in which the originators of many gel-type matrixes have described the development of their materials. In Part II, the story continues to unfold in the same manner. The focus of Part I was the introductory section by Bruce Merrifield; that of Part II is provided by a personal perspective from the other “father” of this field, Robert Letsinger, who independently conceived of solid-phase synthesis and pursued the concept, as applied to DNA synthesis, with equal vigor. His efforts provide the foundation for much of today’s biotechnology. Truly we do stand on the shoulders of giants, 2 who dreamed of things that never were and made those dreams reality. Table 1 summarizes some reasons why there has been so much interest in the development of new supports. Tables 2 and 3 provide an overview of the content of the two parts. Note, however, that the second contribution to Part II is a description of traditional gel form PS beads made with some novel technological improvements. Each section is introduced by personal remarks (D.H. comments). The final section consists of a discussion by Bing Yan and myself on a topic dear to our hearts: we know from the few studies that have been done that the rates and yields of particular transformations are highly dependent on the nature of the matrix. These studies provide some clues as to which would be the best selection for any reaction series. As with Part I, all contributors were given similar instructions: to prepare a personal account of their support development work, focusing on how the structure of the support influences its suitability. 3 The opinions and comments provided by the contributors are, obviously, their own, and occasionally they are in conflict with statements by myself and others (since no contributor has had access to other contributions). I have on several occasions had to suggest modifications to authors, but I have never sought to influence their opinions; differences of opinion are not uncommon in this field and are part of its considerable fascination. This invention provides an apparatus for preparing chemical libraries. The apparatus includes: (1) a carousel comprising a plurality of reaction mounts having at least one reaction well; (2) a rotator that rotates the carousel step-wise; (3) a fluid delivery system; (4) a drain system; and (5) a programmable computer that controls the operation of the apparatus, including the rotator, the fluid delivery system, the drain system and other systems in the apparatus. The preparation of chemical libraries involves rotating the carousel through a plurality of stations. At each station, a physical step in a reaction protocol is carried out on the reaction wells of the mount docked at the station.

The invention relates to a device for carrying out chemical reactions. Said device comprises a reaction slider in which a plurality of through openings are configured as reaction chambers and a selection slider which is arranged on the upper surface of the reaction slider and which has at least one traversing control opening. Said selection slider is displaceable relative to the reaction slider in such a manner that the control opening thereof is aligned with one of the reaction chambers of the reaction slider so that a traversing connection for supply of the reaction chamber with a predetermined reagent is formed. The device is further provided with a slider for chemical agents which is arranged below the reaction slider and which is configured with through openings for feeding chemicals to the individual reaction chambers of the reaction slider. The invention provides a means for impinging the sliders with a predetermined force and further provides a valve arrangement with which chemicals can be fed to the chemical slider as a premix.

Device for carrying out chemical reactions

Biomolecular computing is an area of tremendous activity and potential. It is a t t ract ing the interest of large numbers of excellent researchers, and has been extensively reported on in the popular press. Indeed, two days before this introduction was written, the work of Professors Ogihara and Ray was the subject of a front-page headline of the New York Times's on-line \"CYBERTIMES.\" This issue's column is an expert guest column on this exciting topic.

Oligonucleotides are synthesized and purified in a single column filled with a mixture of nucleoside-loaded and underivatized, high-cross link, non-swelling polystryene beads. A new oligonucleotide production system has been developed to completely automate-the entire process of sequence entry, phosphoramidite synthesis, cleavage, deprotection, purification, quantitation, and sample collection. Up to 48 primer-length, high-purity oligonucleotides can be produced in a 24 hour period of unattended automation on the ABI 3948 DNA Synthesizer. Synthesis, cleavage, and purification occur within the unique OneStepTM column. Analyses by MicroGel capillary electrophoresis, anion-exchange HPLC, PAGE, and enzymatic digestion/HPLC routinely exhibit > 90% product purity. Stringent PCR, automated fluorescent sequencing, and genetic analysis experiments show the positive effects of the total automation of purified oligonucleotide production.
