Meghalaya is known to receive the most torrential rainfall in the world, but the region suffers from water shortage as soon as the rain recedes, and the dry season starts. Changes in rainfall patterns and distribution can have a profound impact on water availability in a watershed, and therefore, examining spatial and temporal variations in rainfall is essential. However, the long-term rainfall variations in Meghalaya are not well explored. In this study, we take up two important watersheds in Meghalaya, i.e. Umiam and Umtru watersheds, to study the spatial and temporal rainfall variations. Using the gridded rainfall data from the Indian Meteorological Department from 1901 to 2018, we show that annual, winter, pre-monsoon, and monsoon rainfall is decreasing, whereas the post-monsoon rainfall is increasing. We use the innovative trend analysis (ITA) method to identify the trends in low-, medium-, and high-intensity rainfall. We find that low- and medium-intensity rainfall is in decreasing trend while high-intensity rainfall is increasing across annual and seasonal time scales. Lastly, we cross-check the trends detected using the innovative trend analysis method with a widely accepted Mann-Kendall (MK) test. We find that the results obtained by using the two methods generally concur; however, the ITA can detect non-monotonic trends in different rainfall intensities and is more sensitive to hidden patterns than the MK test.

Innovative trend analysis of spatial and temporal rainfall variations in Umiam and Umtru watersheds in Meghalaya, India

Light provides the energy for photosynthesis and determines plant morphogenesis and development. Low light compromises photosynthetic efficiency and leads to crop yield loss. It remains unknown how rice responds to low light stress at a proteomic level. In this study, the quantitative proteomic analysis with isobaric tags for relative and absolute quantitation (iTRAQ) was used and 1221 differentially expressed proteins (DEPs) were identified from wild type rice plants grown in control or low light condition (17% light intensity of control), respectively. Bioinformatic analysis of DEPs indicated low light remarkably affects the abundance of chloroplastic proteins. Specifically, the proteins involved in carbon fixation (Calvin cycle), electron transport, and ATPase complex are severely downregulated under low light. Furthermore, overexpression of the downregulated gene encoding rice β subunit of glyceraldehyde-3-phosphate dehydrogenase (OsGAPB), an enzyme in Calvin cycle, significantly increased the CO2 assimilation rate, chlorophyll content and fresh weight under low light conditions but have no obvious effect on rice growth and development under control light. Our results revealed that low light stress on vegetative stage of rice inhibits photosynthesis possibly by decreasing the photosynthetic proteins and OsGAPB gene is a good candidate for manipulating rice tolerance to low light stress.

Proteomic Analysis of Rice Subjected to Low Light Stress and Overexpression of OsGAPB Increases the Stress Tolerance.

Chlorophyll is the molecular basis for the function of photosystems and is also a promising tool for ecological prediction. However, the large-scale patterns of chlorophyll variation in grasslands remain poorly understood. We performed consistent measurements of chlorophyll a, b, a+b, and the a:b ratio (chlorophyll a/b) for 421 species across northern hemisphere grassland transects, recorded their distributions, variations, and influencing factors, and examined their relationships with leaf nitrogen. The results showed that the distributional ranges were 0.52-28.33 (mean 5.49) mg·g-1 dry weight, 0.15-12.11 (mean 1.83) mg·g-1 dry weight, 0.67-39.29 (mean 7.32) mg·g-1 dry weight, and 1.28-7.84 (mean 3.02) for chlorophyll a, b, a+b, and a/b, respectively. The chlorophyll averages differed among regions (higher in the Loess Plateau and the Mongolian Plateau than in the Tibetan Plateau), grassland types (desert grasslands > meadow > typical grasslands), life forms, life spans, and taxonomies. In the entire northern hemisphere grassland, chlorophyll concentrations and chlorophyll a/b were negatively correlated to photosynthetically active radiation and the soil N:P ratio, and positively correlated to the mean annual temperatures. These results implied that chlorophyll in grasslands was shaped by the layered structure of grasses, distinct plateau environments, and phylogeny. The allocation patterns of leaf nitrogen to chlorophyll differed among regions and grassland types, showing that caution is required if simply relating single leaf N or chlorophyll to productivity separately. These findings enhance our understanding of chlorophyll in natural grasslands on a large scale, as well as providing information for ecological predictive models.

Spatial Variation of Leaf Chlorophyll in Northern Hemisphere Grasslands.

