Restraint stress in biomedical research: A review

— Withdrawal of food or immobilization both led to changes in rat brain tryptophan metabolism. Brain tryptophan and 5-hydroxyindolylacetic acid concentrations both increased while changes in 5-hydroxytryptamine were much smaller. Changes were greater upon withdrawal of food. The brain tryptophan change did not appear merely to reflect an overall increase of brain amino acid concentrations, brain tyrosine concentration being only slightly increased by food withdrawal and significantly decreased upon immobilization. Plasma tryptophan did not increase. The changes in brain indole metabolism were not abolished by adrenalectomy. Results are discussed in relation to the regulation of brain serotonin metabolism.

Effects of immobilization and food deprivation on rat brain tryptophan metabolism.

Restraint stress of 30 min increases plasma CS and lowers hypothalamic NE. Restraints of longer durations are associated with an attenuation of these changes. Daily repetitive restraint enhances the CS response on the second day and progressively diminishes it on subsequent days. Whole brain NE increases on the first day and decreases on Day 2 to 5. The CS response to acute restraint is similar is similar in 5 different normotensive rat strains, but is enhanced in the genetically hypertensive SH rat, its normotensive backbreed WKY, and the DOCA hypertensive Sprague-Dawley rat. Comparison with other stressors (electric foot shock and novel environment) indicate that the responses to restraint are different at least in time course, if not qualitatively.

Physiological and biochemical concomitants of restraint stress in rats.

The purpose of this chapter is to review some of the neurochemical responses that occur during stress and to assess their potential significance for the organism. Of the myriad stress responses that have been recorded, two systems stand out: the hypothalamic-pituitary-adrenocortical (HPA) system, and the sympathetic-adrenomedullary (SA) system. While most research has tended to focus on one or other of these systems, we believe the existence of these two parallel systems to be biologically significant. It is the relationship between these two systems that intrigues us, and this will be the focus of our review.

Neurochemical Responses in Stress: Relationships Between the Hypothalamic-Pituitary-Adrenal and Catecholamine Systems

This experiment examined how inescapable tail shock alters the level of dopamine and norepinephrine within various brain regions of the rat and the relationship of these changes to the depression of motor activity produced by the shock. Following exposure to tail shock that is known to interfere with acquisition of active behavioral tasks, animals were briefly tested for spontaneous motor activity and then sacrificed for neurochemical measures. Norepinephrine and dopamine levels in the frontal cortex, brain stem, striatum, olfactory tubercle, hypothalamus, hippocampus, septum, and amygdala were measured by a sensitive radicenzymatic technique. Exposure to 45 min of tail shock did not alter motor activity significantly, but shock sessions of 60 and 75 min duration produced a marked decrease in motor activity. Levels of dopamine were found to be very little changed in all brain regions studied except for the hypothalamus, in which a substantial rise in dopamine level was observed. Norepinephrine levels, in contrast, fell in many brain regions in response to shock. The fall in norepinephrine levels observed in twi brain regions was significantly correlated with the decline in motor activity (brain stemr=+0.70, hypothalamusr=+0.60) These data suggest that deficits in active motor behavior produced by shock parameters similar to those used in this study may reflect concomitant disturbances of noradrenergic function in specific brain regions.

Stress-induced depression of motor activity correlates with regional changes in brain norepinephrine but not in dopamine

1. The effect of a pharmacologically induced increase or depletion of brain monoamines (5-hydroxytryptamine, noradrenaline, dopamine) was investigated with respect to adrenocortical function.

2. No strict correlation was found between the depletion of brain amine stores induced by prenylamine or p-chlorophenylalanine and adrenocortical activation, even at times in which the peak effect of brain amine depletion occurs.

3. Restraint stress causes a manifest increase in brain 5-hydroxytryptamine while decreasing the cerebral noradrenaline and dopamine content. This stress strongly stimulates corticosterone secretion by the adrenals.

4. A monoamine oxidase inhibitor, nialamide, at a dose which causes an evident increase in brain amine concentrations, does not modify plasma corticosterone. At the same dose it was, furthermore, unable to prevent the adrenocortical stimulation induced by restraint stress.

5. Brain amine content does not seem to play an important part in the control of corticotrophin releasing factor in corticotrophin secretion by the pituitary gland. The relationship between hypothalamic monoamines and other neurohumours is discussed.
