Abstract: [Objective] To observe pathological changes and apoptosis in rat brain nerve cells after aconitine poisoning.[Methods] Experimental models of 35 Wistar rats aconitine poisoning were established,and the technology of apoptosis detection was used.The regular HE staining and immunohistochemical staining results were analyzed by computer image analysis system.[Results] Apoptosis never cells counts in poisoning group were higher than those of the control nomal group at different stages.In addition,the apoptosis cells counts were different after different poisoning time in the same organ(P 0.05).[Conclusion] Aconitine poisoning can not be detected by clinical symptoms or poison analysis while pathological changes can be found through the apoptosis detection.

Abstract: [Objective] To investigate the expression characteristic and significance of human telomerase reverse transcriptase(hTERT) and PCNA in brain glioma.[Method] Expressions of hTERT gene in 75 specimens of glioma and 4 specimens of normal brain were detected by in situ hybridization(ISH),PCNA protein was detected by immunohistochemical staining(SABC).[Result] 40 glioma cases positively expressed hTERT.4 cases of the normal brain didn't express hTERT.hTERT expression had relation to the glioma grading.31 glioma cases expressed PCNA positively,including 21 cases who expressed hTERT positively and 10 cases who expressed hTERT negatively.There was a significant difference in positive cell numbers of PCNA between hTERT-positive group and hTERT-negative one(P 0.01).[Conclusion] Expression rate of hTERT is positively correlated with pathologic grading of glioma,the cell which expresses hTERT has high proliferation capacity.

Abstract: [Objective] To evaluate the feasibility of the reconstruction of portal vein/superior mesenteric vein using the ligmentum teres hepatic. [Methods] The data of 4 cases with portal vein/superior mesenteric vein reconstruction using the ligmentum teres hepatis were analyzed retrospectively.[Results] The length of vascular resection was 3.5 cm to 6.5 cm.The length of ligment teres hepatis used during operation was 4.0 to 7.0 cm.The portal vein clamping time during the interposition grafting ranged from 30 to 126 min.No patient died in operation.The follow-up time was 10 to 39 months . The graft of ligment teres hepatis had no thrombosis and kept unblocked .[Conclusion] Portal vein/superior mesenteric vein reconstruction using the ligment teres hepatis during the pancreatoduodenectomy combined with vascular resection is a feasibility method.

Abstract: Objective: To identify protein profiling difference of white adipose tissue between type 2 diabetes and control. Methods: The white adipose tissue of obesity type 2 diabetes and control were used to isolate differentially expressed proteins by two dimensional electrophoresis,the 2-D gels were analysed with PD-Quest 6.2 software, three spots in which the differentially expressed protein was more obvious were selected and analysed with matrix assisted laser desorption/ionlzation time of flying mass spectrometry (MALD1-TOF-MS). Results: There were 300 protein spots or so observed in adipose tissue of control and type 2 diabetes respectively,and the majority of the spots between two types adipose tissue matched each other, only a few expressed differentially. After analysed by MALDI-TOF-MS and database search for three differentially expressed proteins, a new protein and other two sequence-known proteins including thioredoxin and Iipin were identified. Conclusions: Two-dimensional electrophoresis and MALDI-TOF-MS are effective means in the protein profiling analysis of white adipose tissue, the differentially expressed proteins may play an important role during the development of obesity and type 2 diabetes and serve as a new target of diabetes therapy.

Abstract: [Objective] To investigate the relationship between the expression of cholecystokinin(CCK)-A receptor(AR)mRNA of gallbladder wall and the plasma CCK-8 level and emptying function of the gallbladder in gallstone patients [Methods] The gallbladder emptying function was determined preoperatively with B ultrasonograph ,CCK-AR mRNA of gallbladder was studied by RT-PCR,plasma CCK-8 were detected by radioimmunoassay before the cholecystectomy and after the operation in 60 patients of gallstones group and 30 patients of control group [Results] The CCK-AR mRNA of gallstones group was lower than that of the control group(P 001),and so did patients with impaired gallbladder motility than those with normal motility(P 001) Plasma CCK-8 level in gallstone group was significantly higher than that in control group (P 005) Plasma CCK-8 level before the cholecystectomy were significantly higher than that after the operation in the gallstone group (P 005) [Conclusion] CCK-AR and CCK-8 play an important role in the regulation of gallbladder motility

