Abstract: In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide. Our ultimate goal is to encourage intellectual and technical innovation in the field.

Abstract: Intracellular potassium homeostasis is a prerequisite for the optimal operation of plant metabolic machinery and plant's overall performance. It is controlled by K(+) uptake, efflux and intracellular and long-distance relocation, mediated by a large number of K(+) -selective and non-selective channels and transporters located at both plasma and vacuolar membranes. All abiotic and biotic stresses result in a significant disturbance to intracellular potassium homeostasis. In this work, we discuss molecular mechanisms and messengers mediating potassium transport and homeostasis focusing on four major environmental stresses: salinity, drought, flooding and biotic factors. We argue that cytosolic K(+) content may be considered as one of the 'master switches' enabling plant transition from the normal metabolism to 'hibernated state' during first hours after the stress exposure and then to a recovery phase. We show that all these stresses trigger substantial disturbance to K(+) homeostasis and provoke a feedback control on K(+) channels and transporters expression and post-translational regulation of their activity, optimizing K(+) absorption and usage, and, at the extreme end, assisting the programmed cell death. We discuss specific modes of regulation of the activity of K(+) channels and transporters by membrane voltage, intracellular Ca(2+) , reactive oxygen species, polyamines, phytohormones and gasotransmitters, and link this regulation with plant-adaptive responses to hostile environments.

Abstract: Today, vehicles are increasingly being connected to the Internet of Things which enable them to provide ubiquitous access to information to drivers and passengers while on the move. However, as the number of connected vehicles keeps increasing, new requirements (such as seamless, secure, robust, scalable information exchange among vehicles, humans, and roadside infrastructures) of vehicular networks are emerging. In this context, the original concept of vehicular ad-hoc networks is being transformed into a new concept called the Internet of Vehicles (IoV). We discuss the benefits of IoV along with recent industry standards developed to promote its implementation. We further present recently proposed communication protocols to enable the seamless integration and operation of the IoV. Finally, we present future research directions of IoV that require further consideration from the vehicular research community.

Abstract: Transportation is a necessary infrastructure for our modern society. The performance of transportation systems is of crucial importance for individual mobility, commerce, and for the economic growth of all nations. In recent years modern society has been facing more traffic jams, higher fuel prices, and an increase in CO2 emissions. It is imperative to improve the safety and efficiency of transportation. Developing a sustainable intelligent transportation system requires the seamless integration and interoperability with emerging technologies such as connected vehicles, cloud computing, and the Internet of Things. In this article we present and discuss some of the integration challenges that must be addressed to enable an intelligent transportation system to address issues facing the transportation sector such as high fuel prices, high levels of CO2 emissions, increasing traffic congestion, and improved road safety.
