Journal ArticleDOI
The dangers of using Cq to quantify nucleic acid in biological samples; a lesson from COVID-19.
Daniel Evans,Simon Cowen,Martin Kammel,Denise M. O'Sullivan,Graham R. Stewart,Hans-Peter Grunert,Jacob Moran-Gilad,Jasper Verwilt,Jiwon In,Jo Vandesompele,Kathryn A. Harris,Ki Ho Hong,Nathaniel Storey,Suzie Hingley-Wilson,Ulf Dühring,Young-Kyung Bae,Carole A. Foy,Julian Braybrook,Heinz Zeichhardt,Jim F. Huggett +19 more
TL;DR: In this article, the reproducibility and impact of diagnostic cut-offs on clinical sensitivity of COVID-19 testing were investigated and the inter-laboratory variation for a given Cq value was >1000 fold in copies/mL (99% CI).
Journal ArticleDOI
Single base mutations in the nucleocapsid gene of SARS-CoV-2 affects amplification efficiency of sequence variants and may lead to assay failure
Nathaniel Storey,Julianne R Brown,Rui P A Pereira,Denise M. O'Sullivan,Jim F. Huggett,Rachel Williams,Judith Breuer,Kathryn A. Harris +7 more
TL;DR: In this article, the authors demonstrate how two single nucleotide variants (SNVs) altered the amplification curve of a diagnostic PCR targeting the Nucleocapsid (N) gene and illustrate how threshold setting can lead to false negative results even where the variant sequence is amplified.
