Journal ArticleDOI
Detection of neutralizing anti-therapeutic protein antibodies in serum or plasma samples containing high levels of the therapeutic protein
TL;DR: The development of a procedure that uses acid dissociation and affinity adsorption to remove therapeutic protein from assay samples and can detect the presence of neutralizing anti-therapeutic protein antibodies from samples containing high levels of therapeutic protein is described.
Journal ArticleDOI
A novel method for detecting neutralizing antibodies against therapeutic proteins by measuring gene expression.
Yanbin Yu,Christopher Piddington,Dan Fitzpatrick,Brian Twomey,Ren Xu,Steven J. Swanson,Shuqian Jing +6 more
TL;DR: This work has established a novel and non-radioactive bioassay system using branched DNA (bDNA) technology for detecting protein-therapeutic neutralizing antibodies in patient serum that measures the variations of target gene expression that reflect the biologic effect of the therapeutic agent and the capability of the antibodies, if present, to neutralize the therapeutics.
Journal ArticleDOI
Development of a Novel BAFF Responsive Cell Line Suitable for Detecting Bioactive BAFF and Neutralizing Antibodies against BAFF-Pathway Inhibiting Therapeutics
Jenny Hu,Yanbin Yu,Hong Han,Francesca Civoli,Yao Zhuang,John Thomas,Steve Swanson,Shuqian Jing,Shalini Gupta +8 more
TL;DR: The development of a sensitive BAFF responsive cell line via stable transfection of the BAFFR-TNFR1 hybrid receptor into monkey kidney epithelial COS-1 cells is reported here, useful in the development of sensitive bioassays to measure the levels of bioactive BAFF, inhibition of BAFF and neutralizing antibodies against any BAFF pathway-mediated therapeutic proteins.
Journal ArticleDOI
A non-radioactive method for detecting neutralizing antibodies against therapeutic proteins in serum.
TL;DR: This work has established a novel and non-radioactive bioassay system for detecting neutralizing antibodies in patient serum samples that measures the cell metabolic activities that are closely associated with cell proliferation and apoptosis.

Detection of compounds that affect therapeutic activity
Shuqian Jing,Francesca Civoli,Shalini Gupta,Daniel Steven Halperin,Jason J. Pennucci,Steven J. Swanson,Yan Bin Yu +6 more
TL;DR: In this paper, the authors present methods of detecting compounds that affect the activity of a therapeutic substance or composition administered to a subject, and to reagents for use in such methods.
