CD33 modulates TREM2: convergence of Alzheimer loci
Gail Chan,Charles C. White,Phoebe A. Winn,Maria Cimpean,Joseph M. Replogle,Laura R. Glick,Nicole Cuerdon,Katie J. Ryan,Keith A. Johnson,Keith A. Johnson,Julie A. Schneider,David A. Bennett,Lori B. Chibnik,Reisa A. Sperling,Reisa A. Sperling,Elizabeth M. Bradshaw,Philip L. De Jager +16 more
TL;DR: A protein quantitative trait analysis in monocytes from 226 individuals to evaluate cross-talk between Alzheimer loci found the NME8 locus influenced PTK2B and the CD33 risk allele led to greater TREM2 expression.
Journal ArticleDOI
A human microglia-like cellular model for assessing the effects of neurodegenerative disease gene variants.
Katie J. Ryan,Charles C. White,Charles C. White,Kruti R. Patel,Jishu Xu,Jishu Xu,Marta Olah,Marta Olah,Joseph M. Replogle,Michael Frangieh,Michael Frangieh,Maria Cimpean,Maria Cimpean,Phoebe A. Winn,Phoebe A. Winn,Allison McHenry,Allison McHenry,Belinda J. Kaskow,Gail Chan,Nicole Cuerdon,David A. Bennett,Justin D. Boyd,Justin D. Boyd,Jaime Imitola,Wassim Elyaman,Wassim Elyaman,Philip L. De Jager,Philip L. De Jager,Elizabeth M. Bradshaw,Elizabeth M. Bradshaw +29 more
TL;DR: In the PILRB and LRRK2 loci, the cis-eQTL was found in the MDMi cells but not in human peripheral blood monocytes, suggesting that differentiation of monocytes into microglia-like cells led to the acquisition of a cellular state that could reveal the functional consequences of certain genetic variants.
Journal ArticleDOI
A TREM1 variant alters the accumulation of Alzheimer-related amyloid pathology.
Joseph M. Replogle,Gail Chan,Charles C. White,Charles C. White,Towfique Raj,Phoebe A. Winn,Phoebe A. Winn,Denis A. Evans,Reisa A. Sperling,Reisa A. Sperling,Lori B. Chibnik,Elizabeth M. Bradshaw,Julie A. Schneider,David A. Bennett,Philip L. De Jager +14 more
TL;DR: A targeted analysis of the TREM locus is pursued in relation to cognitive decline and pathological features of AD.
Journal ArticleDOI
Genes and Environment in Multiple Sclerosis project: A platform to investigate multiple sclerosis risk
Zongqi Xia,Zongqi Xia,Zongqi Xia,Charles C. White,Charles C. White,Emily K. Owen,Alina Von Korff,Sarah R. Clarkson,Cristin McCabe,Maria Cimpean,Phoebe A. Winn,Ashley Hoesing,Sonya Steele,Irene Cortese,Tanuja Chitnis,Tanuja Chitnis,Howard L. Weiner,Howard L. Weiner,Daniel S. Reich,Lori B. Chibnik,Lori B. Chibnik,Lori B. Chibnik,Philip L. De Jager,Philip L. De Jager,Philip L. De Jager +24 more
TL;DR: Using an integrated genetic and environmental risk score, subjects with twice the MS risk when compared to the average family member are identified, and an initial incidence rate in these subjects is reported that is 30 times greater than that of sporadic MS.
Journal ArticleDOI
Trans-pQTL study identifies immune crosstalk between Parkinson and Alzheimer loci
Gail Chan,Gail Chan,Charles C. White,Charles C. White,Phoebe A. Winn,Maria Cimpean,Joseph M. Replogle,Joseph M. Replogle,Laura R. Glick,Laura R. Glick,Nicole Cuerdon,Nicole Cuerdon,Katie J. Ryan,Katie J. Ryan,Keith A. Johnson,Keith A. Johnson,Julie A. Schneider,David A. Bennett,Lori B. Chibnik,Lori B. Chibnik,Reisa A. Sperling,Reisa A. Sperling,Philip L. De Jager,Philip L. De Jager,Elizabeth M. Bradshaw,Elizabeth M. Bradshaw +25 more
TL;DR: It is found that the rs12456492 PD risk variant affects expression of AD-associated protein CD33 in peripheral monocytes, which suggests that genetic factors for these 2 diseases may converge to influence overlapping innate immune-mediated mechanisms that contribute to neurodegeneration.
