The nucleolus and ribosome biogenesis

The nucleolus of eukaryotic cells was first described in the early 19th century and was discovered in the 1960s to be the seat of ribosome synthesis. Although rRNA transcription, rRNA processing and ribosome assembly have been clearly established as major functions of the nucleolus, recent studies suggest that the nucleolus participates in many other aspects of gene expression as well. Thus, the nucleolus has been implicated in the processing or nuclear export of certain mRNAs. In addition, new results indicate that biosyntheses of signal recognition particle RNA and telomerase RNA involve a nucleolar stage and that the nucleolus is also involved in processing of U6 RNA, one of the spliceosomal small nuclear RNAs. Interestingly, these three nucleolus-associated small nuclear RNAs (signal recognition particle RNA, telomerase RNA and U6 RNA) are components of catalytic ribonucleoprotein machines. Finally, recent work has also suggested that some transfer RNA precursors are processed in the nucleolus. The nucleolus may have evolutionarily descended from a proto-eukaryotic minimal genome that was spatially linked to vicinal RNA processing and ribonucleoprotein assembly events involved in gene read-out. The nucleolus of today's eukaryotes, now surrounded by the chromatin of over 2 billion years of genome expansion, may still perform these ancient functions, in addition to ribosome biosynthesis. The plurifunctional nucleolus concept has a strong footing in contemporary data and adds a new perspective to our current picture of the spatial-functional design of the cell nucleus.

It is common knowledge today that DNA molecules are not simple, rigid double helices, but molecules which have considerable conformational flexibility. This flexibility allows coiling of DNA into complex higher order structures. In most living cells or organelles, including many virus particles, these higher order structures occur on the basis of negative supercoiling or, as it is also called, negative superhelicity. Negative supercoiling is the conformational consequence of a winding deficit in DNA, i.e., the DNA under conditions of defined constraints forms in vivo or in vitro fewer helical rotations of one strand about the other than it assumes if left unconstrained.

In somatic cells the DNA coding for 28S and 18S ribosomal RNA (rRNA) is confined to the chromosomal nucleolus organizer (Birnstiel et al., 1966). However, in oocytes from a wide variety of animals, this nucleolar DNA is selectively amplified and accumulated in the nucleus free of the chromosomes (Gall, 1969). In Xenopus laevis, the best studied example, almost three-quarters of the nuclear DNA in a diplotene oocyte codes exclusively for ribosomal RNA (Brown and Dawid, 1968). The extra ribosomal DNA (rDNA) is synthesized during a three-week period in early meiotic prophase (Gall, 1968; Macgregor, 1968; Bird and Birnstiel, 1971; Watson-Coggins and Gall, 1971) and during diplotene it forms hundreds of extrachromosomal nucleoli, which actively engage in rRNA synthesis (Miller and Beatty, 1969). This paper will summarize our recent studies of the amplification mechanism by autoradiography of isolated rDNA using both light and electron microscopy.

The Mechanism of Gene Amplification in Xenopus Laevis Oöcytes

Electron microscopic analysis of the material released by gentle lysis of prokaryotic and eukaryotic cells has provided information on the in vivo structure of transcription complexes. We have identified active nonribosomal and ribosomal RNA genes in both cell types (for reviews see Miller and Hamkalo, 1972; Hamkalo and Miller, 1973). This paper summarizes the comparative structural features of these two types of transcription in three eukaryotic cells (HeLa, Drosophila, and yeast).
