SUMMARY
Many secondary metabolites found in plants have a role in defence against herbivores, pests and pathogens. In this review, a few examples are described and discussed, and some of the problems in determining the precise role(s) of such metabolites highlighted. The role of secondary metabolites in defence may involve deterrence/anti-feedant activity, toxicity or acting as precursors to physical defence systems. Many specialist herbivores and pathogens do not merely circumvent the deterrent or toxic effects of secondary metabolites but actually utilize these compounds as either host recognition cues or nutrients (or both). This is true of both cyanogenic glucosides and glucosinolates, which art discussed in detail as examples of defensive compounds. Their biochemistry is compared and contrasted. An enormous variety of secondary metabolites are derived from shikimic acid or aromatic amino acids, many of which have important roles in defence mechanisms. Several classes of secondary products are ‘induced’ by infection, wounding or herbivory, and examples of these are given. Genetic variation in the speed and extent of such induction may account, at least in part, for the difference between resistant and susceptible varieties. Both salicylates and jasmonates have been implicated as signals in such responses and in many other physiological processes, though their prescise roles and interactions in signalling and development are not fully understood.

Secondary metabolites in plant defence mechanisms

Plants have been interacting with insects for several hundred million years, leading to complex defense approaches against various insect feeding strategies. Some defenses are constitutive while others are induced, although the insecticidal defense compound or protein classes are often similar. Insect herbivory induce several internal signals from the wounded tissues, including calcium ion fluxes, phosphorylation cascades and systemic- and jasmonate signaling. These are perceived in undamaged tissues, which thereafter reinforce their defense by producing different, mostly low molecular weight, defense compounds. These bioactive specialized plant defense compounds may repel or intoxicate insects, while defense proteins often interfere with their digestion. Volatiles are released upon herbivory to repel herbivores, attract predators or for communication between leaves or plants, and to induce defense responses. Plants also apply morphological features like waxes, trichomes and latices to make the feeding more difficult for the insects. Extrafloral nectar, food bodies and nesting or refuge sites are produced to accommodate and feed the predators of the herbivores. Meanwhile, herbivorous insects have adapted to resist plant defenses, and in some cases even sequester the compounds and reuse them in their own defense. Both plant defense and insect adaptation involve metabolic costs, so most plant-insect interactions reach a stand-off, where both host and herbivore survive although their development is suboptimal.

Plant Defense against Insect Herbivores

The essential role of ruminal microflora in ruminant nutrition provides the potential for improvement in animal production via altering the numbers or activities of specific classes of microorganisms. Successful alterations will be facilitated by an understanding of the microbial ecology of the rumen based on its mechanistic underpinnings. Demonstrated improvements in ruminal fermentation can be traced to their consonance with well-established principles of microbial ecology (niche occupancy, selective pressure, adaptation, and interactions) and the thermodynamics and kinetics of substrate utilization. Application of these principles to several proposed alterations of the ruminal bacterial population allows a prediction of their relative feasibility. Improving fiber digestion, decreasing protein degradation, and detoxifying feed components that are present in low concentrations will be difficult to achieve in the rumen and are best approached by altering the feed, either genetically or with postharvest treatment. By contrast, the detoxification of feed components present in high concentration, and redirection of electron disposal away from methanogenesis, are more productive targets for microbiological research.

Chemical constraints to the use of tropical legumes in animal nutrition

Contents of phenolic compounds in leaf extracts often serve as a measure of plant anti-herbivore defence. This method suffers from the multifunctionality of phenolics and from problems with their colorimetric quantification. Here we present further evidence for the pertinence of these problems. Contents of condensed tannins (CCT) were spectrophotometrically quantified in leaf extracts of 11 closely related mimosoid species, and Spodoptera littoralis caterpillars were reared on artificial diet containing these extracts. The relationship of CCT with caterpillar growth differed considerably among plant species, since both positive and negative correlations were detected. There was, however, a negative correlation of CCT with fungal spore germination, indicating a role of these compounds in resistance to fungi. Detailed knowledge on the structure and biological function of defensive compounds and on the overall composition of leaves is required to estimate a plant's defensive efficacy against a particular group of enemies.

Extraction and quantification of "condensed tannins" as a measure of plant anti-herbivore defence? Revisiting an old problem.

The effect on live-weight gain from dosing cattle with rumen bacteria capable of degrading 3-hydroxy-4(l H)-pyridone (DHP) was measured on cattle grazing leucaena and native grass pastures in south-east Queensland. Dosing increased the growth rate of cattle grazing only leucaena pasture: from 0·52 kg/head per day when not dosed, to 1·03 kg/head per day when dosed, in the period 6·19 weeks after treatment. Dosing did not affect the growth rate of cattle either grazing leucaena with native pasture, or grazing only native pasture. The introduced bacteria spread naturally to untreated cattle after 19 weeks post-dosing.The response to dosing occurred when untreated cattle grazing only leucaena pasture had high urinary concentrations of DHP (maximum 0·28%) and low concentrations of serum thyroxine (< 30 nmol/1). The results show that DHP-induced depressions in growth rate may occur in this environment when cattle graze mainly on leucaena-based pasture. Dosing with DHP-degrading bacteria will overcome this problem.

SUMMARY The C3 and C4 plant components in extrusa samples from oesophageally fistulated cattle can be estimated by δ13Canalysis. However, contamination with saliva containing bicarbonates could cause bias in the estimation of the C3 or C4 components. Saliva from animals fed C4 and C3 diets had δ13C values of – 4.37/103 and –15.97/103, respectively. Both values showed a large positive shift of c. 10 units relative to the diets, but the difference between feeds was maintained in the saliva. On theoretical grounds, contamination of the ingested feed with saliva should have caused a small (1·3–3·4% legume) negative bias when estimating legume percentage in the diet. This did not occur when saliva of known (PC was added to mixtures of C3 and C4 forages which were dried, ground and analysed for δ13C. It is postulated that CO2 was lost from bicarbonate when saliva was dried in contact with fresh plant material, and so no significant bias was detected. It would appear that the δ13C technique can be used to estimate the proportion of C3 and C4 components in oesophageal extrusa samples without significant bias from salivary contamination.
