The effect of exogenously applied natural [indole-3-acetic acid (IAA), phenylacetic acid (PAA), indole-3-butyric acid (IBA)] and synthetic [1-naphthaleneacetic acid (NAA)] auxins on the growth and metabolism of green microalga Chlorella vulgaris was examined. Exogenous auxins acted in a concentration-dependent manner on algal growth. Phytohormones at concentration of 100 μM inhibited algal growth expressed as the number of cells. IAA and IBA displayed the highest biological activity at 0.1 μM, whereas PAA and NAA were characterized by the greatest stimulatory effect on the number of cells at 1 μM. Treatment with IAA and IBA at 0.1 μM or NAA and PAA at 1 μM increased the concentration of photosynthetic pigments, monosaccharides and soluble proteins in C.vulgaris. Moreover, all auxins stimulated enzymatic (ascorbate peroxidase, catalase, superoxide dismutase) and non-enzymatic antioxidant (ascorbate, glutathione) systems in C.vulgaris, and therefore, suppressed lipid peroxidation and hydrogen peroxide accumulation. The data supports the hypothesis that auxins play a central role in the regulation of C.vulgaris growth and metabolism and the components of cellular redox systems that are thought to have a prominent role in the regulation of auxin-dependent processes.

The effect of natural and synthetic auxins on the growth, metabolite content and antioxidant response of green alga Chlorella vulgaris (Trebouxiophyceae)

Cytokinins modulate a number of important developmental processes, including the last phase of leaf development, known as senescence, which is associated with chlorophyll breakdown, photosynthetic apparatus disintegration and oxidative damage. There is ample evidence that cytokinins can slow down all these senescence-accompanying changes. Here, we review relationships between the various mechanisms of action of these regulatory molecules. We highlight their connection to photosynthesis, the pivotal process that generates assimilates, however may also lead to oxidative damage. Thus, we also focus on cytokinin induction of protective responses against oxidative damage. Activation of antioxidative enzymes in senescing tissues is described as well as changes in the levels of naturally occurring antioxidative compounds, such as phenolic acids and flavonoids, in plant explants. The main goal of this review is to show how the biological activities of cytokinins may be related to their chemical structure. New links between molecular aspects of natural cytokinins and their synthetic derivatives with antisenescent properties are described. Structural motifs in cytokinin molecules that may explain why these molecules play such a significant regulatory role are outlined.

Role of Cytokinins in Senescence, Antioxidant Defence and Photosynthesis

Water stress reduces endogenous cytokinin (CK) content and may inhibit CK production. Maintenance of endogenous CK levels by genetic transformation with ipt in leaves and roots undergoing senescence may promote stress tolerance. This study was designed to determine the physiological effects of ipt expression on immature and mature leaves and in roots for plants exposed to different levels of water stress for creeping bentgrass (Agrostis stolonifera). Plants containing the ipt gene, encoding the enzyme adenine isopentenyl phosphotransferase for CK synthesis ligated to a senescence-activated promoter (SAG12), and wild-type ‘Penncross’ (WT) were grown hydroponically in a growth chamber and exposed to water stress by weekly additions of polyethylene glycol 8000 to reduce the growing solution osmotic potential from –0.05 to –0.3, –0.5, –0.7, –1.0, and –1.4 MPa. Immature and mature leaves and roots of SAG12-ipt creeping bentgrass were evaluated for ipt expression, CK content, leaf relative water content (RWC), chlorophyll content (Chl), photochemical efficiency (FvFm), osmotic adjustment (OA), photosynthesis rate (Pn), stomatal conductance (gs), transpiration (E), water use efficiency (WUE), carbon isotope discrimination (D), and root viability. Expression of ipt was detected in all plant parts and a higher CK content, primarily in the form of isopentyladenine (iPa), was found in SAG12-ipt plants but not in the WT plants under water stress. Immature leaves exhibited higher iPa and OA at all treatment levels. Mature leaves of SAG12-ipt plants maintained higher OA, Pn, Chl, WUE, and D, whereas gs and E were relatively unaffected compared to the WT. Roots of SAG12-ipt plants had higher levels of iPa and greater root viability than the WT. The results demonstrate that expression of ipt enhanced the tolerance of creeping bentgrass to water stress, which could be attributed to the positive effects on osmotic adjustment, efficient water use, and maintaining higher photosynthetic rate primarily for mature leaves, as well as increased root viability.

