplinary field is located at the borderline between chemistry, biochemistry, biology and medicine. In this tutorial review, various aspects of bioorganometallic chemistry are introduced, with the main emphasis on medicinal organometallic compounds. Organometallic therapeutics for cancer, HIV and malaria and other medicinal applications are described. It is also shown how rational ligand design has led to new improved therapies much in the same way that an organometallic chemist working in catalysis will design new ligands for improved activities.

Bioorganometallic chemistry—from teaching paradigms to medicinal applications

Anti-cancer drugs

In recent decades, it has generally been recognized that carbenes play an important role as transient intermediates. As a result of a number of stable carbenes having been isolated and investigated in detail, it is not an exaggeration to say that the chemistry of carbenes has been thoroughly investigated and is now well-understood.1 In addition, much attention has also been paid to the heavier analogues of carbenes, i.e., silylenes (R2Si:), germylenes (R2Ge:), stannylenes (R2Sn:), and plumbylenes (R2Pb:). These so-called metallylenes are monomeric species of the polymetallanes. This is especially true of the silylenes, which are believed to be monomers of polysilane. The metallylenes could be expected to be of great importance in fundamental and applied chemistry as a result of their many differences and similarities to carbenes. The valency of the central atom of the heavier carbene analogues (R2M:, M ) Si, Ge, Sn, Pb) is two. That is, its oxidation state is MII and its stability increases as the principal quantum number (n) increases. In fact, dichloroplumbylene and dichlorostannylene, PbCl2 and SnCl2, respectively, are very stable ionic compounds. However, these dihalides exist as polymers or ion pairs both in solution and in the solid state. The dichlorogermylene complex GeCl2 · (dioxane)3 is also known to be stable and isolable, whereas the dihalosilylenes are barely isolable compounds.2 The early silylene research was concerned largely with comparing the chemistry of the dihalosilylenes with that of carbenes. Hence, the chemistry of the metallylenes has been considered mainly from the molecular chemistry point of view.4 In contrast to the carbon atom, the heavier group 14 atoms have a low ability to form hybrid orbitals. They therefore prefer the (ns)2(np)2 valence electron configurations in their divalent species.5 Since two electrons remain as a singlet pair in the ns orbital, the ground state of H2M: (M ) Si, Ge, Sn, Pb) is a singlet, unlike the case of H2C:, where the ground state is a triplet (Figure 1).1a On the basis of theoretical calculations, the singlet-triplet energy differences ∆EST for H2M, [∆EST ) E(triplet) E(singlet)], are found to be 16.7 (M ) Si), 21.8 (M ) Ge), 24.8 (M ) Sn), and 34.8 (M ) Pb) kcal/mol, respectively. That of H2C: is estimated as -14.0 kcal/mol.6 Furthermore, the relative stabilities of the singlet species of R2M: (M ) C, Si, Ge, Sn, Pb; R ) alkyl or aryl) compared to the corresponding dimer, R2MdMR2, are estimated to increase as the element row descends, C < Si < Ge < Sn < Pb. It follows, therefore, that one can expect that a divalent organolead compound such as plumbylene should be isolable as a stable compound. However, some plumbylenes, without any electronic or steric stabilization effects, are known to be thermally unstable and undergo facile disproportionation reactions, giving rise to elemental lead and the corresponding tetravalent organolead compounds.7 On this basis, it could be concluded that it might be difficult to isolate metallylenes as stable compounds under ambient conditions, since they generally exhibit extremely high reactivity toward other molecules as well as themselves. Metallylenes have a singlet ground state with a vacant p-orbital and a lone pair of valence orbitals. This extremely high reactivity must be due to their vacant p-orbitals, since 6 valence electrons is less than the 8 electrons of the “octet rule”. Their lone pair is expected to be inert due to its high s-character. In order to stabilize metallylenes enough to be isolated, either some thermodynamic and/or kinetic stabilization of the reactive vacant p-orbital is required (Figure 2). A range of “isolable” metallylenes have been synthesized through the thermodynamic stabilization of coordinating Cp* ligands, the inclusion of heteroatoms such as N, O, and P, * To whom correspondence should be addressed. Stable Heavier Carbene Analogues

This tutorial review compiles the advances that have been achieved in using transition metal complexes containing N-heterocyclic carbene ligands as components for materials. Applications of metal carbene complexes in fields different from catalysis are remarkably scarce. During the last few years, promising results have been accomplished in particular by utilizing such complexes as antimicrobial and cytotoxic agents, as photoactive sites in luminescent materials, for self-assembly into liquid crystalline materials and metallosupramolecular structures, and as synthons for molecular switches and conducting polymeric materials. These initial achievements clearly underline the great potential of N-heterocyclic carbene complexes in various fields of materials science.

