Journal ArticleDOI

Binding-Site Identification and Genotypic Profiling of Hepatitis C Virus Polymerase Inhibitors

TL;DR: A hepatitis C virus mutant and genotypic recombinant polymerase panel is established as a means of guiding medicinal chemistry through the elucidation of the site of action of novel inhibitors and profiling against genotypes.
Journal ArticleDOI

Minor variant detection in amplicons using 454 massive parallel pyrosequencing: experiences and considerations for successful applications

TL;DR: Because accuracy of the DNA polymerase is vital for reliable UDS results, a comparative analysis of error profiles from seven different DNA polymerases was performed and experimentally assessed in parallel by 454 sequencing and revealed highly reproducible results in amplicon-based UDS.
Journal ArticleDOI

1a/1b Subtype Profiling of Nonnucleoside Polymerase Inhibitors of Hepatitis C Virus

TL;DR: 1a/1b subtype profiling of polymerase inhibitors that bind at each of the four known nonnucleoside binding sites is reported, showing that inhibition of all of the clinical isolates tested is maintained, except for inhibitors that binds at the palm-1 binding site.
Journal ArticleDOI

1,5-Benzodiazepines, a Novel Class of Hepatitis C Virus Polymerase Nonnucleoside Inhibitors

TL;DR: A novel class of HCV polymerase nonnucleoside inhibitors, 1, 5-benzodiazepines (1,5-BZDs), identified by high-throughput screening of a library of small molecules, found to inhibit the RdRp activity of NS5B noncompetitively with GTP and to inhibit a first phosphodiester bond during the polymerization cycle.
