The study investigated the effect of turning frequency on survival of fecal indicator pathogens (E. coli, Enterococcus spp., Salmonella spp. and helminth eggs) during fecal sludge (FS) co-composting with sawdust. Dewatered FS was mixed with sawdust and composted on a pilot scale using different turning frequencies—i.e., 3 days (3TF), 7 days (7TF), and 14 days (14TF). Composting piles were monitored weekly for survival of fecal indicator microorganisms and evolution of selected physical and chemical characteristics for 14 weeks. Our results show that turning frequency has a statistically significant (p < 0.05) effect on pathogen inactivation in FS compost. The 3TF piles exhibited shorter pathogen inactivation periods (8 weeks) than 7TF and 14TF piles (10 weeks). Temperature-time was found to be the major factor responsible for the survival of pathogens in FS composting piles, followed by indigenous microbial activities and toxic by-products (monitored as NH4+-N). Our study findings suggest that even at low composting temperatures, the high turning frequency can enhance pathogen inactivation. This is a significant finding for composting activities in some rural areas where suitable organic solid waste for co-composting with FS to attain the recommended high thermophilic conditions could be greatly lacking.

Effect of Turning Frequency on the Survival of Fecal Indicator Microorganisms during Aerobic Composting of Fecal Sludge with Sawdust

The emptying and transport of faecal sludge (FS) is a fundamental aspect of the sanitation service chain and is mostly carried out by private operators who usually face a lot of challenges. Our review assessed how influencing factors and challenges FS emptiers face are linked and in turn how they act as barriers to improvement initiatives. We conducted a systematic review of peer-reviewed journals on FS emptying in sub-Saharan Africa, South and Southeast Asia published between January 2002 and December 2021. Amongst the 37 journals reviewed, accessibility was mostly documented (n = 18) as a factor which affected choice of emptying method, followed by cost (n = 14), quality of service (n = 13) and then sludge thickness (n = 8). We grouped the types of challenges identified from the publications into five categories of financial, technical and institutional (n = 14, each), followed by health (n = 12) and then social challenges (n = 8). Discussions on initiatives (n = 13) used to improve the emptying business were limited to Cost/affordability of sanitation services and access to finance by FS emptiers, which were noted to be the major barriers to effective implementation of these strategies. This review identified the need for sensitizing the public on FS emptying, financial modelling of manual emptying business and a need to study the relationship between perceptions and emptying behavior of users. Faecal sludge emptying in Sub-Saharan Africa, South and Southeast Asia: A systematic review of emptying technology choices, challenges, and improvement initiatives

Feacal sludge (FS) management is a growing concern in the Global South. Research has shown that FS has a potential for reuse in agriculture due to its high nutrient content. However, it contains many pathogens which present a public health and environmental risks when it is applied to land without proper treatment. Composting is one of the viable options for sanitizing FS biosolids before reuse in agriculture as a soil conditioner. However, pathogen die-off during FS composting and factors responsible for pathogen survival in compost are not well understood. Therefore, we conducted a systematic review to better understand pathogen inactivation during FS composting. The results revealed that although most pathogens are inactivated by the temperature generated during composting, spore-forming bacteria, helminths, and viruses are likely to survive the process. The findings indicated that although temperature-time is the major factor responsible for pathogen inactivation during composting, other factors (change in pH, moisture content, turning frequency, nutrient depletion, toxic by-products) and mechanisms (indigenous microbial activities, microbial antagonistic effect, antibiotic action) also have an influence. In addition, most studies on pathogen inactivation during FS composting are usually based on the use of indicator microorganisms and there are limited studies on specific pathogens. Hence, further research on virus, protozoa and antimicrobial resistance inactivation during FS composting as well as the role of factors and mechanisms involved in the composting process is needed. This research will generate vital formation to inform the design, operation, and optimization of FS composting facilities to produce pathogen-free compost. 

Inactivation of faecal pathogens during faecal sludge composting: a systematic review

On-site sanitation system emptying practices and influential factors in Asian low- and middle-income countries: a Systematic Review

