Artificial mummification was practised in Egypt from ∼ 2600 BC until the fourth century AD. Because of the dry Egyptian climate, however, there are also many natural mummies preserved from earlier as well as later times. To elucidate whether this unique source of ancient human remains can be used for molecular genetic analyses, 23 mummies were investigated for DNA content. One 2,400-yr-old mummy of a child was found to contain DNA that could be molecularly cloned in a plasmid vector. I report here that one such clone contains two members of the Alu family of human repetitive DNA sequences, as detected by DNA hybridizations and nucleotide sequencing. These analyses show that substantial pieces of mummy DNA (3.4 kilobases) can be cloned and that the DNA fragments seem to contain little or no modifications introduced postmortem.

Molecular cloning of Ancient Egyptian mummy DNA

Light microscopy, particularly the use of polarized light, has such a high value for the differential diagnosis of dry bones that it can no longer be neglected. Alterations caused intra vitam by disease or other living conditions can clearly be differentiated by this technique from changes due to postmortem reactions (e.g., pseudopathology). As a reliable diagnosis is the basis not only of the study of case reports but also of the etiology and epidemiology of diseases in ancient populations, paleopathologists would be well-advised to employ histological analysis for their research, to avoid false diagnoses. The necessary basis for such research is the knowledge of the general histology, histogenesis, and growth as well as pathophysiology of bone. Some new techniques which facilitate the practical use of microscopic analysis, such as the preparation of thin-ground sections from undecalcified bone samples and nonrehydrated mummified soft tissues, are described. Selected examples of mechanisms of pathological bone changes, particularly the determination of vestiges of diseases in macerated bones by microscopy, are presented. Emphasis is placed on the differential diagnoses of proliferative reactions (e.g., periosteal processes of long bones and the skull). In this context, the importance of meningeal reactions on the endocranial lamina of the skull for morbidity and mortality in ancient populations is demonstrated. Furthermore, porotic hyperostosis of the skull vault and the orbital roof, i.e., the cribra cranii externa and cribra orbitalia, is discussed. Selected examples of the etiology and epidemiology of ancient diseases are presented (e.g., anemia, scurvy, rickets, and meningeal diseases), and ideas on living conditions and their implications for the origin and the spread of disease are given to establish a better understanding of deficiency and infectious diseases in the past.

Paleohistopathology of bone: A new approach to the study of ancient diseases

Light microscopy should be a routine complementary diagnostic tool for osteoarchaeologists, palaeopathologists and forensic anthropologists. However, this well-established and long existing technique is not widely utilized. It is a popular belief that the preparation of the required sections of bone tissue demands a lot of time, expensive instruments, and a knowledge of plastics. To show that this is not entirely the case, a long existing technique for the manual preparation of ground sections was brushed up and tested. Its application is simple, extremely cheap, rapid and reliable and the final product is a beautiful and intact ca. 10–15 micron or less thin section. A few modifications are proposed which further improve the feasibility of the technique, and which also make it very suitable for less well-preserved, inhumed, and even cremated osteoarchaeological/forensic remains. As in the original method, the modifications need only a few extra, but still very basic and cheap products. A detailed step by step description of the procedure is presented. Preservation of DNA in ancient Egyptian mummies

Tissue from Egyptian mummy material is extremely brittle; hence it was handled in perforated glass tubes during processing. The first (softening) fluid consisted of 96% ethyl alcohol, 30 vol; 1% aqueous formalin, 50 vol; 5% aqueous Na2CO3, 20 vol. It was used in a fluid to tissue volume ratio of 100:1 and allowed to act overnight. A special dehydrating sequence: 80% alcohol, 3-6 hr; 8% phenol in 96% alcohol, and absolute alcohol followed by 3 changes of amyl acetate, 6-18 hr each; 3 changes of 1 % celloidin in methyl benzoate, 24 hr each; then through benzene and embedding in paraffin completed the special technic. This allowed regular sectioning and staining to be done successfully.
