Structure-based design of a benzodiazepine scaffold yields a potent allosteric inhibitor of hepatitis C NS5B RNA polymerase.

TL;DR: Using structure-based design, a 1,5-benzodiazepine NS5B polymerase inhibitor chemotype is optimized into a new sulfone-containing scaffold that yielded potent inhibitor (S)-4c (K(D) = 0.79 nM), which has approximately 20-fold greater affinity for NS 5B than its carbonyl analogue (R)-2c.

TL;DR: 1a/1b subtype profiling of polymerase inhibitors that bind at each of the four known nonnucleoside binding sites is reported, showing that inhibition of all of the clinical isolates tested is maintained, except for inhibitors that binds at the palm-1 binding site.