Globally, about 2.7 billion people depend on onsite sanitation systems (OSS) (e.g., septic tanks) for their sanitation needs. Although onsite sanitation systems help in providing primary treatment for domestic wastewater, they don’t effectively remove nutrients, pathogens, and other inorganic contaminants. Previous studies have posited that the use of post treatment systems which incorporate biochar leads to improved contaminant removal efficiency. However, the mechanism through which contaminants are removed and factors potentially affecting the removal are still understudied. To fill this knowledge gaps, this review discusses factors which affect efficiency of biochar in removing contaminants found in onsite domestic wastewater, modifications applied to improve the efficiency of biochar in removing contaminants, mechanisms through which different contaminants are removed and constraints in the use of biochar for onsite wastewater treatment. It was noted that the removal of contaminants involves a combination of mechanisms which include adsorption, filtration, biodegradation, ion exchange, pore entrapment. The combination of these mechanisms is brought about by the synergy between the properties of biochar and microbes trapped in the biofilm on the surface of the biochar. Future areas of research such as the modification of biochar, use of biochar in the removal of antibiotic resistant genes (ARGs), application of wet carbonization methods and resistance of biochar to physical disintegration are also discussed. This study provides useful information that can be applied in the use of biochar for the treatment of wastewater and guide future design of treatment systems for optimized treatment performance. Biochar as a novel technology for treatment of onsite domestic wastewater: A critical review

In low-income, urban, informal communities lacking sewerage and solid waste services, onsite sanitation (sludges, aqueous effluent) and child feces are potential sources of human fecal contamination in living environments. Working in informal communities of urban Maputo, Mozambique, we developed a quantitative, stochastic, mass-balance approach to evaluate plausible scenarios of localized contamination that could explain why the soil-transmitted helminth Ascaris remains endemic despite nearly universal coverage of latrines that sequester most fecal wastes. We used microscopy to enumerate presumptively viable Ascaris ova in feces, fecal sludges, and soils from compounds (i.e., household clusters) and then constructed a steady-state mass-balance model to evaluate possible contamination scenarios capable of explaining observed ova counts in soils. Observed Ascaris counts (mean = −0.01 log10 ova per wet gram of soil, sd = 0.71 log10) could be explained by deposits of 1.9 grams per day (10th percentile 0.04 grams, 90th percentile 84 grams) of child feces on average, rare fecal sludge contamination events that transport 17 kg every three years (10th percentile 1.0 kg, 90th percentile 260 kg), or a daily discharge of 2.7 kg aqueous effluent from an onsite system (10th percentile 0.09 kg, 90th percentile 82 kg). Results suggest that even limited intermittent flows of fecal wastes in this setting can result in a steady-state density of Ascaris ova in soils capable of sustaining transmission, given the high prevalence of Ascaris shedding by children (prevalence = 25%; mean = 3.7 log10 per wet gram, sd = 1.1 log10), the high Ascaris ova counts in fecal sludges (prevalence = 88%; mean = 1.8 log10 per wet gram, sd = 0.95 log10), and the extended persistence and viability of Ascaris ova in soils. Even near-universal coverage of onsite sanitation may allow for sustained transmission of Ascaris under these conditions.

Persistent Ascaris Transmission Is Possible in Urban Areas Even Where Sanitation Coverage Is High

The role of canines in transmitting antibiotic resistant bacteria to humans in the urban environment is poorly understood. To elucidate this role, we utilized genomic sequencing and phylogenetics to characterize the burden and transmission dynamics of antibiotic resistant Escherichia coli (ABR-Ec) cultured from canine and human feces present on urban sidewalks in San Francisco, California. We collected a total of fifty-nine ABR-Ec from human (n=12) and canine (n=47) fecal samples from the Tenderloin and South of Market (SoMa) neighborhoods of San Francisco. We then analyzed phenotypic and genotypic antibiotic resistance (ABR) of the isolates, as well as clonal relationships based on cgMLST and single nucleotide polymorphisms (SNPs) of the core genomes. Using Bayesian inference, we reconstructed the transmission dynamics between humans and canines from multiple local outbreak clusters using the marginal structured coalescent approximation (MASCOT). Overall, we found human and canine samples to carry similar amounts and profiles of ABR genes. Our results provide evidence for multiple transmission events of ABR-Ec between humans and canines. In particular, we found one instance of likely transmission from canines to humans as well as an additional local outbreak cluster consisting of one canine and one human sample. Based on this analysis, it appears that canine feces act as an important reservoir of clinically relevant ABR-Ec within the urban environment. Our findings support that public health measures should continue to emphasize proper canine feces disposal practices, access to public toilets and sidewalk and street cleaning. Importance: Antibiotic resistance in E. coli is a growing public health concern with global attributable deaths projected to reach millions annually. Current research has focused heavily on clinical routes of antibiotic resistance transmission to design interventions while the role of alternative reservoirs such as domesticated animals remain less well understood. Our results suggest canines are part of the transmission network that disseminates high-risk multidrug resistance in E. coli within the urban San Francisco community. As such, this study highlights the need to consider canines, and potentially domesticated animals more broadly, when designing interventions to reduce the prevalence of antibiotic resistance in the community. Additionally, it showcases the utility of genomic epidemiology to reconstruct the pathways by which antimicrobial resistance spreads.
