Abstract: This study was performed to determine the antimelanogenic effect and tyrosinase inhibitory activities of anthocyanin rich fraction (AN-SLP) from Liriope platyphylla Wang et Tang seeds. Anthocyanins isolated from L. platyphylla seeds revealed the presence of four major anthocyanin components, which were tentatively identified as delphinidin-3-O- glucoside, delphinidin-3-O-rutinoside, petunidin-3-O-rutinoside, and malvidin-3-O-rutinoside using semipreparative HPLC, 1 H-NMR, 13 C NMR, FAB-MS and LC/ES-MS. The inhibitory effect of AN-SLP on tyrosinase activity was studied using in vitro (against mushroom tyrosinase) and ex vivo (against B16 melanoma cell tyrosinase) models. Cellular tyrosinase activity was decreased by AN-SLP treatment in B 16 melanoma cells through dose dependent manner, but AN-SLP did not inhibit mushroom tyrosinase and L-DOPA oxidation directly. AN-SLP showed melanin inhibition by 53.2% at 50 ㎍/㎖ which was 0.7 times more efficient than the antimelanogenic effect of commercial arbutin and kojic acid (36.5%) also did not show cell toxicity. Additionally, AN-SLP inhibited the activity of α-glucosidase and the glycosylation of tyrosinase in melanoma cell. The resulting unsaturated glycosylation of tyrosinase makes it unstable and disturb correct transportation. From theses results, we conclude that AN-SLP could be used as anti-melanogenic agent for skin whitening.

Abstract: Cyanidin-3-O-(2″-xylosyl)-glucoside (C-3-O-(2″-xylosyl)-G) was analyzed as an active constituent from the fruit of Siberian ginseng (Acanthopanax senticosus) using a HPLC diode array detection-electrospray ionization/mass spectrometry analysis system and the effect of C-3-O-(2″-xylosyl)-G on UVB-induced inflammatory signaling in JB6 P+ cells was investigated. C-3-O-(2″-xylosyl)-G inhibited UVB-induced cyclooxygenase (COX)-2 expression and promoter binding activity in JB6 P+ cells and JB6 P+ cells stably transfected with the COX-2 luciferase reporter plasmid. It inhibited both the UVB-induced activator protein-1 (AP-1) and nuclear factor (NF)-κB transactivation in JB6 P+ cells stably transfected with the AP-1 and NF-κB luciferase reporter plasmids. Additionally, C-3-O-(2″-xylosyl)-G significantly suppressed UVB-induced upregulated phosphorylation of c-Jun N terminal kinase, MEK/extracellular signaling kinase, and mitogen activated protein kinase kinase (MAPKK) 3/6 in JB6 P+ cells. These results indicate that C-3-O-(2″-xylosyl)-G may be a promising chemopreventive material that acts by suppressing COX-2 expression and AP-1 and NF-κB transactivation and JNK, MAPKK3/6, and MEK/ERK1/2 phosphorylation.

Abstract: This study was carried out to examine whether the leaf size is likely to be used as a selection criterion for large seed genotype in soybean (Glycine. max (L.) Merr.) breeding program. Two hundred twenty nine soybean germplasms which had collected in Korea, United States, China and Japan were used in this experiment. The area of unifoliate leaf, middle leaflet of first trifoliate and third trifoliate leaf ranged from 3.2 cm 2 to 33.8 cm 2 , 9.2 to 29.5 cm 2 , and 7.2 to 58.9 cm 2 , respectively. One hundred seed weight also showed great variation from 2.7 to 39.0 gram. The average leaf area of unifoliate, middle leaflet of first trifoliate and third trifoliate leaf were 15.7 cm 2 , 18.1 cm 2 and 32.7 cm 2 , respectively, and that of seed average weight was 17.2 gram per one hundred seed. Significantly positive correlations were observed between seed weight and leaf area of unifoliate (r=0.80**), first trifoliate (r=0.75**) and third trifoliate (r=0.67**), respectively. Both the leaf length and leaf width of unifoliate, middle leaflet of first trifoliate and third trifoliate leaf were significantly positively correlated with seed weight and both the correlations of unifoliate were higher than the other leaves. The correlations of leaf width in soybean leaflet were higher than those of leaf length. Leaf length/width (L/W) ratio of upper leaf was higher than that of lower leaf in the leaf size. Both the leaf area and leaf width of unifoliate leaf are the most suitable predictive characteristics of early selection in related to seed weight for soybean breeding program.Keywords : soybean, seed weight, leaf size, correlation, breeding
