A review on goat sperm cryopreservation

Sperm cells are the endpoint of male spermatogenesis and have particular anatomic and metabolic features. Sperm cryopreservation and storage currently require liquid nitrogen or ultralow refrigeration methods for long or short term storage, which requires routine maintenance and extensive space requirements. Conserving sperms have several purposes such as artificial reproductive technologies (ART), species conservation and clinical medicine. The combinations of storage temperature, cooling rate, chemical composition of the extender, cryoprotectant concentration, reactive oxygen species (ROS), seminal plasma composition and hygienic control are the key factors that affect the life-span of spermatozoa. Sperm preservation protocols vary among animal species owing to their inherent particularities that change extenders used for refrigeration and freezing. Extenders for freezing sperm cells contain buffers, carbohydrates (glucose, lactose, raffinose, saccharose and trehalose), salts (sodium citrate, citric acid), egg yolk and antibiotics. The use of different cryoprotectants, like trehalose or glycerol, as well as different concentrations of egg yolk and other constituents in semen extenders are being studied in our laboratory. Several cooling rates have been tested to freeze sperm cells. The use of faster rates (15-60 degrees C/min) gives rise to best sperm survivals after freezing-thawing, but more studies are needed to find the adequate cooling rates for each animal species. Sheep and goat males of some native breeds are being used in studies performed in EZN. Semen from those males has been frozen and stored as part of the Portuguese Animal Germplasm Bank. In small ruminants, individual variations in the quality of frozen semen have been observed, suggesting specific differences in sperm susceptibility to freezing methods, particularly obvious in goat males. Best quality frozen semen from small ruminants is being used in cervical artificial insemination studies aiming to increase productive parameters in selected flocks.

Reproduction in domestic animals is under control by man and the technologies developed to facilitate that control have a major impact on the efficiency of food production. Reproduction is an energy-intensive process. In beef cattle, for example, over 50 % of the total feed consumption required to produce a unit of meat protein is consumed by the dam of the meat animal (Anim Prod 27:367–379, 1978). Sows are responsible for about 20 % of the total feed needed to produce animals for slaughter (Adv Pork Prod 19:223–237, 2008). Accordingly, energy input to produce food from animal sources is reduced by increasing number of offspring per unit time a breeding female is in the herd. Using beef cattle as an example again, life-cycle efficiency for production of weaned calves is positively related to early age at puberty and short calving intervals (J Anim Sci 57:852–866, 1983). Reproductive technologies also dictate the strategies that can be used to select animals genetically for traits that improve production. Of critical importance has been artificial insemination (AI) (Anim Reprod Sci 62:143–172, 2000; Stud Hist Philos Biol Biomed Sci 38:411–441, 2007; Reprod Domest Anim 43:379–385, 2008; J Dairy Sci 92:5814–5833, 2009) and, as will be outlined in this chapter, emerging technologies offer additional opportunities for improvements in genetic selection. Given the central role of reproduction as a determinant of production efficiency and in genetic selection, improvements in reproductive technologies will be crucial to meeting the challenges created by the anticipated increases in world population (from seven billion people in 2011 to an anticipated nine billion by 2050; World population prospects: the 2010 revision, highlights and advance tables. Artificial insemination of dairy goats in France

Reproductive seasonality observed in all breeds of goats originating from temperate latitudes and in some breeds from subtropical latitudes can now be controlled by artificial changes in photoperiod. Short days stimulate sexual activity, while long days inhibit it. This knowledge has allowed the development of photoperiodic treatments to control sexual activity in goats, for both the buck and doe. In the French intensive milk production system, goat AI plays an important role to control reproduction and, in conjunction with progeny testing, to improve milk production. Most dairy goats are inseminated out of the breeding season with deep frozen semen, after induction of oestrus and ovulation by hormonal treatments. This protocol provides a kidding rate of approximately 65%. New breeding strategies have been developed, based on the buck effect associated with AI, to reduce the use of hormones. With the development of insemination with frozen semen, a classical selection programme was set up, including planned mating, progeny testing and the diffusion of proved sires by inseminations in herds. Functional traits have become important for efficient breeding schemes in the dairy goat industries. Based on knowledge gained over the past decade, the emphasis in selective breeding has been placed on functional traits related to udder morphology and health. New windows have been opened based on new molecular tools, allowing the detection and mapping of genes of economic importance.
