Isolation and characterization of bacteria associated with the rhizosphere of halophytes (Salsola stocksii and Atriplex amnicola) for production of hydrolytic enzymes

A halophilic bacterial strain, EG1HP4QL, was isolated from a salt sample from Lake Qarun, Fayoum Province, Egypt. Morphological, physiological, biochemical, and phylogenetic analyses indicated that the strain belonged to the genus Halobacillus. Strain EG1HP4QL produced an extracellular polysaccharide (EPS), with production peaking (5.9 g L−1) during growth on medium S-G containing 2% (w/v) sucrose at 35 °C (pH 8.0). The EPS had significant emulsifying activity (E24 %) against kerosene (65.7 ± 0.8%), o-xylene (64.0 ± 1%), and sunflower oil (44.7 ± 0.5%). The composition of the EPS included two polymers—a negatively charged and a neutral one (~ 3:1)—in which mannose and glucose were the main neutral monosaccharide constituents. Strain EG1HP4QL was able to utilize crude oil (35.3%) as the sole carbon source within 12 days. The minimum inhibitory concentrations of heavy metals [Zn(II), Cd(II), Pb(II), Ni(II), and Cu(II)] for strain EG1HP4QL were 1.0, 2.0, 2.0, 2.5, and 5 mM, respectively.

Biology and Applications of Halophilic Bacteria and Archaea: A Review

Extremophiles are attracting a lot of focus by the researchers in the recent years and amongst them are a group of halophilic fungi inhabiting hypersaline environments ubiquitously distributed globally. In year 2000, the fungi were considered as halophiles which inhabit hypersaline habitats and are ubiquitously distributed throughout the world. However, most of the research on halophilic fungi has been focused on adaptation studies of these fungi in hypersaline habitats or as preliminary reports of biotechnological screenings where some of the applications have been well characterized and researched. However, in the recent years the research has diversified to topics like extremozymes, followed by bioremediation studies. The rapidly changing global environment, upcoming challenges in industrial biotechnology, and unique properties of the metabolites obtained from halophilic organisms and especially obligate halophilic fungi are promising to the prospective future of research in biotechnology.

Biotechnological Applications of Halophilic Fungi: Past, Present, and Future

Halophiles are salt loving microbes optimally growing at high concentrations of salt. Khewra salt mines of Pakistan provide extreme saline conditions where enormous halophilic microbial biota thrives. The present study aimed at isolation and molecular identification of bacterial strains from saline waters of Khewra salt mines. Using halophilic media, nine halophilic bacterial strains from saline water bodies were cultured and studied under optimized growth conditions (NaCl, pH and temperature). Bacterial growth at different NaCl concentrations was measured at 600nm wavelength, showing optimal growth at 1.5M NaCl. 769bp size 16S rRNA gene was amplified for molecular identification of bacterial strains. The amplified genes of the strains FA2.2 and FA3.3 were sequenced and their homology with other bacterial strains was analyzed. The results showed FA2.2 shared maximum homology with Bacillus anthracis strain while FA3.3 showed close resemblance with Staphylococcus saprophyticus subsp. bovis. Isolated halophilic bacterial strains possess potential for various biotechnological applications. They could be manipulated for synthesizing transgenic crops tolerating high salinity boosting the agricultural yield. Moreover extremozymes of these bacteria holds great industrial importance.
