The influence of diet on the distribution of nitrogen isotopes in animals was investigated by analyzing animals grown in the laboratory on diets of constant nitrogen isotopic composition. 
The isotopic composition of the nitrogen in an animal reflects the nitrogen isotopic composition of its diet. The δ^(15)N values of the whole bodies of animals are usually more positive than those of their diets. Different individuals of a species raised on the same diet can have significantly different δ^(15)N values. The variability of the relationship between the δ^(15)N values of animals and their diets is greater for different species raised on the same diet than for the same species raised on different diets. Different tissues of mice are also enriched in ^(15)N relative to the diet, with the difference between the δ^(15)N values of a tissue and the diet depending on both the kind of tissue and the diet involved. The δ^(15)N values of collagen and chitin, biochemical components that are often preserved in fossil animal remains, are also related to the δ^(15)N value of the diet. 
The dependence of the δ^(15)N values of whole animals and their tissues and biochemical components on the δ^(15)N value of diet indicates that the isotopic composition of animal nitrogen can be used to obtain information about an animal's diet if its potential food sources had different δ^(15)N values. The nitrogen isotopic method of dietary analysis probably can be used to estimate the relative use of legumes vs non-legumes or of aquatic vs terrestrial organisms as food sources for extant and fossil animals. However, the method probably will not be applicable in those modern ecosystems in which the use of chemical fertilizers has influenced the distribution of nitrogen isotopes in food sources. 
The isotopic method of dietary analysis was used to reconstruct changes in the diet of the human population that occupied the Tehuacan Valley of Mexico over a 7000 yr span. Variations in the δ^(15)C and δ^(15)N values of bone collagen suggest that C_4 and/or CAM plants (presumably mostly corn) and legumes (presumably mostly beans) were introduced into the diet much earlier than suggested by conventional archaeological analysis.

Influence of Diet On the Distribtion of Nitrogen Isotopes in Animals

Stable isotope analysis (SIA) has emerged as a common tool in ecology and has proven especially useful in the study of animal diet, habitat use, movement, and physiology. SIA has been vigorously applied to the study of marine mammals, because most species live in habitats or undergo large migrations/movements that make them difficult to observe. Our review supplies a complete list of published SIA contributions to marine mammal science and highlights informative case examples in four general research areas: (1) physiology and fractionation, (2) foraging ecology and habitat use, (3) ecotoxicology, and (4) historic ecology and paleoecology. We also provide a condensed background of isotopic nomenclature, highlight several physiological considerations important for accurate interpretation of isotopic data, and identify research areas ripe for future growth. Because it is impossible to conduct controlled laboratory experiments on most marine mammal species, future studies in marine mammal ecology must draw on isotopic data collected from other organisms and be cognizant of key assumptions often made in the application of SIA to the study of animal ecology. The review is designed to be accessible to all audiences, from students unfamiliar with SIA to those who have utilized it in published studies.

Decarbonation and preservation method for the analysis of organic C and N contents and stable isotope ratios of low-carbonated suspended particulate material

Plans for the restoration of aquatic ecosystems are increasingly focusing on the restoration and rehabilitation of self-sustaining native fish communities. Such efforts have not traditionally adopted an ecosystem-based perspective, which considers species as embedded within a broader food web context. In this study, we quantify food web changes in Lake Tahoe (California-Nevada) over the last century based on stable isotope analysis of museum-archived, preserved fish specimens collected during 4 historical periods and under present conditions. We also examine the contemporary food web of nearby Cascade Lake, which is free from most exotic species and contains a species assemblage resembling that of Lake Tahoe prior to historical species introductions. During the last century, the freshwater shrimp Mysis relicta and lake trout (Salvelinus namaycush) have been introduced and established in Lake Tahoe, and the native top predator, Lahontan cutthroat trout (Oncorhynchus clarki henshawi; hereafter LCT), has been extirpated. Isotope analysis indicates that lake trout now occupy a trophic niche similar to that of historical LCT. Fish production has shifted from benthic to pelagic, corresponding with the eutrophication of Lake Tahoe during recent decades. The current Cascade Lake food web resembles that of the historical Lake Tahoe food web. Our isotope-based food web reconstructions reveal long-term food web changes in Lake Tahoe and can serve as the basis for setting historically relevant restoration targets. Unfortunately, the presence of nonnative species, particularly Mysis and lake trout, have dramatically altered the pelagic food web structure; as such, they are barriers to native fish community restoration. Fish community restoration efforts should focus on adjacent ecosystems, such as Cascade Lake, which have a high likelihood of success because they have not been heavily affected by nonnative introductions.

Historical Food Web Structure and Restoration of Native Aquatic Communities in the Lake Tahoe (California–Nevada) Basin

Trophic ecology has benefitted from the use of stable isotopes for the last three decades. However, during the last 10 years, there has been a growing awareness of the isotopic biases associated with some pre-analytical procedures that can seriously hamper the interpretation of food webs. We have assessed the extent of such biases by: (1) reviewing the literature on the topic, and (2) compiling C and N isotopic values of marine invertebrates reported in the literature with the associated sample preparation protocols. The factors considered were: acid-washing, distilled water rinsing (DWR), sample type (whole individuals or pieces of soft tissues), lipid content, and gut contents. Two-level ANOVA revealed overall large and highly significant effects of acidification for both δ13C values (up to 0.9‰ decrease) and δ15 N values (up to 2.1‰ decrease in whole individual samples, and up to 1.1‰ increase in tissue samples). DWR showed a weak overall effect with δ13C increments of 0.6‰ (for the entire data set) or decrements of 0.7‰ in δ15 N values (for tissue samples). Gut contents showed no overall significant effect, whereas lipid extraction resulted in the greatest biases in both isotopic signatures (δ13C, up to −2.0‰ in whole individuals; δ15N, up to +4.3‰ in tissue samples). The study analyzed separately the effects of the various factors in different taxonomic groups and revealed a very high diversity in the extent and direction of the effects. Maxillopoda, Gastropoda, and Polychaeta were the classes that showed the largest isotopic shifts associated with sample preparation. Guidelines for the standardization of sample preparation protocols for isotopic analysis are proposed both for large and small marine invertebrates. Broadly, these guidelines recommend: (1) avoiding both acid washing and DWR, and (2) performing lipid extraction and gut evacuation in most cases.

Effects of sample preparation on stable isotope ratios of carbon and nitrogen in marine invertebrates: implications for food web studies using stable isotopes.

The effects of freezing, oven-drying, and forma- lin and ethanol preservation on stable isotope signatures of 3 unrelated marine species (fish, octopus and kelp) were stud- ied over a period of 3 mo to investigate temporal changes in δ 13 C and δ 15 N. The effects on stable isotope ratios during the short-term confinement of live littorinid snails were also examined. Freezing and oven-drying did not result in a sig- nificant change in the initial isotope ratio for both carbon and nitrogen over the period of investigation.
