The biological activity of selenium is dependent upon its speciation We aim to integrate selenium speciation and metabolism into a discussion of the mechanisms by which selenium exerts its biological activity First, we present the current status of selenium in the prevention of cancer, cardiovascular and neurodegenerative diseases with particular attention paid to the results of major chemoprevention trials involving selenium supplementation A comprehensive review of the current understanding of the metabolism of common dietary selenium compounds – selenite, selenomethionine, methylselenocysteine and selenocystine – is presented, with discussion of the evidence for the various metabolic pathways and their products The antioxidant, prooxidant and other mechanisms of the dietary selenium compounds have been linked to their disease prevention and treatment properties The evidence for these various mechanisms – in vitro, in cells and in vivo – is evaluated with emphasis on the selenium metabolites involved We conclude that dietary selenium compounds should be considered prodrugs, whose biological activity will depend on the activity of the various metabolic pathways in, and the redox status of, cells and tissues These factors should be considered in future laboratory research and in selecting selenium compounds for trials of disease prevention and treatment by selenium supplementation

Which form is that? The importance of selenium speciation and metabolism in the prevention and treatment of disease

Traditional supplements of selenium generally have a low degree of absorption and increased toxicity. Therefore, it is imperative to develop innovative systems as transporters of selenium compounds, which would raise the bioavailability of this element and allow its controlled release in the organism. Nanoscale selenium has attracted a great interest as a food additive especially in individuals with selenium deficiency, but also as a therapeutic agent without significant side effects in medicine. This review is focused on the incorporation of nanotechnological applications, in particular exploring the possibilities of a more effective way of administration, especially in selenium-deficient organisms. In addition, this review summarizes the survey of knowledge on selenium nanoparticles, their biological effects in the organism, advantages, absorption mechanisms, and nanotechnological applications for peroral administration.

Nano-selenium and its nanomedicine applications: a critical review.

Selenium nanoparticles (SeNPs) are gaining importance in the field of medicine owing to their antibacterial and anticancer properties. SeNPs are biocompatible and non-toxic compared to the counterparts, selenite (SeO3 (-2)) and selenate (SeO4 (-2)). They can be synthesized by physical, chemical, and biological methods and have distinct bright orange-red color. Biogenic SeNPs are stable and do not aggregate owing to natural coating of the biomolecules. Various hypotheses have been proposed to describe the mechanism of microbial synthesis of SeNPs. It is primarily a two-step reduction process from SeO4 (-2) to SeO3 (-2) to insoluble elemental selenium (Se(0)) catalyzed by selenate and selenite reductases. Phenazine-1-carboxylic acid and glutathione are involved in selenite reduction. Se factor A (SefA) and metalloid reductase Rar A present on the surface of SeNPs confer stability to the nanoparticles. SeNPs act as potent chemopreventive and chemotherapeutic agents. Conjugation with antibiotics enhances their anticancer efficacy. These also have applications in nanobiosensors and environmental remediation.

Biogenic selenium nanoparticles: current status and future prospects

Selenium (Se) is an essential element for humans and animals, which occurs ubiquitously in the environment. It is present in trace amounts in both organic and inorganic forms in marine and freshwater systems, soils, biomass and in the atmosphere. Low Se levels in certain terrestrial environments have resulted in Se deficiency in humans, while elevated Se levels in waters and soils can be toxic and result in the death of aquatic wildlife and other animals. Human dietary Se intake is largely governed by Se concentrations in plants, which are controlled by root uptake of Se as a function of soil Se concentrations, speciation and bioavailability. In addition, plants and microorganisms can biomethylate Se, which can result in a loss of Se to the atmosphere. The mobilization of Se across soil-plant-atmosphere interfaces is thus of crucial importance for human Se status. This review gives an overview of current knowledge on Se cycling with a specific focus on soil-plant-atmosphere interfaces. Sources, speciation and mobility of Se in soils and plants will be discussed as well as Se hyperaccumulation by plants, biofortification and biomethylation. Future research on Se cycling in the environment is essential to minimize the adverse health effects associated with unsafe environmental Se levels.