Abstract: [Objective] To investigate the relationship between polymorphism of insulin receptor(INSR) gene Exon 17 with hypertensive nephrosclerosis and renal hypertension.[Methods] Polymerase chain relation(PCR) combined with single strand conformation polymorphism(SSCP) was used to detect and analyse the NsiI polymorphism of INSR gene Exon 17 in normal renal function of essential hypertension(EH-NRF),renal failure of hypertensive nephroselerosis(EH-CRF),normal renal function of renal hypertension(RH-NRF),renal failure of renal hypertension (RH-CRF) and normal control subjects(NC).[Results] Three different patterns of the PCR products in SSCP,which called band pattern Ⅰ,Ⅱ respectively,were presented in all four patient groups and normal control subject.The mutation frequency of EH-NRF and EH-CRF was remarkblely higher than those of RH-NRF,RH-CRF and NC(P0.05).there was no diference between patients of EH-NRF and EH-CRF.Among the patients of RH-NRF,RH-CRF and NC,there was no diference in frequency of all band pattern.[Conclusions] ①The polymorphism of INSR gene Exon 17 was associated with EH and the mutation frequency of EH was remarkblety higher(band Ⅰ+Ⅱ) than that of RIt.②The polymorphism of INSR gene Exon 17 has unconcern with EH whether complicated by renal damage and with renal damage complicated by hypertension as well.

Abstract: Objective: To investigate the possibly molecular mechanism of inflammation in chronic obstructive pulmonary disease(COPD).Methods: Peripheral venous blood were collected from 12 patients with COPD,10 patients with asthma,10 healthy cases,and peripheral blood monoenuclear cells(PBMC) were extracted with ficioll-hypague.The mRNA levels of c-fos,c-jun,IL-8 were evaluated by reverse transcription-polymerase chain reaction(RT-PCR).The protein levels of cfos,c-jun were determined by immunocytochemistry(ICC).Results: The c-fos level in COPD group and the asthma group had no difference,but were both higher than that in the control group.The levels of c-jun and IL-8 in COPD group were significantly higher than that in the asthma group and the control group(P0.05).The expression of c-jun had a positive correlation with IL-8(P0.05).Conclusions: The increased expression of c-jun playes an important role in COPD,it may enhance the activity of AP-1 and increase the expression of IL-8.

Abstract: [Objective] To study the suppression effect of Astilbin on cardiac allograft rejection in mice.[Methods] The model of mice cervical heterotopic heart transplantation was set up.forty-eight heart transplanted mice were divided in to 4 groups randomly: group A were fed with normal saline(1 ml per day),group B were fed with CsA 5 mg/kg per day,group C were fed with Astilbin 1 mg/kg per day,and group D were fed with Astilbin 1 ml/kg per day aud CsA 2.5 mg/kg per day.Grafts survival days and transplanted heart beating conditions were observed every day after heart transplantation.[Results] In group B,C and D,the survival time of the grafts was prolonged significantly as compared with group A.The survival time of the grafts in group D was prolonged significantly as compared with group C(P 0.05) and group B (P 0.01).[Conclusions] Astilbin can suppress cardiac allograft rejection,and has synergistic reaction with CsA.

Abstract: [Objective]To investigate the regulatory effects of diammonium glycyrrhizinate(DG) on proinflammatory cytokines TNF-α,IL-6 and IL-8 in rats model with ulcerative colitis(UC)[Methods]30 Spragur-Dawley(SD) female rats were randomized into treatment group,acetic acid control group and normal control group,10 rats in each groupColitis rats in treatment group and acetic acid control group were induced by administrating acetic acid rectallyThe rats in treatment group were treated with DGColon mucosal inflammation and damage were assessed by measuring disease activity index(DAI) mucosal lesion area and histological damage scoresLevels of serum interleukin-6(IL-6),interleukin-8(IL-8) and tumor necrosis factor-alpha(TNF-α) were detected by ELISA[Results]Compared with the acetic acid control group,the DAI,mucosal lesion area and histological damage scores in treatment group were significantly improved,and the serum levels of IL-6,IL-8 and TNF-α of treatment group were significantly decreased(P001)[Conclusion]The present study proved that DG has therapeutic effect on UC,it can down-regulate the serum levels of IL-6,IL-8 and TNF-α

Abstract: Objective:To evaluate the advantages of harvesting skeletonized internal thoracic artery(ITA) in coronary artery bypass grafting.Methods:The internal thoracic artery was mobilized from the satellite veins and surrounding tissue mainly using scissors and clips in 60 patients underwent coronary artery bypass grafting.Results:61 skeletonized ITAs were mobilized,the mean time of harvesting was 26.8 min.The mean blood flow of skeletonized ITA before anastomosis was 86.5ml/min,after anastomosis was 26.4ml/min.Follow-up data showed there were no cardiac deaths or cardiac events.Conclusion:Skeletonized ITA is as safe and effective as ITA with pedicel in coronary artery bypass grafting.

