The fisheries and livestock sectors capture the highest share of protein-rich animal food and demonstrate accelerated growth as an agriculture subsidiary. Environmental pollution, climate change, as well as pathogenic invasions exert increasing stress impacts that lead the productivity momentum at a crossroads. Oxidative stress is the most common form of stress phenomenon responsible for the retardation of productivity in fisheries and livestock. Essential micronutrients play a determinant role in combating oxidative stress. Selenium, one of the essential micronutrients, appears as a potent antioxidant with reduced toxicity in its nanoscale form. In the present review, different methods of synthesis and characterization of nanoscale selenium have been discussed. The functional characterization of nano-selenium in terms of its effect on growth patterns, feed digestibility, and reproductive system has been discussed to elucidate the mechanism of action. Moreover, its anti-carcinogenic and antioxidant potentiality, antimicrobial and immunomodulatory efficacy, and fatty acid reduction in liver have been deciphered as the new phenomena of nano-selenium application. Biologically synthesized nano-selenium raises hope for pharmacologically enriched, naturally stable nanoscale selenium with high ecological viability. Hence, nano-selenium can be administered with commercial feeds for improvising stress resilience and productivity of fish and livestock.

Selenium Nanoparticles for Stress-Resilient Fish and Livestock

Recently, it was observed that there is an increasing application of nanoparticles (NPs) in aquaculture. Manufacturers are trying to use nano-based tools to remove the barriers about waterborne food, growth, reproduction, and culturing of species, their health, and water treatment in order to increase aquaculture production rates, being the safe-by-design approach still unapplied. We reviewed the applications of NPs in aquaculture evidencing that the way NPs are applied can be very different: some are direclty added to feed, other to water media or in aquaculture facilities. Traditional toxicity data cannot be easily used to infer on aquaculture mainly considering short-term exposure scenarios, underestimating the potential exposure of aquacultured species. The main outputs are (i) biological models are not recurrent, and in the case, testing protocols are frequently different; (ii) most data derived from toxicity studies are not specifically designed on aquaculture needs, thus contact time, exposure concentrations, and other ancillary conditions do not meet the required standard for aquaculture; (iii) short-term exposure periods are investigated mainly on species of indirect aquaculture interest, while shrimp and fish as final consumers in aquaculture plants are underinvestigated (scarce or unknown data on trophic chain transfer of NPs): little information is available about the amount of NPs accumulated within marketed organisms; (iv) how NPs present in the packaging of aquacultured products can affect their quality remained substantially unexplored. This experiment was conducted to compare the effects of different sources of selenium (Se) on growth, muscle Se concentration, muscle proximate composition, blood enzymes and antioxidant status of common carp, Cyprinus carpio. Organic Se (selenomethionine, SeMet), inorganic Se (sodium selenite, Na2SeO3), or nanoselenium were each added to the basal diet at 0.7 mg Se kg−1 diet. Four groups of fish with an average weight of 9.69 ± 0.12 g were fed one of the experimental diets for 8 weeks. Nano-Se diet resulted in better growth performance (P < 0.05). Fish fed with Se-supplemented diets showed a higher (P < 0.05) Se content in the muscle and liver tissues. Fish fed nano-Se had the highest liver glutathione peroxidase, superoxide dismutase and catalase activities (P < 0.05). Liver malondialdehyde level was significantly (P < 0.05) lower in fish fed on SeMet and nano-Se diets as compared to control. The carp fed on nano-Se diet had the lowest values of aspartate transaminase and alanine transaminase. Lactate dehydrogenase activity was significantly lower in fish fed on SeMet and nano-Se diets. This study shows that nano-Se acts more efficiently on growth performance and antioxidant defence system of common carp than organic and inorganic sources of Se.

Effects of different dietary selenium sources (sodium selenite, selenomethionine and nanoselenium) on growth performance, muscle composition, blood enzymes and antioxidant status of common carp (Cyprinus carpio)

An 8-week feeding trial was conducted to compare the effects of supplementing (0.7 mg kg-1) different dietary selenium (Se) sources including organic [selenomethionine (SeMet)], inorganic [sodium selenite (Na2SeO3)], and nanoparticulate Se (nano-Se) on physiological responses of common carp, Cyprinus carpio juveniles (9.7 ± 0.1 g). Basal diet without Se supplementation used as control. Fish fed nano-Se supplemented diet had the highest weight gain (97.2 ± 10.8%) and feed efficiency ratio (42.4 ± 0.8%). Intestinal villi height was significantly taller in fish fed nano-Se diet than in the control group in both foregut and midgut sections. Serum glutathione peroxidase and superoxide dismutase activities were significantly higher in nano-Se and SeMet groups than in control and sodium selenite groups. Fish fed Se-supplemented diets had greater red blood cell counts and hematocrit and hemoglobin values than the control group (P < 0.05). Nano-Se and SeMet groups showed a significant increase in white blood cell counts, neutrophil percentage, and serum lysozyme activity than the other groups. Fish fed nano-Se diet had the highest serum hemolytic activity, total immunoglobulin, and total protein and albumin contents, as well as the lowest serum total cholesterol and low density lipoprotein levels (P < 0.05). Overall, significant improvements in growth performance, feed utilization, intestinal morphology, and hemato-immunological and serum biochemical parameters of common carp juveniles suggest nano-Se as an efficient source for providing dietary Se in this species.

Nano-selenium (Se), with its high bioavailability and low toxicity, has attracted wide attention for its potential application in the prevention of oxidative damage in animal tissues. However, the effect of nano-Se of different sizes on the intestinal epithelial cells of the crucian carp (Carassius auratus gibelio) is poorly understood. Our study showed that different sizes and doses of nano-Se have varied effects on the cellular protein contents and the enzyme activities of secreted lactate dehydrogenase, intracellular sodium potassium adenosine triphosphatase, glutathione peroxidase, and superoxide dismutase. It was also indicated that nano-Se had a size-dependent effect on the primary intestinal epithelial cells of the crucian carp. Thus, these findings may bring us a step closer to understanding the size effect and the bioavailability of nano-Se on the intestinal tract of the crucian carp.
