Citric acid is a commodity chemical produced and consumed throughout The World. It is used mainly in the food and beverage industry, primarily as an acidulant. Although it is one of the oldest industrial fermentations, its World production is still in rapid increasing. Global production of citric acid in 2007 was over 1.6 million tones. Biochemistry of citric acid fermentation, various microbial strains, as well as various substrates, technological processes and product recovery are presented. World production and economics aspects of this strategically product of bulk biotechnology are discussed.

Gel entrapment and micro-encapsulation: methods, applications and engineering principles

On the merits of viable-cell immobilisation.

Immobilized cells of Aspergillus niger needed a lower initial sucrose concentration than free cells in order to obtain maximal yields of citric acid production. High sucrose concentrations led to reduced yields and increased polyol formation (glycerol, erythritol, arabitol). Continuous fermentation with media containing low sugar concentrations prevented the formation of polyols. The change from nitrogen-limited to phosphate-limited precultivation of immobilized spores significantly increased the productivity of the mycelium. The ratio of citric acid to residual sugar in the effluent distinctly lay in the direction of citric acid. Inside the alginate beads mainly large bulbous cells were observed.

Influence of sucrose concentration and phosphate limitation on citric acid production by immobilized cells of Aspergillus niger

A simple method is proposed for calculating oxygen pentration depth in immobilized cells by assuming zero order kinetics in the presence of several external oxygen transport resistances. Calculations indicate that typical penetration depths of oxygen for immobilized microbial cells are in the range of 50–200 μ and those for immobilized or encapsulated animal and plant tissue culture are about 500–1000 μ. Based on calculations, oxygen transport in microencapsulation and microcarriers for tissue cultures are not transport-limited, but a slight limitation is expected for those in a hollow fiber reactor.

Estimation of oxygen penetration depth in immobilized cells

Living Aspergillus niger cells were entrapped in polyacrylamide gels and employed in both replacement batch and continuous column bioreactors to produce citric acid from sucrose. With the replacement batch bioreactor, increase of citric acid was observed under conditions of higher aeration and of wider surface of immobilized cells. With the continuous bioreactor, the maximum citric acid yield was 39.1 mg/h per 40 g gels. The biocatalyst activity or half-life was 105 days.