Selenium Cycling Across Soil-Plant-Atmosphere Interfaces: A Critical Review

A bioreductive capacity of a plant, Terminalia arjuna leaf extract, was utilized for preparation of selenium nanoparticles. The leaf extract worked as good capping as well as stabilizing agent and facilitated the formation of stable colloidal nanoparticles. Resulting nanoparticles were characterized using UV-Vis spectrophotometer, transmission electron microscopy (TEM), energy dispersive X-ray analysis (EDAX), Fourier transform infrared spectroscopy (FT-IR), and X-ray diffraction analysis (XRD), respectively. The colloidal solution showed the absorption maximum at 390 nm while TEM and selected area electron diffraction (SAED) indicated the formation of polydispersed, crystalline selenium nanoparticles of size raging from 10 to 80 nm. FT-IR analysis suggested the involvement of O-H, N-H, C=O, and C-O functional group of the leaf extract in particle formation while EDAX analysis indicated the presence of selenium in synthesized nanoparticles. The effect of nanoparticles on human lymphocytes treated with arsenite, As(III), has been studied. Studies on cell viability using MTT assay and DNA damage using comet assay revealed that synthesized selenium nanoparticles showed protective effect against As(III)-induced cell death and DNA damage. Chronic ingestion of arsenic infested groundwater, and prevalence of arsenicosis is a serious public health issue. The synthesized benign nanoselenium can be a promising agent to check the chronic toxicity caused due to arsenic exposure.

Biogenic Synthesis of Selenium Nanoparticles and Their Effect on As(III)-Induced Toxicity on Human Lymphocytes

Sixteen groups of mice were fed diets containing different selenium species to compare their toxicity. Inorganic sodium selenate and sodium hydroselenite, elementary nanoSe, organic Sel-Plex, and Lacto-MicroSelenium were administered for 14 d at concentrations of 0.5, 5, and 50 ppm Se, equivalent to 0.5, 5, and 50 mg Se/kg food, corresponding to an estimated 4, 40, and 400 µg/kg body weight/d Se uptake, respectively. At the end of the treatment, body, liver, spleen, kidney, heart, and brain weights were measured, mice were subjected to necropsy, and histological examinations were performed on the liver. At lower Se doses (0.5 and 5 ppm) a moderate reduction was observed in the number of bone marrow and white blood cells and in granulocyte-macrophage colony-forming units (GM-CFUs) relative to the untreated control group of mice. A comparison of lowest toxic doses of sodium selenite in mice (0.5 ppm) and mallard (10 ppm) indicates that birds are more resistant to Se than rodents. In mice, a small but measurable weight loss was observed after 5 ppm selenate and LactoMicroSe treatment. The most significant changes took place after 50-ppm administration in body and spleen weight, hematology, and liver histology. Toxicity was more pronounced when inorganic Se was applied than after subacute application of Sel-Plex, nanoSe, or LactoMicroSe. To summarize the effects, the authors' 14-d murine subacute toxicity study showed that the toxicity of Se species decreased in the following order: selenate > selenite > nanoSe > Sel-Plex > LactoMicroSe.

Subacute toxicity of nano‐selenium compared to other selenium species in mice

Selenium is well known as an essential trace element since the 20th century, but it can be overdosed easily because of its toxicity. According to the present regulations [1] only the potentially most harmful, inorganic selenium salts can be added to any comestibles. Elemental selenium is considered as the least toxical of all selenium forms and in the same time supplementation with its nano-size particles has the same or better bioavailability compared to its salts. In our experiments we managed to produce nano-size (100-500nm) elemental selenium by using probiotic yogurt bacteria in a fermentation procedure. We developed the laboratory technology of product purification and recovery from bacteria.

Nowadays, the world is facing the problem of environmental pollution because of the increase of man’s needs requires development in life activities, progress industrialization, transportation tools, enhancement of agriculture and exploitation of natural resources. Soil and water resources are extremely exposed to pollution from different aspects. Agrochemicals in particular, have created severe problems, since they release thousands of chemicals to the environment. Several studies on the effect of environmental pollutants on agroecosystem have been carried out. On the other hand, the importance of trace elements as environmental pollutants is well known and well documented in literature. Cu contamination to agricultural soils has been accelerated due to its wide and repeated use in agriculture and horticulture as fertilizers or fungicides to protect vines, citrus trees, and other fruit crops against fungus diseases. Applied Cu from different agrochemical sources to agroenvironment may be adsorbed and are transported to the groundwater table and pollute it besides polluting the soils. The use of Cu-based fungicides in vineyard soils is widely documented worldwide. It has been found that many countries contain concentrations in excess of 100 mg kg−1. Importance of study of transport of Cu arises due to the fact that Cu is absorbed in soils and also reaches the groundwater table, thus polluting both soil and ground water. It is often more important to be able to estimate the mobile fraction, the readily soluble fraction, the exchangeable fraction, or the plant available fraction of Cu content of a soil as a more direct indication of the likelihood of deleterious or toxic effects on soils and groundwater. Therefore, the aim of present work was to highlight the behavior and ecotoxicological effects of copper on horticultural soils.
