Journal ArticleDOI
SAK/PLK4 is required for centriole duplication and flagella development.
Mónica Bettencourt-Dias,Ana Rodrigues-Martins,Lee Carpenter,Maria Giovanna Riparbelli,Laurent Lehmann,Melanie K. Gatt,N. Carmo,Francois Balloux,Giuliano Callaini,David M. Glover +9 more
TL;DR: It is shown that downregulation of SAK in Drosophila cells, by mutation or RNAi, leads to loss of centrioles, the core structures of centrosomes, and that depletion of Sak in human cells also prevents centriole duplication and gives rise to mitotic abnormalities.
Journal ArticleDOI
Asterless is a scaffold for the onset of centriole assembly
Nikola S. Dzhindzhev,Quan D. Yu,Kipp Weiskopf,Kipp Weiskopf,George Tzolovsky,Inês Cunha-Ferreira,Inês Cunha-Ferreira,Maria Giovanna Riparbelli,Ana Rodrigues-Martins,Ana Rodrigues-Martins,Mónica Bettencourt-Dias,Giuliano Callaini,David M. Glover +12 more
TL;DR: Independent functions for Asl as a scaffold for Plk4 and Sas-4 that facilitates self-assembly and duplication of the centriole and organization of pericentriolar material are identified.
Journal ArticleDOI
Revisiting the role of the mother centriole in centriole biogenesis.
Ana Rodrigues-Martins,Maria Giovanna Riparbelli,Maria Giovanna Riparbelli,Maria Giovanna Riparbelli,Giuliano Callaini,Giuliano Callaini,Giuliano Callaini,David M. Glover,David M. Glover,David M. Glover,Mónica Bettencourt-Dias,Mónica Bettencourt-Dias,Mónica Bettencourt-Dias +12 more
TL;DR: It is found that overexpression of SAK/PLK4 could induce amplification of centrioles in Drosophila embryos and their de novo formation in unfertilized eggs.
Journal ArticleDOI
The SCF/Slimb Ubiquitin Ligase Limits Centrosome Amplification through Degradation of SAK/PLK4
Inês Cunha-Ferreira,Inês Cunha-Ferreira,Ana Rodrigues-Martins,Ana Rodrigues-Martins,Inês Bento,Maria Giovanna Riparbelli,Wei Zhang,Ernest D. Laue,Giuliano Callaini,David M. Glover,Mónica Bettencourt-Dias +10 more
TL;DR: It is shown that centrosome amplification is normally inhibited by degradation of SAK/PK4 degradation, mediated by the SCF/Slimb ubiquitin ligase, which plays an important role in cell-cycle progression and how it may go awry in human disease.
Journal ArticleDOI
Stepwise evolution of the centriole-assembly pathway
Zita Carvalho-Santos,Pedro Machado,Pedro Branco,Filipe Tavares-Cadete,Ana Rodrigues-Martins,José B. Pereira-Leal,Mónica Bettencourt-Dias +6 more
TL;DR: It is proposed that the UNIMOD theory explains the conservation of CBB architecture and that taxon- and tissue-specific molecular innovations, gained through emergence, duplication and divergence, play important roles in coordinating CBB biogenesis and function in different cellular contexts.
