Silver nanoparticles as a new generation of antimicrobials.

Here, we present a review of the antibacterial effects of silver nanomaterials, including proposed antibacterial mechanisms and possible toxicity to higher organisms. For purpose of this review, silver nanomaterials include silver nanoparticles, stabilized silver salts, silver–dendrimer, polymer and metal oxide composites, and silver-impregnated zeolite and activated carbon materials. While there is some evidence that silver nanoparticles can directly damage bacteria cell membranes, silver nanomaterials appear to exert bacteriocidal activity predominantly through release of silver ions followed (individually or in combination) by increased membrane permeability, loss of the proton motive force, inducing de-energization of the cells and efflux of phosphate, leakage of cellular content, and disruption DNA replication. Eukaryotic cells could be similarly impacted by most of these mechanisms and, indeed, a small but growing body of literature supports this concern. Most antimicrobial studies are performed in simple aquatic media or cell culture media without proper characterization of silver nanomaterial stability (aggregation, dissolution, and re-precipitation). Silver nanoparticle stability is governed by particle size, shape, and capping agents as well as solution pH, ionic strength, specific ions and ligands, and organic macromolecules—all of which influence silver nanoparticle stability and bioavailability. Although none of the studies reviewed definitively proved any immediate impacts to human health or the environment by a silver nanomaterial containing product, the entirety of the science reviewed suggests some caution and further research are warranted given the already widespread and rapidly growing use of silver nanomaterials.

A review of the antibacterial effects of silver nanomaterials and potential implications for human health and the environment

Nanoparticles (NPs) are increasingly used to target bacteria as an alternative to antibiotics. Nanotechnology may be particularly advantageous in treating bacterial infections. Examples include the utilization of NPs in antibacterial coatings for implantable devices and medicinal materials to prevent infection and promote wound healing, in antibiotic delivery systems to treat disease, in bacterial detection systems to generate microbial diagnostics, and in antibacterial vaccines to control bacterial infections. The antibacterial mechanisms of NPs are poorly understood, but the currently accepted mechanisms include oxidative stress induction, metal ion release, and non-oxidative mechanisms. The multiple simultaneous mechanisms of action against microbes would require multiple simultaneous gene mutations in the same bacterial cell for antibacterial resistance to develop; therefore, it is difficult for bacterial cells to become resistant to NPs. In this review, we discuss the antibacterial mechanisms of NPs against bacteria and the factors that are involved. The limitations of current research are also discussed.

Silver nanoparticles are nanoparticles of silver which are in the range of 1 and 100 nm in size. Silver nanoparticles have unique properties which help in molecular diagnostics, in therapies, as well as in devices that are used in several medical procedures. The major methods used for silver nanoparticle synthesis are the physical and chemical methods. The problem with the chemical and physical methods is that the synthesis is expensive and can also have toxic substances absorbed onto them. To overcome this, the biological method provides a feasible alternative. The major biological systems involved in this are bacteria, fungi, and plant extracts. The major applications of silver nanoparticles in the medical field include diagnostic applications and therapeutic applications. In most of the therapeutic applications, it is the antimicrobial property that is being majorly explored, though the anti-inflammatory property has its fair share of applications. Though silver nanoparticles are rampantly used in many medical procedures and devices as well as in various biological fields, they have their drawbacks due to nanotoxicity. This review provides a comprehensive view on the mechanism of action, production, applications in the medical field, and the health and environmental concerns that are allegedly caused due to these nanoparticles. The focus is on effective and efficient synthesis of silver nanoparticles while exploring their various prospective applications besides trying to understand the current scenario in the debates on the toxicity concerns these nanoparticles pose.

Silver nanoparticles: mechanism of antimicrobial action, synthesis, medical applications, and toxicity effects

In the present study, we report the preparation of silver nanoparticles in the range of 10‐15 nm with increased stability and enhanced anti-bacterial potency. The morphology of the nanoparticles was characterized by transmission electron microscopy. The antibacterial effect of silver nanoparticles used in this study was found to be far more potent than that described in the earlier reports. This effect was dose dependent and was more pronounced against gram-negative bacteria than gram-positive organisms. Although bacterial cell lysis could be one of the reasons for the observed antibacterial property, nanoparticles also modulated the phosphotyrosine profile of putative bacterial peptides, which could thus affect bacterial signal transduction and inhibit the growth of the organisms.

Characterization of enhanced antibacterial effects of novel silver nanoparticles

A precise control of morphology, which plays a significant role towards the application of materials, can be achieved by studying the effects of macromolecules as nucleation templates for minerals in a biomineralization process. The present investigation aims to understand such effects on medically important materials. Thus, Silver (Ag) and Calcium hydroxyapatite [Ca10(PO4)6(OH)2] were synthesized in both biopolymer (gelatin) and synthetic polymer (PVA) media, with aqueous medium as the control to portray a collage of how the interface between the macromolecule and the mineral controls the final morphology of the materials. It was also observed that the change of morphology of the products does influence their performances in specific applications like antibacterial property of the nanoparticles of silver was found to the best when synthesized in aqueous media. Thus, we consider that the organic–inorganic interaction to be of vital significance in the synthesis of complex shapes and sizes of nanoparticles for important applications.

Morphological changes in biomimetically synthesized hydroxyapatite and silver nanoparticles for medical applications

There is an increasing demand for an affordable and easy-to-fabricate material to help patients having a long bone gap. In this paper, we describe the biomimetic synthesis of Hap-Gel in situ nanocomposite powders with varied proportions. Their biocompatibility and bone regeneration abilities were assessed on a rabbit model. The use of Hap crystals and Gel molecule, the soluble form of bone protein, makes the nanocomposites comparable to natural bone in constituents. The application of biomimetic principles improves crystal morphology and the interaction of Hap crystals with the Gel molecules as seen through in vitro characterizations. Out of the various compositions studied, one with 80:20 proportions of Hap to Gel proved to be closest to the characteristics of natural bone. The immunological response to this composite, assessed through intradermal inoculation, did not reveal any reaction. The in vivo implantation studies in the femoral condyle of the animals, as assessed by serial post-operative follow-up radiography and the histological evaluation, revealed a good biocompatibility and bone-regeneration ability of the material. Thus, nanocomposites of Hap-Gel have a great potential for serving as an effective and affordable biomaterial for bone grafting applications.
