Abstract: Objective To investigate the effect of XPG C46 T gene polymorphisms in rectal cancer tissues on neoadjuvant chemoradiotherapy. Methods Forty-five patients with low rectal cancer( Ⅱ-Ⅲ midterm) were given XELOX chemotherapy( oxaliplatin combined with capecitabine) before surgery. CT or MRI examination was conducted to evaluate the efficacy after radiotherapy 6-8 weeks. The XPG C46 T genotype was detected before and after radiotherapy by RT-PCR. Results After neoadjuvant chemoradiotherapy,27 cases were effective,18 cases were non-effective,and the effective rate was 60. 00%. The efficacy of neoadjuvant chemoradiation was not related with gender,age,pathological type and KPS score of colorectal cancer patients( all P 0. 05). There were 18 cases of XPG C46 T genotype C / C in the rectal cancer tissues of patients with effective neoadjuvant chemoradiotherapy,3 cases in patients with non-effective neoadjuvant chemoradiotherapy( P 0. 05); there were respectively 6 cases of C / T and 9 cases in patients with effective and in patients with non-effective neoadjuvant chemoradiotherapy( P 0. 05); T / T were respectively 3 cases and 6 cases in patients with effective and in patients with non-effective neoadjuvant chemoradiotherapy( P 0. 05). Conclusions The XPG C46 T gene C /C genotype ratio in patients with effective neoadjuvant chemoradiotherapy is higher,C / T and T / T genotype ratios are lower than those of the invalid ones. The detection of XPG C46 T gene polymorphism can be used to predict the curative effect of colorectal cancer in Ⅱ-Ⅲ stage.

Abstract: Objective To explore the expression changes of NFKBIA in the primary hepatocellular carcinoma( PHC)tissues and its relationships with the clinicopathological parameters and prognosis. Methods The expression of NFKBIA mRNA in the cancerous tissues and adjacent normal tissues from 74 patients with liver cancer receiving the surgeries was detected by RT-PCR. The expression of AFP,Ki-67 and Connexin-43 in the liver cancer tissues was qualitatively detected by immunohistochemistry. The associations of NFKBIA expression levels with gender,age,diameter of the tumor,differentiation,tumor envelop completeness,liver cirrhosis,tumor thrombus and the expression of AFP,Ki-67 and Connexin-43 were analyzed. Mean survival time( MST) was compared between patients with positive NFKBIA and patients with negative NFKBIA. Results The expression of NFKBIA in the cancer tissues( 1. 13 ± 0. 78) was significantly lower than that in the adjacent normal tissues( 2. 11 ± 1. 56)( P 0. 05). The expression of NFKBIA mRNA in the liver cancer tissues was related with the tumor envelop completeness( P 0. 05),and was not related with gender,age,diameter,differentiation,tumor thrombus,liver cirrhosis and the expression of AFP,Ki-67 and Connexin-43( all P 0. 05). The MST of patients with negative NFKFIA expression( 17 months) was shorter than that of patients with positive NFKBIA expression( 22months),( P 0. 05). Conclusion The expression of NFKBIA decreases in the liver cancer tissues,which may be closely related to tumor envelop completeness. The negative NFKBIA expression predicts the poor prognosis of patients with PHC.