Abstract: [Objective] To observe the clinical efficacy and safety of transurethral KTP laser vaporization for bladder tumors[Methods] 42 bladder tumor cases were randomly assigned to undergo KTP laser vaporization,25 cases of the control underwent standard transurethral resection of bladder tumor(TURBt)The mean operative time,urethral catheter retaining time,postoperative recurrences,the case number of obtaining tumor stages,and the numbers of bladder perforation,postoperative bladder perfusion,and urethral strict were compared between the two groups[Results] The mean operative time,dwelling urethral catheter time,incidences of urethral stricture and postoperative recurrence were not significantly different between the two groups(P005)Compared with TURBt group,more cases in KTP laser group obtained accurate tumor stages,less cases needed bladder perfusion and had bladder perforation(P005)[Conclusions] KTP laser vaporization is feasible and appears to be safe and effective for treatment of bladder tumorsIn addition,it has more advantages in obtaining accurate tumor stage,reducing the incidence of bladder perforation and the amount of bleeding during operation than conventional TURBt,and specially in high-risk patients receiving anticoagulant therapy

Abstract: [Objective]To explore the etiology,pathogenesis,diagnosis and treatment of postsurgical gastroparesis syndrome after subtotal gastrectomy.[Methods] Clinical data of 896 patients with subtotal gastrectomy were analyzed retrospectively.[Results]There were 32 patients suffering from PGS (3.6% in total 896 patients),occurred within 3~10 d after operation.Radiography of upper gastrointestinal tract and gastroscopy were main methods to diagnose PGS.32 patients were cured without operation.[Conclusions] PGS is induced by multiple factors.and organic and mechanical illness must be excluded.The methods without operation are effective to patients with PGS,and operation should be classed as contraindication.

Abstract: [Objective] To observe the proliferation inhibitive effect of grub extract on human breast cancer MCF-7 cell line.[Methods] MTT assay was used to measure the growth inhibition rate of human breast cancer MCF-7 cell line treated with the grub extract.The expression of PCNA,Ki-67 and CyclinD1 was detected by immunohistochemistry before and after exposure to the grub extract.FCM was used to assay the change of cell cycle.[Results] The growth inhibition rate of MCF-7 cells treated with the grub extract was significantly higher than that of control,and the inhibitive effect was time and concentration dependent.The expression of Ki-67,PCNA and CyclinD1 was down-regulated and the cells were cumulated in S phase.[Conclusion] The proliferation of human breast cancer MCF-7 cell line can be inhibited significantly by the grub extract in vitro.It is related with down-regulation of CyclinD1,Ki-67 and PCNA.

Abstract: Objective: To explore the role of Gisenoside-Rg1 on BDNF protein in cerebrum cortex and hippocampus after brain ischemia.Methods: The cerebral infarction in the territory of middle cerebral artery models were established by using 60 adult male SD rats,2 hours later,these rats were performed reperfused and divided randomly into five groups,each with 12 rats.Five groups were performed intraperitoheal injection with saline(5mg/kg),nimodipine(15mg/kg)、 Gisenoside(150mg/kg) Gisenoside(100mg/kg) and Gisenoside(200mg/kg) respectively.Constitution were prepared with frozen section and the sections were stained by immunohistochemistry ABC method by using BDNF(1∶500) antibody.The BDNF protein grayworth and positive neurons amounts were observed. Results: BDNF protein was observed both in cerbrum cortex and hippocampus.The BDNF grayworth in groups 3~5 is lower than that in group 1 and group 2(P0.05,(0.01)).But the positive neurons amounts was significanfly higher than that in the latter.The BDNF grayworth and the positive neurons amounts in cerebrum cortex were both lower than that in hippocampus in group 3 and group 4(P 0.05,0.01).Conclusion: Gisenoside-Rg1 can increase the protein in BDNF-positive neurons both in cerebrum cortex and hippocampus.There is a sigmfficant difference between the cerbrum cortex and hippocumpus.The possible mechanism is not only that BDNF exists in positive neurons but also different parts of the brain have different reaction to drugs and cerebral ischemia.

Abstract: Objective. To observe the effect of metronomic chemotherapy with cyclophosphamide (crX) on tumor growth and angiogenesis on mouse Lewis lung carcinoma (LLC) model. Methods. LLC cells were injected subcuta- neously into C57BL/6 mice on day O. Mice were randomly divided into metronomic group, MTD group and control group. Therapies were given to the groups from the 6th day. Mice weight and tumor diameter were measured every other day. Then tumors were weighed. Microvessel count and proliferating index of tumor cell were performed by immunohistochemical staining with anti-C~l antibody and anti-PCNA antibody. The histological characteristics of the tumor were detected by microscopy. Results. Tumor growth and angiogenesis were inhibited by crx signifi- cantly in the metronomic group, which has better efficacy and lower toxicity than maximum tolerated do.'ie (MTD) group. The average weight of the tumors in control group, metronomic group and MTD group were (3.72:t O. 60) g, (1. 85 :t O. 38 ) g and (1. 73 :t O. 41) g, the average microvessel count were 26. 38 :t 2. 56, 15. 09 :t 3.03 and 23. 51:t 2. 78, PCNA index were 83.88:t 3. 72 ,81. 60:t 4. 21 and 71.80:t 3. 86, respectively. Necrosis was in- creased significantly in the metronomic group. Conclusion. crx inhibits tumor growth and angiogenesis in mouse LLC model significantly. The inhibition may be correlated with the anti-angiogenic effect of crX. (Life Science