Rain-shelter covering is widely applied during cherry fruit development in subtropical monsoon climates with the aim of decreasing the dropping and cracking of fruit caused by excessive rainfall. Under rain-shelter covering, the characteristics of the leaves and fruit of the cherry plant may adapt to the changes in the microclimate. However, the molecular mechanism underlying such adaptation remains unclear, although clarifying it may be helpful for improving the yield and quality of cherry under rain-shelter covering. To better understand the regulation and adaptive mechanism of cherry under rain-shelter covering, 38,621 and 3584 differentially expressed genes were identified with a combination of Illumina HiSeq and single-molecule real-time sequencing in leaves and fruits, respectively, at three developmental stages. Among these, key genes, such as those encoding photosynthetic-antenna proteins (Lhca and Lhcb) and photosynthetic electron transporters (PsbP, PsbR, PsbY, and PetF), were up-regulated following the application of rain-shelter covering, leading to increased efficiency of light utilization. The mRNA levels of genes involved in carbon fixation, namely, rbcL and rbcS, were clearly increased compared with those under shelter-free conditions, resulting in improved CO2 utilization. Furthermore, the transcription levels of genes involved in chlorophyll (hemA, hemN, and chlH) and carotenoid synthesis (crtB, PDS, crtISO, and lcyB) in the sheltered leaves peaked earlier than those in the unsheltered leaves, thereby promoting organic matter accumulation in leaves. Remarkably, the expression levels of key genes involved in the metabolic pathways of phenylpropanoid (PAL, C4H, and 4CL) and flavonoid (CHS, CHI, F3’H, DFR, and ANS) in the sheltered fruits were also up-regulated earlier than of those in the unsheltered fruits, conducive to an increase in anthocyanin content in the fruits. According to the physiological indicators and transcriptional expression levels of the related genes, the adaptive regulation mechanism of cherry plants was systematically revealed. These findings can help understand the effect of rain-shelter covering on Chinese cherry cultivation in rainy regions.

Comparative transcriptome analysis reveals the molecular regulation underlying the adaptive mechanism of cherry (Cerasus pseudocerasus Lindl.) to shelter covering.

Rice grain yield is drastically reduced under low light especially in kharif (wet) season due to cloudy weather during most part of crop growth. Therefore, 50–60% of yield penalty was observed. To overcome this problem, identification of low light tolerant rice genotypes with a high buffering capacity trait such as photosynthetic rate has to be developed. Sedoheptulose-1,7 bisphosphatase, a light-regulated enzyme, plays pivotal role in the Calvin cycle by regenerating the substrate (RuBP) for RuBisCo and therefore, indirectly regulates the influx of CO2 for this crucial process. We found a potential role of SBPase expression and activity in low light tolerant and susceptible rice genotypes by analyzing its influence on net photosynthetic rate and biomass. We observed a significant relationship of yield with photosynthesis, SBPase expression and activity especially under low light conditions. Two tolerant and two susceptible rice genotypes were used for the present study. Tolerant genotypes exhibited significant but least reduction compared to susceptible genotypes in the expression and activity of SBPase, which was also manifested in its photosynthetic rate and finally in the grain yield under low light. However, susceptible genotypes showed significant reduction in SBPase activity along with photosynthesis and grain yield suggesting that tracking the expression and activity of SBPase could form a simple and reliable method to identify the low light tolerant rice cultivars. The data were analyzed using the Indostat 7.5, Tukey–Kramer method through Microsoft Excel 2019 and PAST4.0 software. The significant association of SBPase activity with the grain yield, net assimilation rate, electron transfer rate, biomass and grain weight were observed under low light stress. These traits should be considered while selecting and breeding for low light tolerant cultivars. Thus, SBPase plays a major role in the low light tolerance mechanism in rice.

Role of sedoheptulose-1,7 bisphosphatase in low light tolerance of rice (Oryza sativa L.)

Low light intensity is a great limitation for grain yield and quality in rice. However, yield is not significantly reduced in low light tolerant rice varieties. The work therefore planned for comparative transcriptome profiling under low light stress to decipher the genes involved and molecular mechanism of low light tolerance in rice. At active tillering stage, 50% low light exposure for 1 day, 3 days and 5 days were given to Swarnaprabha (low light tolerant) and IR8 (low light sensitive) rice varieties. Illumina (HiSeq) platform was used for transcriptome sequencing. A total of 6,652 and 12,042 genes were differentially expressed due to low light intensity in Swarnaprabha and IR8, respectively as compared to control. CAB, LRP, SBPase, MT15, TF PCL1 and Photosystem I & II complex related gene expressions were mostly increased in Swarnaprabha upon longer duration of low light exposure which was not found in IR8 as compared to control. Their expressions were validated by qRT-PCR. Overall study suggested that the maintenance of grain yield in the tolerant variety under low light might be results of accelerated expression of the genes which enable the plant to keep the photosynthetic processes moving at the same pace even under low light.
