This study describes the development, optimization, and evaluation of a novel  composite  imprinted  polymer, on  the  basis  of  magnetic  halloysite   nanotubes  (MHNTs-MIPs) using “Quality by Design (QbD)” approach combining computer simulation and response surface methodology. Norfloxacin, methacrylic acid, and ethylene glycol dimethacrylate were used as template, functional monomer and cross-linker, respectively. As a comparison, two MHNTs-MIPs have been prepared with the most suitable functional monomer methacrylic acid (MAA) along with acrylamide (AM). To explain the adsorption behavior, adsorption kinetics and isotherms were studied. Magnetic halloysite nanotubes molecularly imprinted polymers prepared from MAA (MHNTs-MIP1) displayed a high adsorption capacity (349 µg mg-1) toward NOR. A magnetic imprinting  solid phase extraction method  coupled  with  high  performance  liquid  chromatography  (MHNTs-MISPE-HPLC-UV) was  developed  for  the  determination of NOR  in  serum  and  water samples, by applying MHNTs-MIP as a sorbent. The recoveries from 83.76% to 103.30% in water and from 90.46% to 99.78% in serum were obtained. Besides remarquable mechanical properties and specific recognition of MHNTs-MIP toward template molecule. It could be also collected and separated fastly by external magnetic field. Moreover, MHNTs-MIPs could be reused for several cycles with the recovery range from 83.25% to 100.96% for  water sample and from 85.65% to 100.33% for serum sample. These analytical results of serum and  water samples showed that the proposed method based on MHNTs-MIPs is applicable for fast and selective extraction of therapeutic agents from biological fluids and environmental water.

Affinity chromatography is a well-known method dependent on molecular recognition and is used to purify biomolecules by mimicking the specific interactions between the biomolecules and their substrates. Enzyme substrates, cofactors, antigens, and inhibitors are generally utilized as bioligands in affinity chromatography. However, their cost, instability, and leakage problems are the main drawbacks of these bioligands. Biomimetic affinity ligands can recognize their target molecules with high selectivity. Their cost-effectiveness and chemical and biological stabilities make these antibody analogs favorable candidates for affinity chromatography applications. Biomimetics applies to nature and aims to develop nanodevices, processes, and nanomaterials. Today, biomimetics provides a design approach to the biomimetic affinity ligands with the aid of computational methods, rational design, and other approaches to meet the requirements of the bioligands and improve the downstream process. This review highlighted the recent trends in designing biomimetic affinity ligands and summarized their binding interactions with the target molecules with computational approaches.

Inspirations of Biomimetic Affinity Ligands: A Review

In recent years, single-cell or low-input multi-omics techniques have brought a revolution in the study of pre-implantation embryo development. However, single-cell or low-input proteome research in this field is relatively underdeveloped, due to the limited source of mammalian embryo samples, the objective reality of high abundance zona pellucida proteins, and the lack of hypersensitive proteome technology. Here, a comprehensive solution of ultrasensitive proteome technology was developed for single-cell or low-input mouse embryos. Both deep coverage route and high-throughput route could significantly reduce the starting material and enhance the proteomic depth without any customized instrument. Using the deep coverage route, an average of 2,665 or 4,585 protein groups can be identified from 1 or 20 mouse zygotes respectively. Using the high-throughput route, 300 single mouse zygotes can be analysis in 8 days with an average of 2,371 proteins identified. With its popularization, we believe researchers can choose deep coverage or high-throughput technology routes according to their own conditions.

Proteins analysis from an average cell population often overlooks the cellular heterogeneity of expressed effector molecules, and knowledge about the regulations of key biological processes may remain obscure. Therefore, the necessity of single-cell proteomics (SCP) technologies arises. Without microfluidic chip, expensive ultrasonic equipment, or reformed liquid chromatogram (LC) system, we established an Ultra-sensitive and Easy-to-use multiplexed Single-Cell Proteomic workflow (UE-SCP). Specifically, the flexible sorting system ensured outstanding cell activity, high accuracy, remarkable efficiency, and robustness during single-cell isolation. Multiplex isobaric labeling realized the high-throughput analysis in trapped ion mobility spectrometry coupled with quadrupole time-of-flight mass spectrometry (timsTOF MS). Using this pipeline, we achieved single-cell protein quantities to a depth of over 2,000 protein groups in two human cell lines, Hela and HEK-293T. A small batch experiment can identify and quantify more than 3200 protein groups in 32 single cells, while a large batch experiment can identify and quantify about 4000 protein groups in 96 single cells. All the 128 single cells from different cell lines could been unsupervised clustered based on their proteomes. After the integration of data quality control, data cleaning, and data analysis, we are confident that our UE-SCP platform will be easy-to-marketing popularization and will promote biological applications of single-cell proteomics.
