This review covers advances in electrochemical and biochemical sensor development and usage during 2010 and 2011 In choosing scholarly articles to contribute to this review, special emphasis was placed on work published in the areas of reference electrodes, potentiometric sensors, voltammetric sensors, amperometric sensors, biosensors, immunosensors, and mass sensors In the past two years there have been a number of important papers, that do not fall into the general subsections contained within the larger sections Such novel advances are very important for the field of electrochemical sensors as they open up new avenues and methods for future research Each section above contains a subsection titled “Other Papers of Interest” that includes such articles and describes their importance to the field in general For example, while most electrochemical techniques for sensing analytes of interest are based on the changes in potential or current, Shan et al1 have developed a completely novel method for performing electrochemical measurements In their work, they report a method for imaging local electrochemical current using the optical signal of the electrode surface generated from a surface plasmon resonance (SPR) The electrochemical current image is based on the fact that the current density can be easily calculated from the local SPR signal The authors demonstrated this concept by imaging traces of TNT on a fingerprint on a gold substrate

Full articles and reviews were primarily amassed by searching the SciFinder Scholar and ISI Web of Knowledge Additional articles were found through alternate databases or by perusing analytical journals for pertinent publications Due to the reference limitation, only publications written in English were considered for inclusion Obviously, there have been more published accounts of groundbreaking work with electrochemical and biochemical sensors than those covered here This review is a small sampling of the available literature and not intended to cover every advance of the past two years The literature chosen focuses on new trends in materials, techniques, and clinically relevant applications of novel sensors To ensure proper coverage of these trends, theoretical publications and applications of previously reported sensor development were excluded

Reverse electrowetting as a new approach to high-power energy harvesting

Following the development of microfluidic systems, there has been a high tendency towards developing lab-on-a-chip devices for biochemical applications. A great deal of effort has been devoted to improve and advance these devices with the goal of performing complete sets of biochemical assays on the device and possibly developing portable platforms for point of care applications. Among the different microfluidic systems used for such a purpose, digital microfluidics (DMF) shows high flexibility and capability of performing multiplex and parallel biochemical operations, and hence, has been considered as a suitable candidate for lab-on-a-chip applications. In this review, we discuss the most recent advances in the DMF platforms, and evaluate the feasibility of developing multifunctional packages for performing complete sets of processes of biochemical assays, particularly for point-of-care applications. The progress in the development of DMF systems is reviewed from eight different aspects, including device fabrication, basic fluidic operations, automation, manipulation of biological samples, advanced operations, detection, biological applications, and finally, packaging and portability of the DMF devices. Success in developing the lab-on-a-chip DMF devices will be concluded based on the advances achieved in each of these aspects.

A review of digital microfluidics as portable platforms for lab-on a-chip applications.

Electrowetting-on-dielectric (EWOD) lab-on-a-chip systems have already proven their potential within a broad range of bio-assays. Nevertheless, research on the analytical performance of those systems is limited, yet crucial for a further breakthrough in the diagnostic field. Therefore, this paper presents the intrinsic possibilities of an EWOD lab-on-a-chip as a versatile platform for homogeneous and heterogeneous bio-assays with high analytical performance. Both droplet dispensing and splitting cause variations in droplet size, thereby directly influencing the assay's performance. The extent to which they influence the performance is assessed by a theoretical sensitivity analysis, which allows the definition of a basic framework for the reduction of droplet size variability. Taking advantage of the optimized droplet manipulations, both homogeneous and heterogeneous bio-assays are implemented in the EWOD lab-on-a-chip to demonstrate the analytical capabilities and versatility of the device. A fully on-chip enzymatic assay is realized with high analytical performance. It demonstrates the promising capabilities of an EWOD lab-on-a-chip in food-related and medical applications, such as nutritional and blood analyses. Further, a magnetic bio-assay for IgE detection using superparamagnetic nanoparticles is presented whereby the nanoparticles are used as solid carriers during the bio-assay. Crucial elements are the precise manipulation of the superparamagnetic nanoparticles with respect to dispensing and separation. Although the principle of using nano-carriers is demonstrated for protein detection, it can be easily extended to a broader range of bio-related applications like DNA sensing. In heterogeneous bio-assays the chip surface is actively involved during the execution of the bio-assay. Through immobilization of specific biological compounds like DNA, proteins and cells a reactive chip surface is realized, which enhances the bio-assay performance. To demonstrate this potential, on-chip adhesion islands are fabricated to immobilize MCF-7 human breast cancer cells. Viability studies are performed to assess the functionalization efficiency.

A versatile electrowetting-based digital microfluidic platform for quantitative homogeneous and heterogeneous bio-assays

Experimental investigations of the liquid transfer between two separating plates have been carried out, with the aim of increasing the ink transfer ratio in micro-gravure-offset printing. A sessile droplet is divided into two by moving one plate vertically with respect to a fixed plate. The surface contact angles of the sessile droplet on the two plates were of particular interest in this study because the volume ratio of the two droplets varies with the contact angles. Various methods for controlling the surface contact angle, including chemical treatment, plasma surface modification and electrowetting-on-dielectric were assessed. Image analysis of the droplets was carried out to estimate the surface contact angles and to measure the volumes of the droplets. The liquid transfer process and related phenomena, such as satellite droplet generation and the variation in the contact areas between the droplets and the plates, were observed. The experimental results help us to determine the optimal surface contact angles of the plate cylinder, blanket cylinder and substrate for micro-gravure-offset printing.

Liquid transfer between two separating plates for micro-gravure-offset printing

A low voltage, two-level-metal, and multi-layer insulator electrowetting-on-dielectric (EWD) platform is presented. Dispensing 300pl droplets from 140nl closed on-chip reservoirs was accomplished with as little as 11.4V solely through EWD forces, and the actuation threshold voltage was 7.2V with a 1Hz voltage switching rate between electrodes. EWD devices were fabricated with a multilayer insulator consisting of 135nm sputtered tantalum pentoxide (Ta(2)O(5)) and 180nm parylene C coated with 70nm of CYTOP. Furthermore, the minimum actuation threshold voltage followed a previously published scaling model for the threshold voltage, V(T), which is proportional to (t/e(r))(1/2), where t and e(r) are the insulator thickness and dielectric constant respectively. Device threshold voltages are compared for several insulator thicknesses (200nm, 500nm, and 1µm), different dielectric materials (parylene C and tantalum pentoxide), and homogeneous versus heterogeneous compositions. Additionally, we used a two-level-metal fabrication process, which enables the fabrication of smaller and denser electrodes with high interconnect routing flexibility. We also have achieved low dispensing and actuation voltages for scaled devices with 30pl droplets.