Photosynthesis, water use, and root viability under water stress as affected by expression of SAG12-ipt controlling cytokinin synthesis in Agrostis stolonifera

Drought stress limits root growth and inhibits cytokinin (CK) production. Increases in CK production through overexpression of isopentenyltransferase (ipt) alleviate drought damages to promote root growth. The objective of this study was to investigate whether CK-regulated root growth was involved in the alteration of reactive oxygen species (ROS) production and ROS scavenging capacity under drought stress. Wild-type (WT) creeping bentgrass (Agrostis stolonifera L. ‘Penncross’) and a transgenic line (S41) overexpressing ipt ligated to a senescence-activated promoter (SAG12) were exposed to drought stress for 21 d in growth chambers. SAG12-ipt transgenic S41 developed a more extensive root system under drought stress compared to the WT. Root physiological analysis (electrolyte leakage and lipid peroxidation) showed that S41 roots exhibited less cellular damage compared to the WT under drought stress. Roots of SAG12-ipt transgenic S41 had significantly higher endogenous CK content than the WT roots under drought stress. ROS (hydrogen peroxide and superoxide) content was significantly lower and content of total and free ascorbate was significantly higher in S41 roots compared to the WT roots under drought stress. Enzymatic assays and transcript abundance analysis showed that superoxide dismutase, catalase, peroxidase, and dehydroascorbate reductase were significantly higher in S41 roots compared to the WT roots under drought stress. S41 roots also maintained significantly higher alternative respiration rates compared to the WT under drought stress. The improved root growth of transgenic creeping bentgrass may be facilitated by CK-enhanced ROS scavenging through antioxidant accumulation and activation of antioxidant enzymes, as well as higher alternative respiration rates when soil water is limited.

Enhancing cytokinin synthesis by overexpressing ipt alleviated drought inhibition of root growth through activating ROS-scavenging systems in Agrostis stolonifera

Cytokinins are plant hormones that regulate diverse processes in plant development and responses to biotic and abiotic stresses. In this study, we show that Arabidopsis (Arabidopsis thaliana) plants with a reduced cytokinin status (i.e. cytokinin receptor mutants and transgenic cytokinin-deficient plants) are more susceptible to light stress compared with wild-type plants. This was reflected by a stronger photoinhibition after 24 h of high light (approximately 1,000 µmol m−2 s−1), as shown by the decline in maximum quantum efficiency of photosystem II photochemistry. Photosystem II, especially the D1 protein, is highly sensitive to the detrimental impact of light. Therefore, photoinhibition is always observed when the rate of photodamage exceeds the rate of D1 repair. We demonstrate that in plants with a reduced cytokinin status, the D1 protein level was strongly decreased upon light stress. Inhibition of the D1 repair cycle by lincomycin treatment indicated that these plants experience stronger photodamage. The efficiency of photoprotective mechanisms, such as nonenzymatic and enzymatic scavenging systems, was decreased in plants with a reduced cytokinin status, which could be a cause for the increased photodamage and subsequent D1 degradation. Additionally, slow and incomplete recovery in these plants after light stress indicated insufficient D1 repair. Mutant analysis revealed that the protective function of cytokinin during light stress depends on the ARABIDOPSIS HISTIDINE KINASE2 (AHK2) and AHK3 receptors and the type B ARABIDOPSIS RESPONSE REGULATOR1 (ARR1) and ARR12. We conclude that proper cytokinin signaling and regulation of specific target genes are necessary to protect leaves efficiently from light stress.
