Singlet oxygen, O2(a1Δg), the lowest excited electronic state of molecular oxygen, has been known to the scientific community for ∼80 years. It has a characteristic chemistry that sets it apart from the triplet ground state of molecular oxygen, O2(X3Σ−g), and is important in fields that range from atmospheric chemistry and materials science to biology and medicine. For such a “mature citizen”, singlet oxygen nevertheless remains at the cutting-edge of modern science. In this critical review, recent work on singlet oxygen is summarized, focusing primarily on systems that involve light. It is clear that there is indeed still something new under the sun (243 references).

Nanoparticles as vehicles for delivery of photodynamic therapy agents

We report energy-transferring organically modified silica nanoparticles for two-photon photodynamic therapy. These nanoparticles co-encapsulate two-photon fluorescent dye nanoaggregates as an energy up-converting donor and a photosensitizing PDT drug as an acceptor. They combine two features:  (i) aggregation-enhanced two-photon absorption and emission properties of a novel two-photon dye and (ii) nanoscopic fluorescence resonance energy transfer between this nanoaggregate and a photosensitizer, 2-devinyl-2-(1-hexyloxyethyl)pyropheophorbide. Stable aqueous dispersions of the co-encapsulating nanoparticles (diameter ≤ 30 nm) have been prepared in the nonpolar interior of micelles by coprecipitating an organically modified silica sol with the photosensitizer and an excess amount of the two-photon dye which forms fluorescent aggregates by phase separation from the particle matrix. Organically modified silica nanoparticles co-encapsulating photosensitizing drug and aggregation-enhanced two-photon absorbing fluorescent dye aggregates for two-photon photodynamic therapy.

Functional organic materials with enhanced two-photon absorption lead to new technologies in the fields of chemistry, biology, and photonics. In this article we review experimental and theoretical methodologies allowing detailed investigation and analysis of two-photon absorption properties of organic chromophores. This includes femtosecond two-photon excited fluorescence experimental setups and quantum-chemical methodologies based on time-dependent density functional theory. We thoroughly analyze physical phenomena and trends leading to large twophoton absorption responses of a few series of model chromophores focusing on the effects of symmetric and asymmetric donor/acceptor substitution and branching.

Enhanced two-photon absorption of organic chromophores: theoretical and experimental assessments ∗∗

This critical review describes how halocarbons can function as effective binding sites of anions via halogen bonding, the noncovalent interaction whereby halogen atoms accept electron density. The focus is on the binding and coordination of oxyanions, by far the most numerous class of anions in organic chemistry. It is shown how a large variety of inorganic and organic oxyanions can form discrete adducts and 1D, 2D, or 3D supramolecular networks with chloro-, bromo-, and iodocarbons. Specific examples are discussed in order to identify new supramolecular synthons based on halogen bonding and to outline some general principles for the design of effective and selective receptors based on this interaction. The interaction allows for several other anions to self-assemble with halocarbons and mention is also given to the halogen bonding-based coordination of halides, polycyano- and polyoxometallates (72 references).

Halogen bonding: a general route in anion recognition and coordination

Singlet molecular oxygen (a1Δg) has been produced and optically monitored in time-resolved experiments upon nonlinear two-photon excitation of photosensitizers that contain triple bonds as an integral part of the chromophore. Both experiments and ab initio computations indicate that the photophysical properties of alkyne-containing sensitizers are similar to those in the alkene-containing analogues. Most importantly, however, in comparison to the analogue that contains double bonds, the sensitizer containing alkyne moieties is more stable against singlet-oxygen-mediated photooxygenation reactions. This increased stability can be advantageous, particularly with respect to two-photon singlet oxygen imaging experiments in which data are collected over comparatively long time periods.

Two-photon photosensitized production of singlet oxygen: sensitizers with phenylene-ethynylene-based chromophores.

The synthesis and characterization of water-soluble singlet oxygen sensitizers with a phenylene−vinylene motif is presented. The principal motivation for this study was to better understand specific features of a water-soluble molecule that influence the photosensitized production of singlet oxygen upon nonlinear, two-photon excitation of that molecule. To achieve water solubility, sensitizers were synthesized with ionic as well as nonionic substituents. In the ionic approach, salts of N-methylated pyridine, benzothiazole, and 1-methyl-piperazine moieties were used, as were aryl-substituted sulfonic acid moieties. In the nonionic approach, aryl-substituted triethylene glycol moieties were used. Selected photophysical properties of the compounds synthesized were determined, including singlet oxygen quantum yields.
