Albumin is an appealing carrier in nanomedicine because of its unique features. First, it is the most abundant protein in plasma, endowing high biocompatibility, biodegradability, nonimmunogenicity, and safety for its clinical application. Second, albumin chemical structure and conformation allows interaction with many different drugs, potentially protecting them from elimination and metabolism in vivo, thus improving their pharmacokinetic properties. Finally, albumin can interact with receptors overexpressed in many diseased tissues and cells, providing a unique feature for active targeting of the disease site without the addition of specific ligands to the nanocarrier. For this reason, albumin, characterized by an extended serum half-life of around 19 days, has the potential of promoting half-life extension and targeted delivery of drugs. Therefore, this article focuses on the importance of albumin as a nanodrug delivery carrier for hydrophobic drugs, taking advantage of the passive as well as active targeting potential of this nanocarrier. Particular attention is paid to the breakthrough NAB-Technology, with emphasis on the advantages of Nab-Paclitaxel (Abraxane), compared to the solvent-based formulations of Paclitaxel, i.e., CrEL-paclitaxel (Taxol) in a clinical setting. Finally, the role of albumin in carrying anticancer compounds is depicted, with a particular focus on the albumin-based formulations that are currently undergoing clinical trials. The article sheds light on the power of an endogenous substance, such as albumin, as a drug delivery system, signifies the importance of the drug vehicle in drug performance in the biological systems, and highlights the possible future trends in the use of this drug delivery system.

The Uniqueness of Albumin as a Carrier in Nanodrug Delivery.

The present paper aims to summarize the results regarding serum albumin-based nanoparticles (NPs) for drug delivery purposes. In particular, it focuses on the relationship between their preparation techniques and synthesis parameters, as well as their successful clinical application. In spite of the huge amount of consumed material and immaterial sources and promising possibilities, products made from different types of albumin NPs, with the exception of a few, still have not been invented. In the present paper, promising applications of serum albumin nanoparticles (SANPs) for different biomedical purposes, such as carriers, delivery systems and contrast agents, are also discussed. The most frequent utilization of the NPs for certain diseases, i.e., cancer therapy, and future prospects are also detailed in this study.

Serum Albumin Nanoparticles: Problems and Prospects.

Albumin-based nanoparticles are an emerging platform for the delivery of various chemotherapeutics because of their biocompatibility, safety, and ease of surface modification for specific targeting. The most widely used method for the preparation of albumin nanoparticles is by desolvation process using glutaraldehyde (GLU) as a cross-linker. However, limitations of GLU like toxicity and interaction with drugs force the need for alternative cross-linkers. In the present study, several cross-linking systems were evaluated for the preparation of Bovine Serum Albumin (BSA) nanoparticles (ABNs) encapsulating Doxorubicin (Dox). Based on the results obtained from morphological characterization, in vitro release, and therapeutic efficacy in cells, N-succinimidyl 3-(2-pyridyldithio) propionate (SPDP)-modified ABNs (ABN-SPDP) was chosen. Since ABN-SPDP are formed with disulfide linkage, the drug release is facilitated under a highly reducing environment present in the tumor sites. The cytotoxicity studies of those ABN-SPDP were performed in three different breast cell lines, highlighting the mechanism of cell death. The Dox-encapsulated ABN-SPDP showed toxicity in both the breast cancer cells (MCF-7 and MDA-MB-231), but, remarkably, a negligible effect was observed in non-tumoral MCF-10A cells. In addition to the hydrophilic Dox, this system could be used as a carrier for hydrophobic drugs like SN38. The system could be employed for the preparation of nanoparticles based on human serum albumin (HSA), which further enhances the feasibility of this system for clinical use. Hence, the albumin nanoparticles developed herein present an excellent potential for delivering various drugs in cancer therapy.

Albumin-Based Nanoparticles for the Delivery of Doxorubicin in Breast Cancer.

Albumin and functionalized albumin nanoparticles: production strategies, characterization, and target indications

A sensor array mimicking a chemical nose/tongue based on bovine serum albumin nanoparticles (BSANsn) has been developed for the fluorescence pattern recognition of metal ions in biofluids. Three types of BSANsn (BSANs10, BSANs20, and BSANs40) show the same excitation/emission peak at 478/526 nm. According to the differential fluorescence variation, the sensor array shows particular fluorescence response patterns depending upon metal ions. Upon principal component analysis (PCA), it was found that the sensor array can distinguish 18 metal ions clearly at a concentration of as low as 10 μM. Moreover, different concentrations of metal ions and mixed metal ions of diverse kinds or valence states can be differentiated by the sensor in biofluids. In addition, the results were well consistent with those obtained with the traditional ICP-AES method.

Fluorescent pattern recognition of metal ions by nanoparticles of bovine serum albumin as a chemical nose/tongue.

Albumin nanoparticles are considerable in nanotechnology due to their high solubility in aqueous media, their ability to interact with various compounds, and their ease of nanoparticle tailoring from them. Due to the undesirable effects of glutaraldehyde (GT) as a cross-linker, the aim of this study was to investigate different cross-linking agents such as tannic acid, ascorbic acid, citric acid, sorbitol, and glucose (GLU) and to optimize particle production. For this purpose, final particle size, polydispersity index, and zeta potential were measured. Nanoparticles of different cross-linkers were compared to those prepared with GT using UV–Vis and FTIR spectra. Optimal conditions were defined as 20 mg/mL bovine serum albumin (BSA) solution (2 mL), pH 9, temperature of 4 °C, ethanol as desolvation agent, and GLU as cross-linker. BSA nanoparticles were obtained 125.1 nm in size, with a polydispersity index of 0.132 at the optimum point of the desolvation process. Studying FTIR and UV–Vis results showed that cross-linking occurred in the presence of all cross-linkers.
