Curcumin is the active ingredient of turmeric powder, a natural spice used for generations in traditional medicines. Curcumin’s broad spectrum of antioxidant, anticarcinogenic, antimutagenic, and anti-inflammatory properties makes it particularly interesting for the development of pharmaceutical compounds. Because of curcumin’s various effects on the function of numerous unrelated membrane proteins, it has been suggested that it affects the properties of the bilayer itself. However, a detailed atomic-level study of the interaction of curcumin with membranes has not been attempted. A combination of solid-state NMR and differential scanning calorimetry experiments shows curcumin has a strong effect on membrane structure at low concentrations. Curcumin and its two related compounds, that is, demethoxycurcumin and bis-demethoxycurcumin (curcuminoids) are the main secondary metabolites of Curcuma longa and other Curcuma spp. Curcumin is commonly used as coloring agent as well as food additive; curcumin has also shown some therapeutic activities. This review summarizes stability of curcumin in solutions, spectroscopy characteristics of curcumin (UV, IR, Raman, MS, and NMR), polymorphism forms, method of analysis in both of biological and nonbiological samples, and metabolite studies of curcumin. For analysis of curcumin and its related compounds in complex matrices, application of LC–MS/MS is recommended.

To enhance the dissolution and oral absorption of poorly soluble curcumin, self-emulsifying drug delivery system (SEDDS) was developed. SEDDS is composed of oil, surfactant, and cosurfactant and evaluated for its physicochemical properties, anti-angiogenic and anti-inflammatory activity in colitis induced rat model. SEDDS formulations were characterized for enhancement in solubility, in vitro dissolution, surface morphology, IR, DSC and X-ray studies. Formulation SF-2 had shown maximum solubility and in vitro release. In addition, SF-2 was found to have angio-inhibitory activity, as demonstrated by inhibition of angiogenesis in chorioallantoin membrane assay (CAM). Anti-inflammatory activity was evaluated using DSS induced colitis rat model. Curcumin and SF-2 treated rats showed faster weight gain and the whole colon length appeared to be significantly longer than in pure curcumin and DSS treated controls. Novel Self-emulsifying Formulation of Curcumin with Improved Dissolution, Antiangiogenic and Anti-inflammatory Activity

A simple, precise, rapid and accurate, binary reverse phase high performance liquid chromatographic method has been developed for the determination of curcumin a natural drugs with short run time. Chromatographic separation was achieved by using Merck C15 (250 cm X 4.6 mm) Column with mobile phase acetonitril: tetrahydrofuran: 2% acetic acid 50:30:20 (2%) was used. The flow rate was 0.7 ml/min. The retention time was 4.587 minutes. The limit of detection and limit of quantification of curcumin were between 3.68 to 8.125 ng/ml for 50-μL injection volumes. The percentage recovery of curcumin was found to be 97.2 to 98.4 and Relative standard deviation was 0.345 % and 1.160 %. The developed HPLC method can therefore be applied to both in vitro studies of curcumin formulations as well as drug estimation in biological samples.

Antioxidant, antimicrobial and cytotoxicity properties of the methanolic extract from Grewia tiliaefolia Vahl

A simple, precise, rapid and accurate, binary reverse phase high performance liquid chromatographic method has been developed for the determination of curcumin, Demethoxy curcumin and Bismethoxy curcumin with short run time. Chromatographic separation was achieved by using Merck C 15-- (250 cm × 4.6 mm) Column with mobile phase tetrahydrofuran: 1% citric acid 35: 65 was used. The flow rate was 1.2 ml/min. The retention time for curcumin was 15.892 minutes. The proposed method was validated for sensitivity, selectivity, linearity, accuracy, precision, ruggedness, robustness and solution stability. The limit of detection and quantification of curcumin was between 1 to 500 ng/mL respectively for 50-µL injection volumes. Limit of detection and Limit of quantification was calculated by visualization and statistical methods. The % recovery of sample spiked standard ranged within 95-105 %. Method, system, interday and intraday precision was found to be within the limits of acceptance criteria. Method was found to be rugged when different analyst carried out analysis. The method was found to be sensitive and efficient with 4886.77 theoretical plates and 0.2046 mm HETP. The method was suitable for the quality control of curcumin therefore can be applied to both in vitro studies of curcumin formulations as well as drug estimation in biological samples.
