Journal ArticleDOI
Differential efficacies of Cas nucleases on microsatellites involved in human disorders and associated off-target mutations.
Lucie Poggi,Lucie Poggi,Lisa Emmenegger,Stéphane Descorps-Declère,Bruno Dumas,Guy-Franck Richard,Guy-Franck Richard +6 more
TL;DR: In this article, the efficacy of several CRISPR-Cas nucleases to induce recombination within disease-related microsatellites, in Saccharomyces cerevisiae, was measured.
Book ChapterDOI
Monitoring Double-Strand Break Repair of Trinucleotide Repeats Using a Yeast Fluorescent Reporter Assay.
TL;DR: A GFP reporter assay in Saccharomyces cerevisiae is described allowing for the quantification of nuclease efficacy at inducing a DSB, by monitoring the reconstitution of a functional GFP gene whose expression can be rapidly quantified by flow cytometry.

Differential efficacies of Cas nucleases on microsatellites involved in human disorders and associated off-target mutations
Lucie Poggi,Lucie Poggi,Lisa Emmenegger,Lisa Emmenegger,Stéphane Descorps-Declère,Bruno Dumas,Guy-Franck Richard,Guy-Franck Richard +7 more
TL;DR: It is demonstrated that secondary structure formation on the guide RNA was a major determinant of nuclease efficacy, and the most efficient of all CRISPR-Cas nucleases was Streptococcus pyogenes Cas9.
