Difficulty in swallowing is common among all age groups, especially in elderly and pediatrics. Fast dissolving tablets constitute an innovative dosage forms that overcome the problems of swallowing and provide a quick onset of action. Fast-dissolving drug delivery system, in most cases, is a tablet that dissolves or disintegrates in the oral cavity without the need of water or chewing. Fast-dissolving drug delivery system must include substances to mask the taste of the active ingredient. This masked active ingredient is then swallowed by the patient’s saliva along with the soluble and insoluble excipients. Loratadine is antihistaminic drugs which is H1receptor blocker used in the treatment of hay fever and other dermatalogical diseases. In the present study, loratadine fast dissolving tablets were prepared by solid dispersion and using croscarmellose sodium, sodium starch glycolate and polyplasdone XL as super disintegrants. The preformulation study indicated that it is suitable for direct compression. The drug contents of all the tablets were found to be uniform with low standard deviation, indicating efficient mixing and showed quick disintegration and higher dissolution.

Fast Dissolving Tablets

The objective of present research work was to develop methotrexate loaded chitosan anchored shell nanoparticles for drug delivery in breast cancer. Chitosan nanoparticles (CS-NPs) were synthesized by ionic gelation of chitosan using sodium tripolyphosphate (STPP). The optimized nanoparticles were characterized for particle size and polydispersity index (PDI) revealed particle size were found to be between 57.08 nm to169.5 nm and PDI 0.252 to 0.639 respectively. The results signpost that stirring speed during ionic gelation reaction was also decisive parameters for the size of the nanoparticles obtained. Further characterization involved to show polymer-drug interaction was FTIR and DSC. Cariprazine is an oral antipsychotic approved in the US for the treatment of schizophrenia, acute bipolar mania, and most recently, bipolar depression. The aim of this systematic review is to describe the efficacy, tolerability and safety of cariprazine for the treatment of depressive episodes associated with bipolar I disorder (bipolar depression) in adults. Cariprazine in transdermal dosage form would be a lot of fascinating to regulate the mood for extended period by achieving the plasma concentration well higher than the therapeutic levels. With psychiatrical patients, schedules of daily medication should be followed to avoid adverse reactions which are notably difficult. Therefore, delivery of such drugs by transdermal route can be done. Development and maintenance of low dose drugs therapy is desired, which will limit the consequences of significant adverse effect and direct the issues of helpless consistence in the patients. Ideal quantity of the psychotropic drug can be given by transdermal route with least adverse reactions to overcome the psychic conditions. In this research work, we have synthesized the novel electrically responsive graft co-polymers using natural polymers with polyacrylamide and utilized for the preparation of cariprazine containing electrically responsive transdermal therapeutic systems (ETTS). The ETTS developed using electrically-responsive graft copolymers of PAAm-g-GGH having crosslinked with glutaraldehyde have confirmed that there was augmented drug permeation under the influence of electric stimuli, while it was reduced without electric current. The Cariprazine permeability got decreased as the concentration of crosslinking agent was increased. Further, higher drug permeability rate observed with increasing electric current strength from 2 to 8 mA. There was a pulsatile pattern of drug permeability under ‘on and ‘off’ electric stimulus mode. Hence, these graft copolymers are competent biomaterials those could be used for the development of electro-sensitive transdermal therapeutic systems for on-demand drug delivery.

Development and Evaluation of (ETTS) Externally Triggered Transdermal Therapeutic Systems for Cariprazine by using newly synthesized graft Natural co-polymer- PAAm-g-GGH

Therefore, delivery of such drugs by transdermal route can be done. Development and evaluation of antipsychotic drugs for externally triggered transdermal therapeutic systems

The objective of present study was to formulate directly compressible orodispersible tablets of quetiapine fumarate by sublimation method with a view to enhance patient compliance. A full 32 factorial design was used to investigate the effect of two variables viz., concentration of Indion 414 and camphor. Indion 414 (3-5 % w/w) was used as superdisintegrant and camphor (5-15 % w/w) as subliming agent. The tablets were evaluated for thickness, weight variation, hardness, friability, content uniformity, wetting time, porosity, in vitro disintegration time and in vitro drug release. In vitro dissolution profile revealed faster and maximum drug from formulation F3. SEM study show formation of pores on tablet surface after sublimation of the sublimating agent, thus providing a sufficiently porous structure. This permitted the selection of a batch of tablets with desired disintegration time and improved dissolution rate after oral administration. The F3 batch containing the Indion 414 (5%) and Camphor (5%) w/w of total formulation weight had shown good the disintegration time of 18.66 seconds and with improved dissolution rate and desirable friability. Further studies will be required to evaluate the performance of dosage form in vivo and In Vitro In vivo Correlation.