Beyond catalysis: N-heterocyclic carbene complexes as components for medicinal, luminescent, and functional materials applications.

The Tetrameric Aluminum(I) Compound [{Al(η5‐C5Me5)}4]

6-Substituted fulvenes are interesting and easily accessible starting materials for the synthesis of novel substituted titanocenes via reductive dimerisation, carbolithiation or hydridolithiation reactions, which are followed by a transmetallation reaction with titanium tetrachloride in the latter two cases. Depending on the substitution pattern, these titanocenes prove to be bioorganometallic anti-cancer drugs, which have significant potential against advanced or metastatic renal-cell cancer. Patients bearing these stages of kidney cancer have a poor prognosis so far and therefore real progress in the area of metal-based anti-cancer drugs may come from this simple and effective synthetic approach. This tutorial review provides an insight into the synthesis of fulvene-derived titanocenes and their activity in preclinical experiments.

Bioorganometallic fulvene-derived titanocene anti-cancer drugs

Die tetramere Aluminium(I)‐Verbindung [{Al(η5‐C5Me5)}4]

From the reaction of 1-methylimidazole (1a), 4,5-dichloro-1H-imidazole (1bII) and 1-methylbenzimidazole (1c) with p-cyanobenzyl bromide (2a), non-symmetrically substituted N-heterocyclic carbene (NHC) [(3a–c)] precursors, 5,6-dimethyl-1H-benzimidazole (1d) and 4,5-diphenyl-1H-imidazole (1e) with p-cyanobenzyl bromide (2a) and benzyl bromide (2b), symmetrically substituted N-heterocyclic carbene (NHC) [(3d–f)] precursors were synthesised. These NHC-precursors were then reacted with silver(I) acetate to yield the NHC–silver complexes (1-methyl-3-(4-cyanobenzyl)imidazole-2-ylidene)silver(I)acetate (4a), (4,5-dichloro-1-(4-cyanobenzyl)-3-methyl)imidazole-2-ylidene)silver(I)acetate (4b), (1-methyl-3-(4-cyanobenzyl)benzimidazole-2-ylidene)silver(I)acetate (4c), (1,3-bis(4-cyanobenzyl)5,6-dimethylbenzimidazole-2-ylidene) silver(I) acetate (4d), (1,3-dibenzyl-5,6-dimethylbenzimidazole-2-ylidene) silver(I) acetate (4e) and (1,3-dibenzyl-4,5-diphenylimidazol-2-ylidene) silver(I) acetate (4f) respectively. Three NHC-precursors 3c–e and four NHC–silver complexes 4b and 4d–f were characterised by single crystal X-ray diffraction. Preliminary in vitro antibacterial activity of the NHC-precursors and NHC–silver complexes was investigated against Gram-positive bacteria Staphylococcus aureus, and Gram-negative bacteria Escherichia coli using the qualitative Kirby–Bauer disk-diffusion method. NHC–silver complexes have shown very high antibacterial activity compared to the NHC-precursors. All six NHC–silver complexes were tested for their cytotoxicity through MTT based in vitro tests on the human renal-cancer cell line Caki-1 in order to determine their IC50 values. NHC–silver complexes 4a–f were found to have IC50 values of 6.2 (±1.0), 7.7 (±1.6), 1.2 (±0.6), 10.8 (±1.9), 24.2 (±1.8) and 13.6 (±1.0) μM, respectively. These values represent improved cytotoxicity against Caki-1, most notably for 4c, which is a three times more cytotoxic than cisplatin (IC50 value = 3.3 μM) itself.

Novel benzyl-substituted N-heterocyclic carbene–silver acetate complexes: synthesis, cytotoxicity and antibacterial studies

Late transition metal complexes that bear N-heterocyclic carbene (NHC) ligands have seen a speedy growth in their use as both, metal-based drug candidates and potentially active homogeneous catalysts in a plethora of C-C and C-N bond forming reactions. This review article focuses on the recent developments and advances in preparation and characterization of NHC-metal complexes (metal: silver, gold, copper, palladium, nickel and ruthenium) and their biomedical applications. Their design, syntheses and characterization have been reviewed and correlated to their antimicrobial and anticancer efficacies. All these initial discoveries help validate the great potential of NHC-metal derivatives as a class of effective antimicrobial and anticancer agents.
