Abstract: Ox red blood cells sensitized with rabbit anti-ox red cell antibody adhered opsonically to freshly prepared human peripheral blood lymphocytes. When tested by a centrifugation technique 24–59% of live lymphocytes in normal individuals formed rosettes with the indicator red cells while 41–87% of the lymphocytes in chronic lymphatic leukaemia reacted. After storage at + 4 °C the lymphocytes gradually lost the operative receptor; killing the cells by freezing and thawing, by sodium azide or by fixation in formaldehyde also resulted in loss of capacity to bind the indicator cells. Pretreatment of the lymphocytes with high doses of anti-human IgM in diluted serum resulted in a definite, although incomplete, inhibition. Also, soluble antigen-antibody complexes were inhibitory, although strong inhibition was difficult to show. Undiluted, heated normal human or rabbit serum was highly inhibitory. The high incidence of lymphocytes showing antibody opsonic adherence in patients with chronic lymphatic leukaemia, together with the finding that most lymphocytes binding sensitized ox red cells did not bind unsensitized sheep red cells (a possible T-cell marker) suggests that lymphocytes showing antibody opsonic adherence represent a subpopulation of peripheral blood lymphocytes. This subpopulation might be an analogue of the B-cell population in mice.

Abstract: Antinuclear antibody (ANA) titers in complement immunofluorescence (CIF) and conventional indirect immunofluorescence (IIF) staining were established and CIF/IIF ratios of ANA titers calculated. The following properties of conjugates seem to be indicated for determinations of CIF/IIF ratios: (1) anti-C4 conjugate dilutions used for CIF staining of ANA should be adjusted to contain ½ unit of antibody/ml; (2) anti-IgG conjugates used for this purpose should be adjusted to ¼ unit/ml; (3) for determinations of CIF/IIF ratios of ANA titers, the two conjugates used should have comparable F/P ratios or optical sensitivity ratios. Using such a system a preliminary double-blind study of 54 selected sera from patients with various collagen diseases was undertaken. After completion of titrations and decoding, it was found that 11 sera of patients with a diagnosis of systemic lupus erythematosus (SLE) all had ANA titers of 200 or more and all fixed the C4 component of complement. The CIF/IIF ratio of these 11 sera fell in the range of 1:6 to 1:1. Of the remaining 43 sera 4 fixed complement. Of these, 2 had antinucleolar antibodies rather than true ANA. Of the other 2 sera which fixed C4, one was from a case of discoid lupus erythematosus and another from a patient with rheumatoid arthritis. Their CIF/IIF ratios were 1:8 and 1:21, respectively. Tests for ANA patterns of 25 coded sera in another laboratory yielded peripheral staining with 3 of 4 SLE sera and with none of the other sera included.

Abstract: The percentage of lymphoid cells carrying immunoglobulin surface determinants was examined for the yolk sac, bursa of Fabricius, thymus, bone marrow, blood lymphocytes, spleen, cecal tonsils and Harder’s glands in normal White Leghorn chickens from the 11th day of incubation until the 26th week after hatching, using direct immunofluorescent staining with a fluoresceinated rabbit anti-chicken immunoglobulin serum. The results clearly show that under the test conditions employed surface immunoglobulins are markers of a bursa cell population or of bursa-derived cells, whereas thymus cells react only minimally with anti-immunoglobulin conjugates in young chickens. The migration pattern and maturation of bursa cells were studied chronologically as reflected in appearance, quantity and morphology of immunoglobulin surface determinant bearing cells in different organs.

Abstract: The K antibody response was studied in immunized rabbits and in children with urinary tract infection. Hyperimmunization of rabbits with K antigen-containing bacteria gave a relatively marked and regular antibody response with both β-mercaptoethanol sensitive and resistant antibodies. Purified K antigen only in a few animals gave very low K antibody titres. Tolerance induction in rabbits was possible using purified K antigen. In patients, K antibodies were very seldom demonstrated. Five occasions of pyelonephritis out of 9 in 8 patients with acute pyelonephritis caused by E. coli O6:K2a,2c or O6:K13 and only 2 patients out of 17 having K1-containing strains gave an antibody response to the K antigen although they all formed antibodies to the·antigen of the infecting bacteria. No K antibodies were found in patients with cystitis.

Abstract: The pathogenic retina antigen, previously localized by immunofluorescence to the photoreceptor cell layers, was identified as a soluble protein having an estimated molecular weight of 55,000. The antigen was separated from guinea pig retina homogenate by successive fractionation techniques using ultra-centrifugation, ammonium sulfate precipitation, and chromatography. Electrophoretic and immunoelectrophoretic analyses indicated that the antigen was obtained in a highly purified form, and as little as 5 μg mixed with adjuvant produced 100% incidence of uveitis in guinea pigs. Formation of a double precipitin band was observed by immunodiffusion and immunoelectrophoresis on testing both crude and purified preparations against homologous guinea pig antiserum. Although this finding suggested antigenic heterogeneity among molecular components of the antigen, the antigen could not be separated into two components by electrophoresis or chromatography.

Abstract: Although uveitis has been widely identified as the main feature of the autoimmune disease produced in several vertebrate species immunized with retina, the present study shows that retinitis is the main feature of the disease in rats. These species differences can be correlated with species differences in intraocular vascularization, and the hematogenous pathways by which the inflammatory cells reach the ocular target.
