Journal ArticleDOI
Production of infectious hepatitis C virus in tissue culture from a cloned viral genome
Takaji Wakita,Thomas Pietschmann,Takanobu Kato,Takanobu Kato,Tomoko Date,Michiko Miyamoto,Zijiang Zhao,Krishna K. Murthy,Anja Habermann,Hans-Georg Kräusslich,Masashi Mizokami,Ralf Bartenschlager,T. Jake Liang +12 more
TL;DR: It is shown that the JFH1 genome replicates efficiently and supports secretion of viral particles after transfection into a human hepatoma cell line (Huh7) and provides a powerful tool for studying the viral life cycle and developing antiviral strategies.
Journal ArticleDOI
Nonhepatic Cell Lines HeLa and 293 Support Efficient Replication of the Hepatitis C Virus Genotype 2a Subgenomic Replicon
TL;DR: Sequencing analysis revealed that mutations in HCV-derived regions were not essential for replication in these cells, as some clones displayed no mutations.
Journal ArticleDOI
Genotype 2a Hepatitis C Virus Subgenomic Replicon Can Replicate in HepG2 and IMY-N9 Cells
Tomoko Date,Takanobu Kato,Michiko Miyamoto,Zijiang Zhao,Kotaro Yasui,Masashi Mizokami,Takaji Wakita +6 more
TL;DR: In this paper, the replication of JFH-1 replicon was tested in hepatitis C virus genotype 2a subgenomic replicon, and in IMY-N9, a cell line developed by fusing human hepatocytes and HepG2 cells.
Journal ArticleDOI
Nuclear Localization of Japanese Encephalitis Virus Core Protein Enhances Viral Replication
Yoshio Mori,Tamaki Okabayashi,Tetsuo Yamashita,Zijiang Zhao,Takaji Wakita,Kotaro Yasui,Futoshi Hasebe,Masayuki Tadano,Eiji Konishi,Kohji Moriishi,Yoshiharu Matsuura +10 more
TL;DR: The results indicate that nuclear localization of JEV core protein plays crucial roles not only in the replication in mammalian cells in vitro but also in the pathogenesis of encephalitis induced by JEV in vivo.
Journal ArticleDOI
Characterization of the E-138 (Glu/Lys) mutation in Japanese encephalitis virus by using a stable, full-length, infectious cDNA clone.
TL;DR: It is demonstrated clearly that a single Glu-to-Lys mutation at aa 138 of the envelope protein affects multiple steps of the viral life cycle and may induce substantial attenuation of Japanese encephalitis virus.
